Serum microRNA array analysis identifies miR-140-3p, miR-33b-3p and miR-671-3p as potential osteoarthritis biomarkers involved in metabolic processes.
Background: MicroRNAs (miRNAs) in circulation have emerged as promising biomarkers. In this study, we aimed to identify a circulating miRNA signature for osteoarthritis (OA) patients and in combination with bioinformatics analysis to evaluate the utility of selected differentially expressed miRNAs in the serum as potential OA biomarkers. Methods: Serum samples were collected from 12 primary OA patients, and 12 healthy individuals were screened using the Agilent Human miRNA Microarray platform interrogating 2549 miRNAs. Receiver Operating Characteristic (ROC) curves were constructed to evaluate the diagnostic performance of the deregulated miRNAs. Expression levels of selected miRNAs were validated by quantitative real-time PCR (qRT-PCR) in all serum and in articular cartilage samples from OA patients (n = 12) and healthy individuals (n = 7). Bioinformatics analysis was used to investigate the involved pathways and target genes for the above miRNAs. Results: We identified 279 differentially expressed miRNAs in the serum of OA patients compared to controls. Two hundred and five miRNAs (73.5%) were upregulated and 74 (26.5%) downregulated. ROC analysis revealed that 77 miRNAs had area under the curve (AUC) > 0.8 and p < 0.05. Bioinformatics analysis in the 77 miRNAs revealed that their target genes were involved in multiple signaling pathways associated with OA, among which FoxO, mTOR, Wnt, pI3K/akt, TGF-β signaling pathways, ECM-receptor interaction, and fatty acid biosynthesis. qRT-PCR validation in seven selected out of the 77 miRNAs revealed 3 significantly downregulated miRNAs (hsa-miR-33b-3p, hsa-miR-671-3p, and hsa-miR-140-3p) in the serum of OA patients, which were in silico predicted to be enriched in pathways involved in metabolic processes. Target-gene analysis of hsa-miR-140-3p, hsa-miR-33b-3p, and hsa-miR-671-3p revealed that InsR and IGFR1 were common targets of all three miRNAs, highlighting their involvement in regulation of metabolic processes that contribute to OA pathology. Hsa-miR-140-3p and hsa-miR-671-3p expression levels were consistently downregulated in articular cartilage of OA patients compared to healthy individuals. Conclusions: A serum miRNA signature was established for the first time using high density resolution miR-arrays in OA patients. We identified a three-miRNA signature, hsa-miR-140-3p, hsa-miR-671-3p, and hsa-miR-33b-3p, in the serum of OA patients, predicted to regulate metabolic processes, which could serve as a potential biomarker for the evaluation of OA risk and progression.Peer reviewedFinal Published versio
Visual analytics of contact tracing policy simulations during an emergency response
Epidemiologists use individual-based models to (a) simulate disease spread over dynamic contact networks and (b) to investigate strategies to control the outbreak. These model simulations generate complex ‘infection maps’ of time-varying transmission trees and patterns of spread. Conventional statistical analysis of outputs offers only limited interpretation. This paper presents a novel visual analytics approach for the inspection of infection maps along with their associated metadata, developed collaboratively over 16 months in an evolving emergency response situation. We introduce the concept of representative trees that summarize the many components of a time-varying infection map while preserving the epidemiological characteristics of each individual transmission tree. We also present interactive visualization techniques for the quick assessment of different control policies. Through a series of case studies and a qualitative evaluation by epidemiologists, we demonstrate how our visualizations can help improve the development of epidemiological models and help interpret complex transmission patterns
Inactivation of α2-Macroglobulin by Activated Human Polymorphonuclear Leukocytes
The proteolytic activity of trypsin releases the dye Remazol
Brilliant Blue from its high molecular weight substrate, the skin
powder (Hide Powder Azure, Sigma), with an increase in absorbance at
595 nm. Active α2- macroglobulin (80 μg/ml) totally inhibits the
proteolytic activity of trypsin (14 μg/ml) by trapping this
protease. But after a 20 min incubation of α2-macroglobulin at 37°C
with 2 × 106 human polymorphonuclear leukocytes activated by
N-formyl-L-methionyl-L-leucyl-L-phenylalanine
(10−7 M) and cytochalasin B (10−8 M), 100% of trypsin
activity was recovered, indicating a total inactivation of
α2-macroglobuHn. Incubation with granulocyte myeloperoxidase also
inactivates α2-macroglobulin. Hypochlorous acid, a by-product of
myeloperoxidase activity, at a concentration of 10−7 M also
inactivates α2-macroglobulin, which indicates that an important
cause of α2-macroglobulin inactivation by activated
polymorphonuclear leukocytes could be the activity of myeloperoxidase
Contribution à la mise en place d’une meilleure méthode d’estimation des données sur le paludisme en Côte d’Ivoire
peer reviewedIntroduction Selon les services sanitaires, le paludisme constitue le premier
motif de consultation et d’hospitalisation. Il y a cependant contradiction de
ces données avec celles du service national de la statistique de routine, d’après
lesquelles l’incidence des quinze dernières années varie seulement de 4%à 15 %.
L’introduction récente des tests rapides de diagnostic ne fournit pas d’éclairage
suffisant au débat, car reposant sur la notion de portage d’antigènes dont la
prévalence est habituellement élevée dans la population. Notre objectif est de
proposer une méthode d’estimation des données, en attendant que soit porter
à grande échelle le diagnostic parasitologique qui fournirait des données plus
crédibles.
Méthodes Un panel d’experts constitué pour déterminer une méthode a procédé
par brainstorming couplé à une méta-analyse.
Résultats L’approche proposée consiste à sélectionner deux districts dans
chacun des faciès épidémiologiques connus du pays. Tous les cas suspects y
seront enregistrés et documentés par goutte épaisse et frottis sur douze mois pour
tenir compte de toutes les saisons. Dans ces districts, les données de la statistique
de routine seront produites concomitamment à celles basées sur l’examen
parasitologique. Ces dernières serviront d’« étalon-or » pour les comparaisons
intra-faciès et nationale. Les facteurs correcteurs intra-faciès et national seront
déterminés par comparaison et utilisés pour ajuster les données de routine pendant
une période de cinq ans avant de déterminer à nouveaux les facteurs
correcteurs. Discussion L’enjeu du paludisme pour le pays commande que les données
relatives à ce thème soient conformes à la réalité. La présente approche devrait

Safety and technical efficacy of early minimally invasive endoscopy-guided surgery for intracerebral haemorrhage:the Dutch Intracerebral haemorrhage Surgery Trial pilot study
Background: Previous randomised controlled trials could not demonstrate that surgical evacuation of intracerebral haemorrhage (ICH) improves functional outcome. Increasing evidence suggests that minimally invasive surgery may be beneficial, in particular when performed early after symptom onset. The aim of this study was to investigate safety and technical efficacy of early minimally invasive endoscopy-guided surgery in patients with spontaneous supratentorial ICH. Methods: The Dutch Intracerebral Haemorrhage Surgery Trial pilot study was a prospective intervention study with blinded outcome assessment in three neurosurgical centres in the Netherlands. We included adult patients with spontaneous supratentorial ICH ≥10mL and National Institute of Health Stroke Scale (NIHSS) score ≥2 for minimally invasive endoscopy-guided surgery within 8 h after symptom onset in addition to medical management. Primary safety outcome was death or increase in NIHSS ≥4 points at 24 h. Secondary safety outcomes were procedure-related serious adverse events (SAEs) within 7 days and death within 30 days. Primary technical efficacy outcome was ICH volume reduction (%) at 24 h. Results: We included 40 patients (median age 61 years; IQR 51–67; 28 men). Median baseline NIHSS was 19.5 (IQR 13.3–22.0) and median ICH volume 47.7mL (IQR 29.4–72.0). Six patients had a primary safety outcome, of whom two already deteriorated before surgery and one died within 24 h. Sixteen other SAEs were reported within 7 days in 11 patients (of whom two patients that already had a primary safety outcome), none device related. In total, four (10%) patients died within 30 days. Median ICH volume reduction at 24 h was 78% (IQR 50–89) and median postoperative ICH volume 10.5mL (IQR 5.1–23.8). Conclusions: Minimally invasive endoscopy-guided surgery within 8 h after symptom onset for supratentorial ICH appears to be safe and can effectively reduce ICH volume. Randomised controlled trials are needed to determine whether this intervention also improves functional outcome. Trial registration: Clinicaltrials.gov : NCT03608423, August 1st, 2018.</p
Amyloid Precursor Protein and Proinflammatory Changes Are Regulated in Brain and Adipose Tissue in a Murine Model of High Fat Diet-Induced Obesity
Background: Middle age obesity is recognized as a risk factor for Alzheimer’s disease (AD) although a mechanistic linkage remains unclear. Based upon the fact that obese adipose tissue and AD brains are both areas of proinflammatory change, a possible common event is chronic inflammation. Since an autosomal dominant form of AD is associated with mutations in the gene coding for the ubiquitously expressed transmembrane protein, amyloid precursor protein (APP) and recent evidence demonstrates increased APP levels in adipose tissue during obesity it is feasible that APP serves some function in both disease conditions. Methodology/Principal Findings: To determine whether diet-induced obesity produced proinflammatory changes and altered APP expression in brain versus adipose tissue, 6 week old C57BL6/J mice were maintained on a control or high fat diet for 22 weeks. Protein levels and cell-specific APP expression along with markers of inflammation and immune cell activation were compared between hippocampus, abdominal subcutaneous fat and visceral pericardial fat. APP stimulation-dependent changes in macrophage and adipocyte culture phenotype were examined for comparison to the in vivo changes. Conclusions/Significance: Adipose tissue and brain from high fat diet fed animals demonstrated increased TNF-a and microglial and macrophage activation. Both brains and adipose tissue also had elevated APP levels localizing to neurons and macrophage/adipocytes, respectively. Probucol Suppresses Enterocytic Accumulation of Amyloid-β Induced by Saturated Fat and Cholesterol Feeding
Amyloid-β (Aβ) is secreted from lipogenic organs such as intestine and liver as an apolipoprotein of nascent triacylglycerol rich lipoproteins. Chronically elevated plasma Aβ may compromise cerebrovascular integrity and exacerbate amyloidosis—a hallmark feature of Alzheimer’s disease (AD). Probucol is a hypocholesterolemic agent that reduces amyloid burden in transgenic amyloid mice, but the mechanisms for this effect are presently unclear. In this study, the effect of Probucol on intestinal lipoprotein-Aβ homeostasis was explored. Wild-type mice were fed a control low-fat diet and enterocytic Aβ was stimulated by high-fat (HF) diet enriched in 10% (w/w) saturated fat and 1% (w/w) cholesterol for the duration of 1 month. Mice treated with Probucol had the drug incorporated into the chow at 1% (w/w). Quantitative immunofluorescence was utilised to determine intestinal apolipoprotein B (apo B) and Aβ abundance. We found apo B in both the perinuclear region of the enterocytes and the lacteals in all groups. However, HF feeding and Probucol treatment increased secretion of apo B into the lacteals without any change in net villi abundance. On the other hand, HF-induced enterocytic perinuclear Aβ was significantly attenuated by Probucol. No significant changes in Aβ were observed within the lacteals. The findings of this study support the notion that Probucol suppresses dietary fat induced stimulation of Aβ biosynthesis and attenuate availability of apo B lipoprotein-Aβ for secretion
