    Evaluation of Front Detection Methods for Satellite-Derived SST Data Using In Situ Observations

    Sea surface temperature (SST) fronts detected in Advanced Very High Resolution Radiometer (AVHRR) data using automated edge-detection algorithms were compared to fronts found in continuous measurements of SST made aboard a ship of opportunity. Two histograms (a single-image and a multi-image method) and one gradient algorithm were tested for the occurrence of two types of errors: (a) the detection of false fronts and (b) the failure to detect fronts observed in the in situ data. False front error rates were lower for the histogram methods (27%–28%) than for the gradient method (45%). Considering only AVHRR fronts for which the SST gradient along the ship track was greater than 0.1°C km−1, error rates drop to 14% for the histogram methods and 29% for the gradient method. Missed front error rates were lower using the gradient method (16%) than the histogram methods (30%). This error rate drops significantly for the histogram methods (5%–10%) if fronts associated with small-scale SST features (km) are omitted from the comparison. These results suggest that frontal climatologies developed from the application of automated edge-detection methods to long time series of AVHRR images provide acceptably accurate statistics on front occurrence

    Observation of diurnal variations in mesoscale eddy sea-surface currents using GOCI data

    The surface current field of a mesoscale eddy in the East Sea (Sea of Japan) was derived from consecutive Geostationary Ocean Color Imager chlorophyll-a (chl-a) concentration images using the normalized maximum cross-correlation method. The estimated current field of the eddy exhibited anticyclonic structure demonstrated by the objective dynamic thresholds of correlation coefficients. The eddy periphery was defined by fitting an ellipse to subjectively selected points from the frontal map of chl-a concentration data. Radial distribution and hourly variation of the current speed around the eddy were presented. In terms of the magnitude and direction, the estimated current field was in good agreement with altimeter-based current field and current vectors from surface drifters. Diurnal variations in the current speeds of the mesoscale eddies showed a quadratic relation to the wind speed

    Resistance of Dictyostelium discoideum membranes to saponin permeabilization

    <p>Abstract</p> <p>Background</p> <p>Saponin is a mild detergent commonly used to permeabilize cells prior to immunofluorescence labeling of intracellular proteins. It has previously been used to that effect in <it>Dictyostelium discoideum </it>amoebae.</p> <p>Findings</p> <p>We show that saponin, contrary to Triton X-100 or alcohol, permeabilizes at best incompletely membranes of <it>Dictyostelium</it>. In cells exposed to osmotic stress, almost complete resistance to saponin permeabilization was observed.</p> <p>Conclusions</p> <p>Saponin should be used with special care to permeabilize <it>Dictyostelium </it>membranes. This unsusual property is presumably linked to the specific sterol composition of <it>Dictyostelium </it>membranes. It may also represent an adaptation of <it>Dictyostelium </it>to harsh conditions or to natural compounds encountered in its natural environment.</p

    Design, Realization, and Characterization of Advanced Adhesives for Joining Ultra-Stable C/C Based Components

    The aim of this work is to develop high-performance adhesives to join carbon fiber reinforced composites (C/C) for use in aerospace applications; in order to guarantee sound mechanical strength, a low coefficient of thermal expansion, and ease of application on large components. Several different adhesive formulations, based on phenolic or cyanate-ester resins (charged with the maximum experimentally feasible amount of carbon-based fillers), are developed and tested. The measurements of the lap shear strength at room temperature of the C/C joined by means of one phenolic and one cyanate ester-based resin demonstrates that these formulations are the most suitable for the given application. A complete characterization, by means of viscosimetry, dilatometry, and thermal gravimetric analysis, coupled with gas analysis by means of mass spectroscopy, confirms that the phenolic-based formulation is the most promising joining material. A nano-indenter is used to obtain its Young modulus and hardness, both inside the joint and as a bulk cured adhesive

    Properties of small molecular drug loading and diffusion in a fluorinated PEG hydrogel studied by ^1H molecular diffusion NMR and ^(19)F spin diffusion NMR

    R_f-PEG (fluoroalkyl double-ended poly(ethylene glycol)) hydrogel is potentially useful as a drug delivery depot due to its advanced properties of sol–gel two-phase coexistence and low surface erosion. In this study, ^1H molecular diffusion nuclear magnetic resonance (NMR) and ^(19)F spin diffusion NMR were used to probe the drug loading and diffusion properties of the R_f-PEG hydrogel for small anticancer drugs, 5-fluorouracil (FU) and its hydrophobic analog, 1,3-dimethyl-5-fluorouracil (DMFU). It was found that FU has a larger apparent diffusion coefficient than that of DMFU, and the diffusion of the latter was more hindered. The result of ^(19)F spin diffusion NMR for the corresponding freeze-dried samples indicates that a larger portion of DMFU resided in the R_f core/IPDU intermediate-layer region (where IPDU refers to isophorone diurethane, as a linker to interconnect the R_f group and the PEG chain) than that of FU while the opposite is true in the PEG–water phase. To understand the experimental data, a diffusion model was proposed to include: (1) hindered diffusion of the drug molecules in the R_f core/IPDU-intermediate-layer region; (2) relatively free diffusion of the drug molecules in the PEG-water phase (or region); and (3) diffusive exchange of the probe molecules between the above two regions. This study also shows that molecular diffusion NMR combined with spin diffusion NMR is useful in studying the drug loading and diffusion properties in hydrogels for the purpose of drug delivery applications

    Management of Myelodysplastic Syndrome Relapsing after Allogeneic Hematopoietic Stem Cell Transplantation: A Study by the French Society of Bone Marrow Transplantation and Cell Therapies

    To find out prognostic factors and to investigate different therapeutic approaches, we report on 147 consecutive patients who relapsed after allogeneic hematopoietic stem cell transplantation (allo-HSCT) for myelodysplastic syndrome (MDS). Sixty-two patients underwent immunotherapy (IT group, second allo-HSCT or donor lymphocyte infusion), 39 received cytoreductive treatment alone (CRT group) and 46 were managed with palliative/supportive cares (PSC group). Two-year rates of overall survival (OS) were 32%, 6%, and 2% in the IT, CRT, and PSC groups, respectively (P < .001). In multivariate analysis, 4 factors adversely influenced 2-year rates of OS: history of acute graft-versus-host disease (hazard ratio [HR], 1.83; 95% confidence interval [CI], 1.26 to 2.67; P ¼ .002), relapse within 6 months (HR, 2.69; 95% CI, .82 to 3.98; P < .001), progression to acute myeloid leukemia (HR, 2.59; 95% CI, 1.75 to 3.83; P < .001), and platelet count < 50 G/L at relapse (HR, 1.68; 95% CI, 1.15 to 2.44; P ¼.007). A prognostic score based on those factors discriminated 2 risk groups with median OSs of 13.2 versus 2.4 months, respectively (P < .001). When propensity score, prognostic score, and treatment strategy were included in Cox model, immunotherapy was found to be an independent factor that favorably impacts OS (HR, .40; 95% CI, .26 to .63; P < .001). In conclusion, immunotherapy should be considered when possible for MDS patients relapsing after allo-HSCT
