Solid behavior of anisotropic rigid frictionless bead assemblies
We investigate the structure and mechanical behavior of assemblies of
frictionless, nearly rigid equal-sized beads, in the quasistatic limit, by
numerical simulation. Three different loading paths are explored: triaxial
compression, triaxial extension and simple shear. Generalizing recent results
[1], we show that the material, despite rather strong finite sample size
effects, is able to sustain a finite deviator stress in the macroscopic limit,
along all three paths, without dilatancy. The shape of the yield surface is
adequately described by a Lade-Duncan (rather than Mohr-Coulomb) criterion.
While scalar state variables keep the same values as in isotropic systems,
fabric and force anisotropies are each characterized by one parameter and are
in one-to-one correspondence with principal stress ratio along all three
loading paths.The anisotropy of the pair correlation function extends to a
distance between bead surfaces on the order of 10% of the diameter. The tensor
of elastic moduli is shown to possess a nearly singular, uniaxial structure
related to stress anisotropy. Possible stress-strain relations in monotonic
loading paths are also discussed
The iron chelator Deferasirox causes severe mitochondrial swelling without depolarization due to a specific effect on inner membrane permeability
The iron chelator Deferasirox (DFX) causes severe toxicity in patients for reasons that were previously unexplained. Here, using the kidney as a clinically relevant in vivo model for toxicity together with a broad range of experimental techniques, including live cell imaging and in vitro biophysical models, we show that DFX causes partial uncoupling and dramatic swelling of mitochondria, but without depolarization or opening of the mitochondrial permeability transition pore. This effect is explained by an increase in inner mitochondrial membrane (IMM) permeability to protons, but not small molecules. The movement of water into mitochondria is prevented by altering intracellular osmotic gradients. Other clinically used iron chelators do not produce mitochondrial swelling. Thus, DFX causes organ toxicity due to an off-target effect on the IMM, which has major adverse consequences for mitochondrial volume regulation

Finite element damage analysis of an underwater glider–ship collision
Underwater gliders, which are profiling autonomous underwater vehicles designed to make oceanographic measurements, are increasingly used in the coastal ocean. As they regularly surface for data transmission, gliders increasingly pose a risk for fast ships. In order to estimate the extent of damage due to collision, 3D finite element simulations of collisions between a glider and a high-speed craft with a glass-fiber reinforced plastic hull are performed. Different collision scenarios such as impact locations, angles of attack and speeds are examined. The results are compared to an analytical solution based on simplifying assumptions. Although both methods reveal consistent results, it is shown that finite element simulations are required to account for the 3D shape of the ship. The results indicate that at ship velocities exceeding 7.5 m/s (14.6 kt) the glider penetrates the ship’s hull causing severe damage to its structure
