    Fast stereoscopic images with ray-traced volume rendering

    Journal ArticleOne of the drawbacks of standard volume rendering techniques is that it is often difficult to comprehend the three-dimensional structure of the volume from a single frame; this is especially true in cases where there is no solid surface. Generally, several frames must be generated and viewed sequentially, using motion parallax to relay depth. Another option is to generate a single stereoscopic pair, resulting in clear and unambiguous depth information in both static and moving images. Methods have been developed which take advantage of the coherence between the two halves of a stereo pair for polygon rendering and ray-tracing, generating the second half of the pair in significantly less time than that required to completely render a single image. This paper reports the results of implementing these techniques with parallel ray-traced volume rendering. In tests with different data types, the time savings is in the range of 70 - 80%

    Sero-Prevalence and Factors Associated with Toxoplasma gondii Infection among Pregnant Women Attending Antenatal Care in Mwanza, Tanzania.

    Serological screening of pregnant women for Toxoplasma gondii-specific antibodies is not practiced as an antenatal care in Tanzania; and there is a limited data about sero-prevalence of T. gondii infection in developing countries. We therefore conducted this study to determine the sero-prevalence and factors associated with T. gondii infection among pregnant women attending antenatal care clinics in Mwanza, Tanzania. Between 1st November 2012 and 31st May 2013 a total of 350 pregnant women attending antenatal care clinics in Mwanza were enrolled and screened for IgG and IgM antibodies against T. gondii using the ELISA technique. Of 350 pregnant women, 108 (30.9%) were sero-positive for T. gondii-specific antibodies. The risk of contracting T. gondii infection increases by 7% with each yearly increase in a woman's age (OR=1.07, 95% CI: 1.02 - 1.11, p=0.002). The sero-positivity rate of T. gondii-specific antibodies was higher among pregnant women from the urban than those from rural communities (41.5% versus 22.0%); [OR=2.2, 95% CI; 1.4 - 3.7, p=0.001]. Likewise employed/business women were more likely to get T. gondii infection than peasants (40.0% versus 25.9%) [OR=1.9, 95% CI: 1.2 - 3.0, p=0.006]. Sero-prevalence of T. gondii-specific antibodies is high among pregnant women in Mwanza with a significant proportion of women at risk of contracting primary T. gondii infections. Screening of T. gondii infections during antenatal care should be considered in Tanzania as the main strategy to minimize congenital toxoplasmosis

    Histochemical demonstration of endothelial superoxide and hydrogen peroxide generation in ischemic and reoxygenated rat tissues

    Objective: The aims were to test and evaluate two novel and independent histochemical methods for detecting the initial postischemic burst of superoxide and hydrogen peroxide in buffer perfused rat tissues during reflow after 60 min warm ischemia. Methods: The first is a high manganese/diaminobenzidine technique, in which superoxide oxidises Mn2+ to Mn3+, which in turn oxidizes diaminobenzidine to form amber colored polymers, observable by light microscopy. The second is a high iron/diaminobenzidine technique, in which hydrogen peroxide oxidizes diethylenetriaminepenta-acetate chelated Fe2+ to form intermediate species, which in turn oxidize diaminobenzidine similarly to Mn3+. Various isolated organs of the rat were rendered ischemic for 60 min, and reperfused with oxygen or air equilibrated buffers containing diaminobenzidine and either Mn2+ or Fe2+. Tissues were fixed by perfusion with Trump’s solution and processed for light microscopy. Results: Both manganese and iron methods consistently showed the appearance of reaction product on the luminal surfaces of arterial, capillary, and venular endothelial cells in lung, heart, and intestine of the rat during the first 2 to 3 min of reoxygenation after ischemia. The histochemical reactions were nearly absent in non-manganese treated and non-iron-treated controls. Superoxide dismutase strongly inhibited Mn2+/diaminobenzidine reaction product formation and catalase strongly inhibited Fe2+/diaminobenzidine reaction product formation, when tested in specially perfused lung preparations in which these specific antioxidant enzymes were concentrated. Conclusions: These histochemical techniques provide direct, visual evidence that a burst of reactive oxygen species is generated in postischemic rat tissues. The Mn2+/diaminobenzidine and Fe2+/diaminobenzidine techniques permit investigation of the endothelium derived reactive oxygen by simple laboratory procedures available to almost any investigator at low marginal cost. The endothelial oxidants so revealed may be of pathophysiological significance in a variety of cardiovascular disorders

    Molecular epidemiology of human rhinovirus infections in Kilifi, coastal Kenya

    This study reports pediatric surveillance over 3 years for human rhinovirus (HRV) at the District Hospital of Kilifi, coastal Kenya. Nasopharyngeal samples were collected from children presenting at outpatient clinic with no signs of acute respiratory infection, or with signs of upper respiratory tract infection, and from children admitted to the hospital with lower respiratory tract infection. Samples were screened by real-time reverse transcriptase polymerase chain reaction (real-time RT-PCR) and classified further to species by nucleotide sequencing of the VP4/VP2 junction. Of 441 HRV positives by real-time RT-PCR, 332 were classified to species, with 47% (155) being HRV-A, 5% (18) HRV-B, and 48% (159) HRV-C. There was no clear seasonal pattern of occurrence for any species. The species were present in similar proportions in the inpatient and outpatient sample sets, and no significant association between species distribution and the severity of lower respiratory tract infection in the inpatients could be determined. HRV sequence analysis revealed multiple but separate clusters in circulation particularly for HRV-A and HRV-C. Most HRV-C clusters were distinct from reference sequences downloaded from GenBank. In contrast, most HRV-A and HRV-B sequences clustered with either known serotypes or strains from elsewhere within Africa and other regions of the world. This first molecular epidemiological study of HRV in the region defines species distribution in accord with reports from elsewhere in the world, shows considerable strain diversity and does not identify an association between any species and disease severity

    Hot water treatment of seeds to control seedborne pathogens is an important tool for organic seed production. Reducing seed moisture content may have the potential to increase carrot (Daucus carota L. var. sativus D.C.) seed tolerance to treatment. Two hot water seed treatment experiments were conducted. The first studied the effect of seed moisture content (SMC), treatment temperature and treatment duration on germination. Maximum safe treatment temperature and durations were established at 50°C and 30-40 min. Germination decreased slightly from 68% at 5% SMC to 63% at 20% SMC (LSD 1.2) for all durations. The second experiment studied the effect of initial SMC and treatment durations on infestation of seed by Alternaria radicina and seed germination. Treatment at 50°C for 30 min for all SMC compared to the control resulted in a decrease in A. radicina infestation from 69.2 to 1.7%. Reducing SMC from 20 to 5% for all durations resulted in a small decrease in infestation from 25% to 18% (LSD 1.5). Reducing SMC to 5% prior to hot water treatment may be a commercially viable means of minimising reductions in seed viability and decreasing fungal infestation levels

    Electromechanical Coupling between Skeletal and Cardiac Muscle: Implications for Infarct Repair

    Skeletal myoblasts form grafts of mature muscle in injured hearts, and these grafts contract when exogenously stimulated. It is not known, however, whether cardiac muscle can form electromechanical junctions with skeletal muscle and induce its synchronous contraction. Here, we report that undifferentiated rat skeletal myoblasts expressed N-cadherin and connexin43, major adhesion and gap junction proteins of the intercalated disk, yet both proteins were markedly downregulated after differentiation into myo-tubes. Similarly, differentiated skeletal muscle grafts in injured hearts had no detectable N-cadherin or connexin43; hence, electromechanical coupling did not occur after in vivo grafting. In contrast, when neonatal or adult cardiomyocytes were cocultured with skeletal muscle, ∼10% of the skeletal myotubes contracted in synchrony with adjacent cardiomyocytes. Isoproterenol increased myotube contraction rates by 25% in coculture without affecting myotubes in monoculture, indicating the cardiomyocytes were the pacemakers. The gap junction inhibitor heptanol aborted myotube contractions but left spontaneous contractions of individual cardiomyocytes intact, suggesting myotubes were activated via gap junctions. Confocal microscopy revealed the expression of cadherin and connexin43 at junctions between myotubes and neonatal or adult cardiomyocytes in vitro. After microinjection, myotubes transferred dye to neonatal cardiomyocytes via gap junctions. Calcium imaging revealed synchronous calcium transients in cardiomyocytes and myotubes. Thus, cardiomyocytes can form electromechanical junctions with some skeletal myotubes in coculture and induce their synchronous contraction via gap junctions. Although the mechanism remains to be determined, if similar junctions could be induced in vivo, they might be sufficient to make skeletal muscle grafts beat synchronously with host myocardium
