    4D Antarctica: a new effort aims to help bridge the gap between Antarctic crust and lithosphere structure and geothermal heat flux

    Seismology, satellite-magnetic and aeromagnetic data, and sparse MT provide the only available geophysical proxies for large parts of Antarctica\u2019s Geothermal Heat Flux (GHF) due to the sparseness of direct measurements. However, these geophysical methods have yielded significantly different GHF estimates. This restricts our knowledge of Antarctica\u2019s contrasting tectono-thermal provinces and their influence on subglacial hydrology and ice sheet dynamics. For example, some models derived from aeromagnetic data predict remarkably high GHF in the interior of the West Antarctic Rift System (WARS), while other satellite magnetic and seismological models favour instead a significantly colder rift interior but higher GHF stretching from the Marie Byrd Land dome towards the Antarctic Peninsula, and beneath parts of the Transantarctic Mountains. Reconciling these differences in West Antarctica is imperative to better comprehend the degree to which the WARS influences the West Antarctic Ice Sheet, including thermal influences on GIA. Equally important, is quantifying geothermal heat flux variability in the generally colder but composite East Antarctic craton, especially beneath its giant marine-based basins. Here we present a new ESA project- 4D Antarctica that aims to better connect international Antarctic crust and lithosphere studies with GHF, and assess its influence on subglacial hydrology by analysing and modelling recent satellite and airborne geophysical datasets. The state of the art, hypotheses to test, and methodological approaches for five key study areas, including the Amundsen Sea Embayment, the Wilkes Subglacial Basin and the Totten catchment, the Recovery and Pensacola-Pole Basins and the Gamburtsev Sublgacial Mountains/East Antarctic Rift System are highlighted

    Revealing the crustal architecture of the least understood composite craton on Earth: East Antarctica

    East Antarctica hosts one of the largest Precambrian cratons on Earth. Meager coastal exposures and sediment provenance studies provide glimpses into up to 3 billion years of its geological history. Extensive ice sheet cover hampers however our knowledge of crustal architecture, and consequently the geodynamic processes responsible for the growth and amalgamation of East Antarctica have remained elusive. Here we exploit recent aerogeophysical exploration efforts to help unveil the large-scale crustal architecture of East Antarctica. We focus on three sectors of East Antarctica: the Transantarctic Mountains and Wilkes Basin area; the Recovery/Dronning Maud Land area and the Gamburtsev Province. These areas provide new insights into both the margins of the so called Mawson craton and the processes that affected its interior. A 1,900 km-long linear magnetic and gravity boundary is imaged along the western flank of the Wilkes Basin and interpreted here as a crustal-scale Paleoproterozoic suture zone (ca 1.7 Ga) that inverted a former passive margin. Two ribbon-like Archean and Paleoproterozic microcontinents were assembled during this stage, resembling modes of amalgamation of Paleoproterozoic microcontinental ribbons in Australia. The proposed Proterozoic sutures and microcontinent boundaries also influenced Neoproterozoic rifted margin and early Cambrian back-arc basins in the Wilkes Basin/Transantarctic Mountains region. In the Recovery/Dronning Maud Land region our new potential field compilations reveal a wide tract of anastomising crustal-scale shear zones, likely of Pan-African age that flank and variably deform the margins of several distinct Archean, Paleo-Mesoproterozoic and Grenvillian age crustal blocks. In the Gamburtsev Province new magnetic and gravity models provide insights into the Gamburtsev Suture (Ferraccioli et al., 2011, Nature) that separates the Ruker Province from an inferred Grenvillian-age orogenic Gamburtsev Province with remarkably thick crust (up to 60 km thick) and thick lithosphere (over 200 km thick). We suggest that a recently inferred Tonian-age accretionary belt identified in the Sor Rondane region continues further inland in the Gamburtsev Province and was likely also reactivated during Pan-African age transpression linked to Gondwana assembly

    New Views of East Antarctica- from Columbia to Gondwana

    East Antarctica is a keystone in the Gondwana, Rodinia and the Columbia supercontinents. Recent aerogeophysical research, augmented by satellite magnetic, gravity and seismological data is unveiling the crustal architecture of the continent. This is helping comprehend the impact of supercontinental processes such as subduction, accretion, rifting and intraplate tectonics on its evolution. A mosaic of Precambrian basement provinces is apparent in interior East Antarctica (Ferraccioli et al., 2011, Nature). A major suture separates the Archean-Neoproterozoic Ruker Province from an inferred Grenvillian-age orogenic Gamburtsev Province with remarkably thick crust (up to 60 km thick) and thick lithosphere (over 200 km thick). The age of the suturing and its linkages with supercontinental assembly is debated with both Rodinia and Gondwana candidates being proposed. Further east, magnetic highs delineate a Paleo to Mesoproterozoic Nimrod-South Pole igneous province (Goodge and Finn, 2010 JGR) that flanks a composite Mawson Continent- including the Gawler Craton of South Australia (Aitken et al., 2014 GRL). An over 1,900 km long magnetic and gravity lineament is imaged along the western flank of the Wilkes Subglacial Basin and is interpreted here as a major Paleoproterozoic suture zone linked to the collision of Laurentia and East Antarctica within Columbia. The proposed suture played a pivotal role helping localise Neoproterozoic Rodinia rifted margin evolution and forming a backstop for the Ross-Delamerian cycle of Gondwana amalgamation. Aeromagnetic and gravity imaging help determine the extent of a Keweenawan-age (ca 1.1 Ga) large igneous province in the Coats Land Block -isotopically tied with the Mid-Continent Rift System of Laurentia (Loewy et al., 2011 Geology). Imprints of Grenvillian magmatic arc accretion link together the Namaqua-Natal and Maud belts in South Africa and Dronning Maud Land within Rodinia. The aeromagnetically distinct Southeast Dronning Maud Land province (Mieth and Jokat, 2014 GR) may represent a separate 1000-900 Ma Oceanic Arc Superterrane (Jacobs et al., 2015 Prec. Res.). New geophysical views of the Shackleton Range suture lend weight to more complex collisional and indentation tectonic models for the Pan-African age assembly of Gondwan

    Advancing diagnostics to address antibacterial resistance: The diagnostics and devices committee of the Antibacterial Resistance Leadership Group

    Diagnostics are a cornerstone of the practice of infectious diseases. However, various limitations frequently lead to unmet clinical needs. In most other domains, diagnostics focus on narrowly defined questions, provide readily interpretable answers, and use true gold standards for development. In contrast, infectious diseases diagnostics must contend with scores of potential pathogens, dozens of clinical syndromes, emerging pathogens, rapid evolution of existing pathogens and their associated resistance mechanisms, and the absence of gold standards in many situations. In spite of these challenges, the importance and value of diagnostics cannot be underestimated. Therefore, the Antibacterial Resistance Leadership Group has identified diagnostics as 1 of 4 major areas of emphasis. Herein, we provide an overview of that development, highlighting several examples where innovation in study design, content, and execution is advancing the field of infectious diseases diagnostics

    Janus-faced EPHB4-associated disorders: novel pathogenic variants and unreported intrafamilial overlapping phenotypes.

    PURPOSE: Several clinical phenotypes including fetal hydrops, central conducting lymphatic anomaly or capillary malformations with arteriovenous malformations 2 (CM-AVM2) have been associated with EPHB4 (Ephrin type B receptor 4) variants, demanding new approaches for deciphering pathogenesis of novel variants of uncertain significance (VUS) identified in EPHB4, and for the identification of differentiated disease mechanisms at the molecular level. METHODS: Ten index cases with various phenotypes, either fetal hydrops, CM-AVM2, or peripheral lower limb lymphedema, whose distinct clinical phenotypes are described in detail in this study, presented with a variant in EPHB4. In vitro functional studies were performed to confirm pathogenicity. RESULTS: Pathogenicity was demonstrated for six of the seven novel EPHB4 VUS investigated. A heterogeneity of molecular disease mechanisms was identified, from loss of protein production or aberrant subcellular localization to total reduction of the phosphorylation capability of the receptor. There was some phenotype-genotype correlation; however, previously unreported intrafamilial overlapping phenotypes such as lymphatic-related fetal hydrops (LRFH) and CM-AVM2 in the same family were observed. CONCLUSION: This study highlights the usefulness of protein expression and subcellular localization studies to predict EPHB4 variant pathogenesis. Our accurate clinical phenotyping expands our interpretation of the Janus-faced spectrum of EPHB4-related disorders, introducing the discovery of cases with overlapping phenotypes

    Nomograms of Iranian fetal middle cerebral artery Doppler waveforms and uniformity of their pattern with other populations' nomograms

    <p>Abstract</p> <p>Background</p> <p>Doppler flow velocity waveform analysis of fetal vessels is one of the main methods for evaluating fetus health before labor. Doppler waves of middle cerebral artery (MCA) can predict most of the at risk fetuses in high risk pregnancies. In this study, we tried to obtain normal values and their nomograms during pregnancy for Doppler flow velocity indices of MCA in 20 – 40 weeks of normal pregnancies in Iranian population and compare their pattern with other countries' nomograms.</p> <p>Methods</p> <p>During present descriptive cross-sectional study, 1037 normal pregnant women with 20<sup>th</sup>–40<sup>th </sup>week gestational age were underwent MCA Doppler study. All cases were studied by gray scale ultrasonography initially and Doppler of MCA afterward. Resistive Index (RI), Pulsative Index (PI), Systolic/Diastolic ratio (S/D ratio), and Peak Systolic Velocity (PSV) values of MCA were determined for all of the subjects.</p> <p>Results</p> <p>Results of present study showed that RI, PI, S/D ratio values of MCA decreased with parabolic pattern and PSV value increased with simple pattern, as gestational age progressed. These changes were statistically significant (P = 0.000 for all of indices) and more characteristic during late weeks of pregnancy.</p> <p>Conclusion</p> <p>Values of RI, PI and S/D ratio indices reduced toward the end of pregnancy, but PSV increased. Despite the trivial difference, nomograms of various Doppler indices in present study have similar pattern with other studies.</p

    Agentic learning: the pedagogical implications of young trans people’s online learning strategies

    This paper proposes anew conceptualisation of learning in the age of the internet, increasing systemic rigidity of formal education and intensified media manipulation and partiality. Using empirical data and drawing on Social Activity Method it elaborates the different strategies young trans people recruit in their self-learning and contends that these constitute a type of learning where the control of pedagogy, the learning environment and the subject matter lies to a significant extent, with the learner, taking place in spaces free from the influence of hegemonic transphobia. This type of learning appears to constitute an effective but complex one. As, in this instance, the learning is taking place in a wider cultural environment where the subject matter is often suppressed and subject to ideological misrepresentation by hegemonic control of the public sphere, this study suggests that learning by providing learners with greater control over pedagogy and learning environment is effective

    High body mass index is not associated with atopy in schoolchildren living in rural and urban areas of Ghana

    <p>Abstract</p> <p>Background</p> <p>Factors which determine the development of atopy and the observed rural-urban gradient in its prevalence are not fully understood. High body mass index (BMI) has been associated with asthma and potentially atopy in industrialized countries. In developing countries, the transition from rural to urban areas has been associated with lifestyle changes and an increased prevalence of high BMI; however, the effect of high BMI on atopy remains unknown in this population. We therefore investigated the association between high BMI and atopy among schoolchildren living in rural and urban areas of Ghana.</p> <p>Methods</p> <p>Data on skin prick testing, anthropometric, parasitological, demographic and lifestyle information for 1,482 schoolchildren aged 6-15 years was collected. Atopy was defined as sensitization to at least one tested allergen whilst the Centres for Disease Control and Prevention (CDC, Atlanta) growth reference charts were used in defining high BMI as BMI ≥ the 85<sup>th </sup>percentile. Logistic regression was performed to investigate the association between high BMI and atopy whilst adjusting for potential confounding factors.</p> <p>Results</p> <p>The following prevalences were observed for high BMI [Rural: 16%, Urban: 10.8%, p < 0.001] and atopy [Rural: 25.1%, Urban: 17.8%, p < 0.001]. High BMI was not associated with atopy; but an inverse association was observed between underweight and atopy [OR: 0.57, 95% CI: 0.33-0.99]. Significant associations were also observed with male sex [Rural: OR: 1.49, 95% CI: 1.06-2.08; Urban: OR: 1.90, 95% CI: 1.30-2.79], and in the urban site with older age [OR: 1.76, 95% CI: 1.00-3.07], family history of asthma [OR: 1.58, 95% CI: 1.01-2.47] and occupational status of parent [OR: 0.33, 95% CI: 0.12-0.93]; whilst co-infection with intestinal parasites [OR: 2.47, 95% CI: 1.01-6.04] was associated with atopy in the rural site. After multivariate adjustment, male sex, older age and family history of asthma remained significant.</p> <p>Conclusions</p> <p>In Ghanaian schoolchildren, high BMI was not associated with atopy. Further studies are warranted to clarify the relationship between body weight and atopy in children subjected to rapid life-style changes associated with urbanization of their environments.</p

    Quantitative Proteomics Reveals Myosin and Actin as Promising Saliva Biomarkers for Distinguishing Pre-Malignant and Malignant Oral Lesions

    Oral cancer survival rates increase significantly when it is detected and treated early. Unfortunately, clinicians now lack tests which easily and reliably distinguish pre-malignant oral lesions from those already transitioned to malignancy. A test for proteins, ones found in non-invasively-collected whole saliva and whose abundances distinguish these lesion types, would meet this critical need.To discover such proteins, in a first-of-its-kind study we used advanced mass spectrometry-based quantitative proteomics analysis of the pooled soluble fraction of whole saliva from four subjects with pre-malignant lesions and four with malignant lesions. We prioritized candidate biomarkers via bioinformatics and validated selected proteins by western blotting. Bioinformatic analysis of differentially abundant proteins and initial western blotting revealed increased abundance of myosin and actin in patients with malignant lesions. We validated those results by additional western blotting of individual whole saliva samples from twelve other subjects with pre-malignant oral lesions and twelve with malignant oral lesions. Sensitivity/specificity values for distinguishing between different lesion types were 100%/75% (p = 0.002) for actin, and 67%/83% (p<0.00001) for myosin in soluble saliva. Exfoliated epithelial cells from subjects' saliva also showed increased myosin and actin abundance in those with malignant lesions, linking our observations in soluble saliva to abundance differences between pre-malignant and malignant cells.Salivary actin and myosin abundances distinguish oral lesion types with sensitivity and specificity rivaling other non-invasive oral cancer tests.
