    Ligand regulation of the quaternary organization of cell surface M3 muscarinic acetylcholine receptors analyzed by fluorescence resonance energy transfer (FRET) imaging and homogenous time-resolved FRET

    Flp-In T-REx 293 cells expressing a wild type human M muscarinic acetylcholine receptor construct constitutively and able to express a Receptor Activated Solely by Synthetic Ligand (RASSL) form of this receptor on demand maintained response to the muscarinic agonist carbachol but developed response to clozapine-N-oxide only upon induction of the RASSL. The two constructs co-localized at the plasma membrane and generated strong ratiometric fluorescence resonance energy transfer (FRET) signals consistent with direct physical interactions. Increasing levels of induction of the FRET-donor RASSL did not alter wild type receptor FRET-acceptor levels substantially. However, ratiometric FRET was modulated in a bell-shaped fashion with maximal levels of the donor resulting in decreased FRET. Carbachol, but not the antagonist atropine, significantly reduced the FRET signal. Cell surface homogenous time-resolved FRET, based on SNAP-tag technology and employing wild type and RASSL forms of the human M receptor expressed stably in Flp-In TREx 293 cells, also identified cell surface dimeric/oligomeric complexes. Now, however, signals were enhanced by appropriate selective agonists. At the wild type receptor large increases in FRET signal to carbachol and acetylcholine were concentration-dependent with EC values consistent with the relative affinities of the two ligands. These studies confirm the capacity of the human M muscarinic acetylcholine receptor to exist as dimeric/oligomeric complexes at the surface of cells and demonstrate that the organization of such complexes can be modified by ligand binding. However, conclusions as to the effect of ligands on such complexes may depend on the approach used

    The aim of this work is to explore the possibility of transitioning a fuel powered island to a renewable powered one. This transition is analyzed for the real MV/LV distribution system of the island of Pantelleria, in the Mediterranean Sea. Particularly, this work is focused on a renewable source nowadays totally unused: wave energy. Thanks to the innovative generator prototype designed by Department of Energy of University of Palermo (Italy), wave energy is able to represent a primary source for the production of electric energy in the Mediterranean islands. The procedures applied in the present article, as well as the main equations used, are the result of previous applications made in different technical fields that show a good replicability

    Desalination is commonly adopted nowadays to overcome the freshwater scarcity in some areas of the world if brackish water or salt water is available. Different kinds of technologies have been proposed in the last century. In this paper, the state of the mainstream solutions is reported, showing the current commercial technologies like reverse osmosis (RO), Multi-Stages Flash desalination (MSF) and Multi-Effect Distillation (MED), and the new frontiers of the research with the aim of exploiting renewable sources such as wind, solar and biomass energy. In these cases, seawater treatment plants are the same as traditional ones, with the only difference being that they use a renewable energy source. Thus, classifications are firstly introduced, considering the working principles, the main energy input required for the treatment, and the potential for coupling with renewable energy sources. Each technology is described in detail, showing how the process works and reporting some data on the state of development. Finally, a statistical analysis is given concerning the spread of the various technologies across the world and which of them are most exploited. In this section, an important energy and exergy analysis is also addressed to quantify energy losses

Research finds this field very promising in the future of renewable energies, especially in areas where there are specific climatic and morphological characteristics to exploit large amounts of energy from the sea. In general, this kind of energy is referred to as six energy resources: waves, tidal range, tidal current, ocean current, ocean thermal energy conversion, and saline gradient. This review has the aim to list several wave-energy converter power plants and to analyze their years of operation. In this way, a focus is created to understand how many wave-energy converter plants work on average and whether it is indeed an established technology

    The Spitzer search for the transits of HARPS low-mass planets - II. Null results for 19 planets

    Short-period super-Earths and Neptunes are now known to be very frequent around solar-type stars. Improving our understanding of these mysterious planets requires the detection of a significant sample of objects suitable for detailed characterization. Searching for the transits of the low-mass planets detected by Doppler surveys is a straightforward way to achieve this goal. Indeed, Doppler surveys target the most nearby main-sequence stars, they regularly detect close-in low-mass planets with significant transit probability, and their radial velocity data constrain strongly the ephemeris of possible transits. In this context, we initiated in 2010 an ambitious Spitzer multi-Cycle transit search project that targeted 25 low-mass planets detected by radial velocity, focusing mainly on the shortest-period planets detected by the HARPS spectrograph. We report here null results for 19 targets of the project. For 16 planets out of 19, a transiting configuration is strongly disfavored or firmly rejected by our data for most planetary compositions. We derive a posterior probability of 83% that none of the probed 19 planets transits (for a prior probability of 22%), which still leaves a significant probability of 17% that at least one of them does transit. Globally, our Spitzer project revealed or confirmed transits for three of its 25 targeted planets, and discarded or disfavored the transiting nature of 20 of them. Our light curves demonstrate for Warm Spitzer excellent photometric precisions: for 14 targets out of 19, we were able to reach standard deviations that were better than 50ppm per 30 min intervals. Combined with its Earth-trailing orbit, which makes it capable of pointing any star in the sky and to monitor it continuously for days, this work confirms Spitzer as an optimal instrument to detect sub-mmag-deep transits on the bright nearby stars targeted by Doppler surveys.Comment: Accepted for publication in Astronomy and Astrophysics. 23 pages, 21 figure

    Evidence of vacuum birefringence from the polarisation of the optical emission from an Isolated Neutron Star

    Isolated Neutron Stars are some of the most exciting stellar objects known to astronomers: they have the most extreme magnetic fields, with values up to 101510^{15} G, and, with the exception of stellar-mass black holes, they are the most dense stars, with densities of ≈1014\approx 10^{14} g cm−3^{-3}. As such, they are perfect laboratories to test theories of electromagnetism and nuclear physics under conditions of magnetic field and density unattainable on Earth. In particular, the interaction of radiation with strong magnetic fields is the cause of the {\em vacuum birefringence}, an effect predicted by quantum electrodynamics in 1936 but that lacked an observational evidence until now. Here, we show how the study of the polarisation of the optical radiation from the surface of an isolated neutron star yielded such an observational evidence, opening exciting perspectives for similar studies at other wavelengths.Comment: 5 pages, 1 figure, Contributed to the 13th Patras Workshop on Axions, WIMPs and WISPs, Thessaloniki, May 15 to 19, 201
