    Giant vesicles at the prolate-oblate transition: A macroscopic bistable system

    Giant phospholipid vesicles are shown to exhibit thermally activated transitions between a prolate and an oblate shape on a time scale of several seconds. From the fluctuating contour of such a vesicle we extract ellipticity as an effective reaction coordinate whose temporal probability distribution is bimodal. We then reconstruct the effective potential from which we derive an activation energy of the order of kBTk_BT in agreement with theoretical calculations. The dynamics of this transition is well described within a Kramers model of overdamped diffusion in a bistable potential.     ROSAT HRI observations of Centaurus A

    We present results from a sensitive high-resolution X-ray observation of the nearby active galaxy Centaurus A (NGC 5128) with the ROSAT HRI. The 65~ksec X-ray image clearly distinguishes different components of the X-ray emission from Cen A: the nucleus and the jet, the diffuse galaxy halo, and a number of individual sources associated with the galaxy. The luminosity of the nucleus increased by a factor of two compared to an earlier ROSAT observation in 1990. The high spatial resolution of the ROSAT HRI shows that most of the knots in the jet are extended both along and perpendicular to the jet axis. We report the detection of a new X-ray feature, at the opposite side of the X-ray jet which is probably due to compression of hot interstellar gas by the expanding southwestern inner radio lobe.Comment: To be published in Astrophys.     Pré-compostagem de cadáveres de bovinos acometidos pelo botulismo.

    Foram pré-compostados, em células individuais e isoladas, cinco cadáveres de bovinos acometidos pelo botulismo com a finalidade de monitorar a presença de esporos de Clostridium botulinum e de toxina botulínica antes e após o processo de decomposição em leira estática com fonte de carbono. O diagnóstico da intoxicação nos animais foi baseado nas características clínico-patológicas, epidemiológicas ou laboratoriais. Dos cinco bovinos com evolução clínica crônica de botulismo, cujos cadáveres foram pré-compostados, três foram acometidos pela toxina tipo D, um pelo complexo CD e um dos animais foi negativo na tentativa de detecção da toxina e de esporos da bactéria nas vísceras pelo bioensaio e neutralização em camundongo. O processo de pré-compostagem foi realizado em leira estática, com o uso de material carbonáceo umidificado como substrato e esquartejamento do animal, vedado individualmente com lona plástica e sem aeração por um período de 50 dias. A temperatura das leiras foi monitorada durante o período e oscilou de 40,5-52,4ºC. Após a abertura das leiras, pôde-se constatar a completa decomposição de todo material mole, com redução significativa do seu peso (de 26,5-44,5%), restando apenas os ossos. Não foi detectado esporo ou toxina botulínica no interior dos ossos (n=5 para cada cadáver). Nas 200 amostras examinadas do homogeneizado restante (n=40 para cada cadáver), em apenas duas amostras de uma leira foram detectados esporos de C. botulinum tipo C, enquanto que todas foram negativas para a tentativa de detecção da toxina botulínica pelo bioensaio em camundongo. Da forma como foi avaliado o processo de pré-compostagem de bovinos mortos pela intoxicação botulínica não contribuiu para a proliferação de C. botulinum. Five carcasses of cattle that died from botulism were composted in individual isolated beds with the purpose of monitoring the effect on spores of Clostridium botulinum and of botulinum toxin. The diagnosis of botulinum poisoning was based on clinical and pathological, epidemic and/or laboratorial characteristics. Of the five bovines with clinical signs of chronic botulism, three were affected by the type D toxin, one by the CD complex, and one was negative for toxin and spores of C. botulinum in the viscera using a mouse bioassay. The composting process was carried out in beds with carcasses cut up and mixed with sawdust as a carbon source. They were kept anaerobic under a plastic sheet for 50 days. The temperature of the beds was monitored during the period and oscillated from 40.5 to 52.4ºC. After this time, soft tissue had complete decomposed with a significant loss of weight (26.5-44.5%). The remaining bones no longer contained spores or botulinum toxin in the 5 samples taken from each carcass. Only 2 of the 200 samples of homogenized remaining (40 for each carcass) contained spores of C. botulinum type C. All samples were negative for botulinum toxin using the mouse bioassay.     On Shape Transformations and Shape Fluctuations of Cellular Compartments and Vesicles

    We discuss the shape formation and shape transitions of simple bilayer vesicles in context with their role in biology. In the first part several classes of shape changes of vesicles of one lipid component are described and it is shown that these can be explained in terms of the bending energy concept in particular augmented by the bilayer coupling hypothesis. In the second part shape changes and vesicle fission of vesicles composed of membranes of lipid mixtures are reported. These are explained in terms of coupling between local curvature and phase separation

    Effects of shading on biological nitrogen fixation in bean plants

    Conduziram-se dois experimentos com o objetivo de verificar o efeito da redução da luminosidade e da temperatura, causado pelo sombreamento, na fixação biológica do N2 em feijoeiro (Phaseolus vulgaris L.). Quando se realizou um sombreamento de 25%, houve um aumento, em relação à luminosidade total, do peso de matéria seca, atividade da nitrogenase, nitrogênio total transportado na seiva do xilema, percentagem de N transportado sob a forma de ureídos, e do nitrogênio-total acumulado na parte aérea. Nesse nível de sombreamento, o aumento na eficiência nodular compensou a redução no peso dos nódulos. O efeito favorável do sombreamento de 25% foi atribuído à redução das temperaturas máximas diurnas nos vasos sombreados e a redução da energia solar foi compensada por um aumento na área foliar. No nível de sombreamento de 40%, embora ocorresse redução da temperatura na zona radicular e aumento na área foliar, isso não foi suficiente para compensar a menor atividade fotossintética. De modo geral, as temperaturas elevadas não prejudicaram a formação e crescimento dos nódulos, mas sim o seu funcionamento, refletido na menor atividade da nitrogenase e no menor acúmulo de nitrogênio na parte aérea.Two experiments were performed to investigate the effect of decreasing luminosity and temperature caused by shading on biological nitrogen fixation of common beans (Phaseolus vulgaris L.). When the shading was at 25% level, there was an increase, in relation to full light, of the dry weight, nitrogenase activity, total nitrogen in xylem sap, rate of N as ureides and total nitrogen in shoots. At this level of shading, an increase in nodular efficiency compensated for the decrease in nodule weight. The favourable effect of reducing 25% of the luminosity was caused by the decrease in maximal diurnal temperature in the shaded pots and the decrease of solar energy was compensated by and increase in leaf area. At 40% shading, there was a decrease in the rhizosphere temperature and an increase in leaf area but this was not enough to compensate for the lower photosynthetic activity. Generally the high temperature did not affect the infection process and the growth of the nodules, but altered nodule metabolism, decreasing nitrogenase activity and total nitrogen in shoots

    Vesicle shape, molecular tilt, and the suppression of necks

    Can the presence of molecular-tilt order significantly affect the shapes of lipid bilayer membranes, particularly membrane shapes with narrow necks? Motivated by the propensity for tilt order and the common occurrence of narrow necks in the intermediate stages of biological processes such as endocytosis and vesicle trafficking, we examine how tilt order inhibits the formation of necks in the equilibrium shapes of vesicles. For vesicles with a spherical topology, point defects in the molecular order with a total strength of +2+2 are required. We study axisymmetric shapes and suppose that there is a unit-strength defect at each pole of the vesicle. The model is further simplified by the assumption of tilt isotropy: invariance of the energy with respect to rotations of the molecules about the local membrane normal. This isotropy condition leads to a minimal coupling of tilt order and curvature, giving a high energetic cost to regions with Gaussian curvature and tilt order. Minimizing the elastic free energy with constraints of fixed area and fixed enclosed volume determines the allowed shapes. Using numerical calculations, we find several branches of solutions and identify them with the branches previously known for fluid membranes. We find that tilt order changes the relative energy of the branches, suppressing thin necks by making them costly, leading to elongated prolate vesicles as a generic family of tilt-ordered membrane shapes.Comment: 10 pages, 7 figures, submitted to Phy.
