    Effect of Acute Exercise on Plasma Nitric Oxide Level in Humans

    Objective: Since nitric oxide (NO) plays an important role in regulating vascular tone and changes in the plasma concentration of the end product of NO (nitrite/nitrate) have been observed during exercise, we studied the influence of acute exercise on nitrite/nitrate plasma levels in a group of semiprofessional football players and in younger and older sedentary subjects. Methods: The subjects exercised for 10 min on a cycle ergometer and blood samples were obtained at rest and immediately after exercise. Plasma samples were analysed for nitrite/nitrate. Results: The acute physical exercise induced a significant increase in nitrite/nitrate plasma levels both in sedentary and in active subjects. No difference was evident between the younger and older subjects, both in the basal level and after exercise, indicating that the changes could not be due to age. The higher basal plasma level of nitrite/nitrate observed in the active subjects compared with sedentary groups indicates that the state of physical conditioning had an effect on resting NO concentration. Conclusion: It seems that either acute exercise, even for a short time, or training can induce an increase in circulating NO

    No more time to stay ‘single’ in the detection of Anisakis pegreffii, A. simplex (s. s.) and hybridization events between them: a multi-marker nuclear genotyping approach

    A multi-marker nuclear genotyping approach was performed on larval and adult specimens of Anisakis spp. (N = 689) collected from fish and cetaceans in allopatric and sympatric areas of the two species Anisakis pegreffii and Anisakis simplex (s. s.), in order to: (1) identify specimens belonging to the parental taxa by using nuclear markers (allozymes loci) and sequence analysis of a new diagnostic nuclear DNA locus (i.e. partial sequence of the EF1 α−1 nDNA region) and (2) recognize hybrid categories. According to the Bayesian clustering algorithms, based on those markers, most of the individuals (N = 678) were identified as the parental species [i.e. A. pegreffii or A. simplex (s. s.)], whereas a smaller portion (N = 11) were recognized as F1 hybrids. Discordant results were obtained when using the polymerase chain reaction–restriction fragment length polymorphisms (PCR–RFLPs) of the internal transcribed spacer (ITS) ribosomal DNA (rDNA) on the same specimens, which indicated the occurrence of a large number of ‘hybrids’ both in sympatry and allopatry. These findings raise the question of possible misidentification of specimens belonging to the two parental Anisakis and their hybrid categories derived from the application of that single marker (i.e. PCR–RFLPs analysis of the ITS of rDNA). Finally, Bayesian clustering, using allozymes and EF1 α−1 nDNA markers, has demonstrated that hybridization between A. pegreffii and A. simplex (s. s.) is a contemporary phenomenon in sympatric areas, while no introgressive hybridization takes place between the two species

    New parasite records for the sunfish Mola mola in the Mediterranean Sea and their potential use as biological tags for long-distance host migration

    Studies describing the parasite fauna of sunfish species from the Mediterranean Sea are to date limited, despite information gained through parasitological examination may reveal unknown ecological and biological aspects of both hosts and parasites. Moreover, recent molecular studies on sunfish taxonomy revealed the presence of two species belonging to the genus Mola in the Mediterranean basin, namely M. mola and M. alexandrini. These two fish taxa have long been synonymized or confused among them, which implies that the majority of the studies carried out so far reported the parasites infecting both species under a single host species, generally referred to as M. mola. We hereby investigated the parasite fauna of a 43 cm long M. mola specimen from the Mediterranean Sea, whose identification was confirmed by molecular tool, and provided the first evidence of the occurrence of the nematode Anisakis simplex (s.s.) and of the cestode Gymnorhynchus isuri in Mola species anywhere. The use of helminth species as biological tags for the sunfish is also discussed

    Genetic and morphological studies were carried out on acanthocephalans belonging to Corynosoma Luhe, 1904 and referable to the species C. cetaceum Johnston & Best, 1942 and C. australe Johnston, 1937, which were recovered from both definitive and intermediate hosts in Argentinian waters. The aims were to estimate the level of genetic differentiation between the two taxa at any stage of their life-cycle, to provide genetic ( allozyme) markers for their recognition and to analyse the systematic status of both taxa. Acanthocephalans were collected from the stomach and intestine of Arctocephalus australis (Zimmerman), the intestine of Mirounga leonina (Linnaeus) and the stomach of Pontoporia blainvillei Gervais & D'Orbigny (definitive hosts) in Argentinian waters. Alternative alleles at all the 13 enzymatic loci studied were observed for C. australe and C. cetaceum. The specimens from the stomach of both P. blainvillei and A. australis were identified, on the basis of the great number of diagnostic loci found, as C. cetaceum; those from intestine of both A. australis and M. leonina as C. australe. A high level of genetic differentiation (D-Nei= infinity: I-Nei= 0.00) between the two taxa was found, suggesting a generic distinction between the two species. Cystacanths of the two species from the body-cavity of the fish Cynoscion guatucupa (Cuvier) collected from the same geographical area were identified genetically. Morphological patterns, such as the number of hooks and hook rows on the proboscis, the distribution of somatic and genital armature, and other morphometric and meristic differences, in addition to ecological data, enabled the identification of these two species at cystacanth, juvenile and adult stages. However, a number of morphological and morphometric features of the Argentinian material were different to those of C. australe and C. cetaceum described from other regions of the world

    Enhanced second harmonic generation from resonant GaAs gratings

    We study second harmonic generation in nonlinear, GaAs gratings. We find large enhancement of conversion efficiency when the pump field excites the guided mode resonances of the grating. Under these circumstances the spectrum near the pump wavelength displays sharp resonances characterized by dramatic enhancements of local fields and favorable conditions for second harmonic generation, even in regimes of strong linear absorption at the harmonic wavelength. In particular, in a GaAs grating pumped at 1064nm, we predict second harmonic conversion efficiencies approximately five orders of magnitude larger than conversion rates achievable in either bulk or etalon structures of the same material.Comment: 8 page

    Broadband Brewster transmission through 2D metallic gratings

    Recently, we have introduced a mechanism to achieve ultrabroadband light funnelling and total transmission through 1D narrow metallic gratings at a specific incidence angle, the so-called plasmonic Brewster angle. This phenomenon is based on impedance matching between the guided modes supported by ultranarrow linear slits and transverse-magnetic waves at oblique incidence. In this paper, we demonstrate that such phenomenon, representing the equivalent of Brewster transmission for plasmonic screens, can also occur in 2D metallic gratings of various structural forms and shapes, and that it may be made insensitive to the azimuthal, or polarization, angle u. This finding may have relevant implications to realize large funneling, absorption and squeezing of light in perforated metallic screens

    Gap solitons in a nonlinear quadratic negative index cavity

    By integrating the full Maxwell's equations we predict the existence of gap solitons in a quadratic, Fabry-Perot negative index cavity. An intense, fundamental pump pulse shifts the band structure that forms when magnetic and electric plasma frequencies are different so that a weak, second harmonic pulse initially tuned inside the gap is almost entirely transmitted. The process is due cascading, which occurs far from phase matching conditions, and causes pulse compression. A nonlinear polarization spawns a dark soliton, while a nonlinear magnetization produces a bright soliton

    Sub-wavelength diffraction-free imaging with low-loss metal-dielectric multilayers

    We demonstrate numerically the diffraction-free propagation of sub-wavelength sized optical beams through simple elements built of metal-dielectric multilayers. The proposed metamaterial consists of silver and a high refractive index dielectric, and is designed using the effective medium theory as strongly anisotropic and impedance matched to air. Further it is characterised with the transfer matrix method, and investigated with FDTD. The diffraction-free behaviour is verified by the analysis of FWHM of PSF in the function of the number of periods. Small reflections, small attenuation, and reduced Fabry Perot resonances make it a flexible diffraction-free material for arbitrarily shaped optical planar elements with sizes of the order of one wavelength.Comment: 5 pages, 4 figure

    Genetic evidence of two sibling species within the Contracoecum ogmorhini Johnson & Mawson 1941 complex (Nematoda; Anisakidae) from otariid seals in boreal and austral regions

    Genetic variation of Contracaecum ogmorhini (sensu lato) populations from different otariid seals of the northern and southern hemisphere was studied on the basis of 18 enzyme loci as well as preliminary sequence analysis of the mitochondrial cyt b gene (260 bp). Samples were collected from Zalophus californianus in the boreal region and from Arctocephalus pusillus pusillus, A. pusillus doriferus and A. australis from the austral region. Marked genetic heterogeneity was found between C. ogmorhini (sensu lato) samples from the boreal and austral region, respectively. Two loci (Mdh-2 and NADHdh) showed fixed differences and a further three loci (Iddh, Mdh-1 and 6Pgdh) were highly differentiated between boreal and austral samples. Their average genetic distance was DNei = 0.36 at isozyme level. At mitochondrial DNA level, an average proportion of nucleotide substitution of 3.7% was observed. These findings support the existence of two distinct sibling species, for which the names C. ogmorhini (sensu stricto) and C. margolisi n. sp., respectively, for the austral and boreal taxon, are proposed. A description for C. margolisi n. sp. is provided. No diagnostic morphological characters have so far been detected; on the other hand, two enzyme loci, Mdh-2 and NADHdh, fully diagnostic between the two species, can be used for the routine identification of males, females and larval stages. Mirounga leonina was found to host C. ogmorhini (s.s.) inmixed infections with C. osculatum (s.l.) (of which C. ogmorhini (s.l.) was in the past considered to be a synonym) and C. miroungae; no hybrid genotypes were found,confirming the reproductive isolation of these three anisakid species.
