    Mycobacterium tuberculosis acg Gene Is Required for Growth and Virulence In Vivo

    Mycobacterium tuberculosis dosRS two-component regulatory system controls transcription of approximately 50 genes including hspX, acg and Rv2030c, in response to hypoxia and nitric oxide conditions and within macrophages and mice. The hspX lies between acg and Rv2030c. However, the functions of the dosR regulated genes in vitro and in vivo are largely unknown. Previously, we demonstrated that deletion of hspX gene produced a mutant which grew faster in macrophages and in mice. In this study, we attempted to determine the functions of acg and Rv2030c by gene inactivation. We demonstrate that Rv2030c is dispensable for virulence and growth. However, deletion of acg produced a mutant which is attenuated in both resting and activated macrophages and in acute and persistent murine infection models. Surprisingly, deletion of acg did not compromise the viability of the mutant to nitrosative and oxidative stresses in vitro and in vivo. In addition, when the WT and the acg mutants were treated with antibiotics such as the prodrugs nitrofurantoin and nitrofuran, the acg mutant became more sensitive than the WT strain to these drugs. This suggests that Acg may not function as a nitroreductase. These data indicate that acg encodes an essential virulence factor for M. tuberculosis and enables it to grow and survive in macrophages and in mouse organs

    Prime ministers and the electoral cost of using the confidence vote in legislative bargaining: evidence from France

    Do prime ministers pay an electoral penalty for using procedural force to pass laws? Influential theories of parliamentary governance and legislative bargaining assume that the use of the confidence vote procedure – parliamentary governments’ most powerful legislative weapon – entails an electoral cost, but evidence on this important claim has been scarce. This article provides the first estimates of how prime ministers’ public approval responds to their use of the confidence vote. Analysing time series data from France 1979–2008, it is found that prime ministers experience a considerable drop in approval after their use of the confidence vote that is not accounted for by standard economic and political covariates. The effect size is similar to a 1 per cent decline in economic growth. The findings help explain French prime ministers’ selective use of the confidence vote procedure. They also suggest that political costs constrain the bargaining power conferred by the confidence vote

    A Video Game-Integrated Electromyography Biofeedback Device for Use in Physical Therapy

    We present a novel system to be used in the rehabilitation of patients with forearm injuries. The system uses surface electromyography (sEMG) recordings from a wireless sleeve to control video games designed to provide engaging biofeedback to the user. An integrated hardware/software system uses a neural net to classify the signals from a user’s muscles as they perform one of a number of common forearm physical therapy exercises. These classifications are used as input for a suite of video games that have been custom-designed to hold the patient’s attention and decrease the risk of noncompliance with the physical therapy regimen necessary to regain full function in the injured limb. The data is transmitted wirelessly from the on-sleeve board to a laptop computer using a custom-designed signal-processing algorithm that filters and compresses the data prior to transmission. We believe that this system has the potential to significantly improve the patient experience and efficacy of physical therapy using biofeedback that leverages the compelling nature of video games
