    Investigations of a THGEM-based imaging detector

    We present the results of our recent studies on a Thick Gas Electron Multiplier (THGEM)-based imaging detector prototype. It consists of two 100x100 mm^2 THGEM electrodes in cascade, coupled to a resistive anode. The event location is recorded with a 2D double-sided readout electrode equipped with discrete delay-lines and dedicated electronics. The THGEM electrodes, produced by standard printed-circuit board and mechanical drilling techniques, a 0.4 mm thick with 0.5 mm diameter holes spaced by 1 mm. Localization resolutions of about 0.7 mm (FWHM) were measured with soft x-rays, in a detector operated with atmospheric-pressure Ar/CH4; good linearity and homogeneity were achieved. We describe the imaging-detector layout, the resistive-anode 2D readout system and the imaging properties. The THGEM has numerous potential applications that require large-area imaging detectors, with high-rate capability, single-electron sensitivity and moderate (sub-mm) localization resolution.Comment: Published in JINST, 22 pages, 18 figure

    Female mate choice across spatial scales: influence of lek and male attributes on mating success of blue-crowned manakins

    Lekking males compete for females within and among leks, yet female choice is expected to work differently at each of these spatial scales. We used paternity analyses to examine how lek versus male attributes influence mate choice in the blue-crowned manakin Lepidothrix coronata. We tested the hypotheses that females prefer (i) to mate at larger leks where a larger number of potential mates can be assessed, (ii) to mate with unrelated or highly heterozygous males expected to produce high-quality offspring, (iii) to mate with males that display at higher rates, and that (iv) display honestly reflects male genetic quality. Our results show that (i) males at larger leks are not more likely to sire young, although females nesting close to small leks travel further to reach larger leks, (ii) siring males are not less related to females or more heterozygous than expected, (iii) within a lek, high-display males are more likely to sire young, and (iv) both male heterozygosity and display rate increased with lek size, and as a result display does not reliably reflect male genetic quality across leks. We suggest that female mate choice in this species is probably driven by a Fisherian process rather than adaptive genetic benefits

    Globally dispersed Y chromosomal haplotypes in wild and domestic sheep

    To date, investigations of genetic diversity and the origins of domestication in sheep have utilised autosomal microsatellites and variation in the mitochondrial genome. We present the first analysis of both domestic and wild sheep using genetic markers residing on the ovine Y chromosome. Analysis of a single nucleotide polymorphism (oY1) in the SRY promoter region revealed that allele A-oY1 was present in all wild bighorn sheep (Ovis canadensis), two subspecies of thinhorn sheep (Ovis dalli), European Mouflon (Ovis musimon) and the Barbary (Ammontragis lervia). A-oY1 also had the highest frequency (71.4%) within 458 domestic sheep drawn from 65 breeds sampled from Africa, Asia, Australia, the Caribbean, Europe, the Middle East and Central Asia. Sequence analysis of a second locus, microsatellite SRYM18, revealed a compound repeat array displaying fixed differences, which identified bighorn and thinhorn sheep as distinct from the European Mouflon and domestic animals. Combined genotypic data identified 11 male-specific haplotypes that represented at least two separate lineages. Investigation of the geographical distribution of each haplotype revealed that one (H6) was both very common and widespread in the global sample of domestic breeds. The remaining haplotypes each displayed more restricted and informative distributions. For example, H5 was likely founded following the domestication of European breeds and was used to trace the recent transportation of animals to both the Caribbean and Australia. A high rate of Y chromosomal dispersal appears to have taken place during the development of domestic sheep as only 12.9% of the total observed variation was partitioned between major geographical regions

    Susceptibility of Italian agile frog populations to an emerging strain of Ranavirus parallels population genetic diversity.

    Western populations of the Italian agile frog (Rana latastei) experience widespread genetic depletion. Based on population genetic theory, molecular models of immunity and previous empirical studies, population genetic depletion predicts increased susceptibility of populations to emergent pathogens. We experimentally compared susceptibility of R. latastei populations upon exposure to an emerging strain of Ranavirus, frog virus 3 (FV3), using six populations spanning the geographical range and range of population genetic diversity found in nature. Our findings confirm this prediction, suggesting that the loss of genetic diversity accompanying range expansion and population isolation is coincident with increased mortality risk from an emergent pathogen. Loss of heterozygosity and escape from selection imposed by immunologically cross-reactive pathogens may potentially generate range-wide variation in disease resistance
