Boolean versus continuous dynamics on simple two-gene modules
We investigate the dynamical behavior of simple modules composed of two genes
with two or three regulating connections. Continuous dynamics for mRNA and
protein concentrations is compared to a Boolean model for gene activity. Using
a generalized method, we study within a single framework different continuous
models and different types of regulatory functions, and establish conditions
under which the system can display stable oscillations. These conditions
concern the time scales, the degree of cooperativity of the regulating
interactions, and the signs of the interactions. Not all models that show
oscillations under Boolean dynamics can have oscillations under continuous
dynamics, and vice versa.Comment: 8 pages, 10 figure
Media use during adolescence: the recommendations of the Italian Pediatric Society.
BACKGROUND: The use of media device, such as smartphone and tablet, is currently increasing, especially among the youngest. Adolescents spend more and more time with their smartphones consulting social media, mainly Facebook, Instagram and Twitter because. Adolescents often feel the necessity to use a media device as a means to construct a social identity and express themselves. For some children, smartphone ownership starts even sooner as young as 7 yrs, according to internet safety experts. MATERIAL AND METHODS: We analyzed the evidence on media use and its consequences in adolescence. RESULTS: In literature, smartphones and tablets use may negatively influences the psychophysical development of the adolescent, such as learning, sleep and sigh. Moreover, obesity, distraction, addiction, cyberbullism and Hikikomori phenomena are described in adolescents who use media device too frequently. The Italian Pediatric Society provide action-oriented recommendations for families and clinicians to avoid negative outcomes. CONCLUSIONS: Both parents and clinicians should be aware of the widespread phenomenon of media device use among adolescents and try to avoid psychophysical consequences on the youngest
Classification of human actions into dynamics based primitives with application to drawing tasks
We develop the study of primitives of human motion, which we refer to as movemes. The idea is to understand human motion by decomposing it into a sequence of elementary building blocks that belong to a known alphabet of dynamical systems. How can we construct an alphabet of movemes from human data? In this paper we address this issue by introducing the notion of well-posednes. Using examples from human drawing data, we show that the well-posedness notion can be applied in practice so to establish if sets of actions, viewed as signals in time, can define movemes
Enhancement of cytosine arabinoside-induced apoptosis in human myeloblastic leukemia cells by NFkB/Rel- specific decoy oligodeoxynucleotides
The activity of NF-kB/Rel nuclear factors is known to inhibit
apoptosis in various cell types. We investigated whether the
subtraction of NF-kB/Rel activity influenced the response of
11 AML (M1, M2 and M4) patients’ cells to AraC. To this
end we used a phosphorothioate double-stranded decoy oligodeoxynucleotide
(ODN) carrying the NF-kB/Rel- consensus
sequence. Cell incubation with this ODN, but not its
mutated (scrambled) form used as a control, resulted in
abating the NF-kB/Rel nuclear levels in these cells, as verified
by electrophoretic mobility shift assay (EMSA) of cells’
nuclear extracts. We incubated the leukemic cells with AraC
(32 or 1 mM), in either the absence or presence of the decoy or the scrambled ODN, and analyzed cell apoptosis. The
spontaneous cell apoptosis detectable in the absence of
AraC (,25%) was not modulated by the oligonucleotide
presence in cell cultures. On the other hand, in 10 of the 11
samples tested, the decoy kB, but not the scrambled ODN
significantly (P ,0.01 in a Student’s t test) enhanced cell
apoptotic response to AraC. Such an effect was particularly
remarkable at low AraC doses (1 mM). These findings indicate
that NF-kB/Rel activity influences response to AraC in
human primary myeloblastic cells, and suggests that the
inhibition of NF-kB/Rel factors can improve the effect of
chemotherapy in AM

Cow's milk allergy (CMA) is one of the most common food allergies in children. Epigenetic mechanisms have been suggested to play a role in CMA pathogenesis. We shown that DNA methylation of Th1/Th2 cytokine genes and FoxP3 affects CMA disease course. Preliminary evidence suggest that also the miRNome could be implicated in the pathogenesis of allergy. Main study outcome was to comparatively evaluate miRNome in children with CMA and in healthy controls

Retinoic Acid Specifically Enhances Embryonic Stem Cell Metastate Marked by Zscan4
Pluripotency confers Embryonic Stem Cells (ESCs) the ability to differentiate in ectoderm, endoderm, and mesoderm derivatives, producing the majority of cell types. Although the majority of ESCs divide without losing pluripotency, it has become evident that ESCs culture consists of multiple cell populations with different degrees of potency that are spontaneously induced in regular ESC culture conditions. Zscan4, a key pluripotency factor, marks ESC subpopulation that is referred to as high-level of pluripotency metastate. Here, we report that in ESC cultures treated with retinoic acid (RA), Zscan4 ESCs metastate is strongly enhanced. In particular, we found that induction of Zscan4 metastate is mediated via RA receptors (RAR-alpha, RAR-beta, and RAR-gamma), and it is dependent on phosphoinositide-3-kinase (PI3K) signaling. Remarkably, Zscan4 metastate induced by RA lacks canonical pluripotency genes Oct3/4 and Nanog but retained both self-renewal and pluripotency capabilities. Finally we demonstrated that the conditional ablation of Zscan4 subpopulation is dispensable for both endoderm and mesoderm but is required for ectoderm lineage. In conclusion, our research provides new insights about the role of RA signaling during ESCs high pluripotency metastate fluctuation
Triple Michelson Interferometer for a Third-Generation Gravitational Wave Detector
The upcoming European design study `Einstein gravitational-wave Telescope'
represents the first step towards a substantial, international effort for the
design of a third-generation interferometric gravitational wave detector. It is
generally believed that third-generation instruments might not be installed
into existing infrastructures but will provoke a new search for optimal
detector sites. Consequently, the detector design could be subject to fewer
constraints than the on-going design of the second generation instruments. In
particular, it will be prudent to investigate alternatives to the traditional
L-shaped Michelson interferometer. In this article, we review an old proposal
to use three Michelson interferometers in a triangular configuration. We use
this example of a triple Michelson interferometer to clarify the terminology
and will put this idea into the context of more recent research on
interferometer technologies. Furthermore the benefits of a triangular detector
will be used to motivate this design as a good starting point for a more
detailed research effort towards a third-generation gravitational wave

Relative Entropy of Coherent States on General CCR Algebras
For a subalgebra of a generic CCR algebra, we consider the relative entropy between a general (not necessarily pure) quasifree state and a coherent excitationthereof. We give a unified formula for this entropy in terms of single-particle modular data. Further, we investigate changes of the relative entropy along subalgebras arising from an increasing family of symplectic subspaces; here convexity of the entropy (as usually considered for the Quantum Null Energy Condition) is replaced with lower estimates for the second derivative, composed of “bulk terms” and “boundary terms”. Our main assumption is that the subspaces are in differential modular position, a regularity condition that generalizes the usual notion of half-sided modular inclusions.
