    Epitope characterization of sero-specific monoclonal antibody to Clostridium botulinum neurotoxin type A.

    Botulinum neurotoxins (BoNTs) are extremely potent toxins that can contaminate foods and are a public health concern. Anti-BoNT antibodies have been described that are capable of detecting BoNTs; however there still exists a need for accurate and sensitive detection capabilities for BoNTs. Herein, we describe the characterization of a panel of eight monoclonal antibodies (MAbs) generated to the non-toxic receptor-binding domain of BoNT/A (H(C)50/A) developed using a high-throughput screening approach. In two independent hybridoma fusions, two groups of four IgG MAbs were developed against recombinant H(C)50/A. Of these eight, only a single MAb, F90G5-3, bound to the whole BoNT/A protein and was characterized further. The F90G5-3 MAb slightly prolonged time to death in an in vivo mouse bioassay and was mapped by pepscan to a peptide epitope in the N-terminal subdomain of H(C)50/A (H(CN)25/A) comprising amino acid residues (985)WTLQDTQEIKQRVVF(999), an epitope that is highly immunoreactive in humans. Furthermore, we demonstrate that F90G5-3 binds BoNT/A with nanomolar efficiency. Together, our results indicate that F90G5-3 is of potential value as a diagnostic immunoreagent for BoNT/A capture assay development and bio-forensic analysis

    Aerogel Scattering Filters for Cosmic Microwave Background Observations

    We present the design and performance of broadband and tunable infrared-blocking filters for millimeter and sub-millimeter astronomy composed of small scattering particles embedded in an aerogel substrate. The ultra-low-density (< 100 mg/cu cm) aerogel substrate provides an index of refraction as low as 1.05, removing the need for anti-reflection coatings and allowing for broadband operation from DC to above 1 THz. The size distribution of the scattering particles can be tuned to provide a variable cutoff frequency. Aerogel filters with embedded high-resistivity silicon powder are being produced at 40-cm diameter to enable large-aperture cryogenic receivers for cosmic microwave background polarimeters, which require large arrays of sub-Kelvin detectors in their search for the signature of an inflationary gravitational-wave background

    Harlequin Syndrome with Crossed Sympathetic Deficit of the Face and Arm

    Harlequin syndrome is characterized by unilateral hyperhidrosis and flushing, which are predominantly induced by heat or exercise. Usually, the sympathetic deficits confine to the face. Rarely, the autonomic deficits involve the arm or the parasympathetic neurons in the ciliary ganglia. We report a 43-yr-old woman who presented with facial flushing and sweating in the right side, which were mainly induced by exercise. The facial flushing accompanied relative coldness in the right arm. Valsalva maneuver, cold pressure and 0.125% pilocarpine test, and computed tomography of the chest were normal. The crossed sympathetic deficit in the left face and right arm suggested that the lesions were multifocal. The sympathetic impairment in our patient may lie on a spectrum of pre- and postganglionic autonomic dysfunction, which was observed in Holmes-Adie, Ross, and Guillain-Barré syndrome

    Freight transportation demand is projected to grow to 27.5 billion tons in 2040, and to nearly 30.2 billion tons in 2050. This report describes the current and future demand for freight transportation in terms of tons and ton-miles of commodities moved by truck, rail, water, pipeline, and air freight carriers. It outlines the economic, logistics, transportation, and policy and regulatory factors that shape freight demand, the trends and 2050 outlook for these factors, and their anticipated effect on freight demand. After describing federal policy actions that could influence future freight demand, the report then summarizes the capabilities of available analytical models for forecasting freight demand. This is one in a series of reports produced as a result of the Transportation Energy Futures project, a Department of Energy-sponsored multi-agency effort to pinpoint underexplored strategies for reducing GHGs and petroleum dependence related to transportation
