Phospholipids and the beta-andrenergic response

The phospholipid polar headgroup composition of membranes of C6 cells was modified in vivo by growth for 24h in media supplemented with the polar headgroup precursors, N, N'-dimethylethanolamine, N-monomethylethanolamine or ethanolamine. These modifications were achieved without alteration of the cholesterol and phospholipid content of the membranes, and without changes in the fatty acyl or protein composition of these membranes. No changes were found in the physical properties of membranes isolated from these cells . Enriching the cell membranes with PDME, PNME or PE elevated the basal intracellular cAMP content, but decreased the degree of stimulation of intracellular cAMP content in response to β-adrenergic stimulation. This reduction of the β-adrenergic response appeared to be due to enhanced phosphodiesterase activity. β-adrenergic stimulation did not affect bulk physical properties of the membrane. Exposure to isoproterenol increased the amount of [3H] methyl label recovered from TLC plates but the different distribution of label indicated that such analysis is insufficient to demonstrate specific phospholipid methylation. This system is a useful means of studying the relationship between phospholipid composition, phospholipid metabolism and the β-adrenergic response in the C6 cell line

Multi-residue enantioselective determination of emerging drug contaminants in seawater by solid phase extraction and liquid chromatography-tandem mass spectrometry.

This study proposes a new multi-residue enantioselective method for the determination of emerging drug contaminants in sea water by solid phase extraction (SPE) and liquid chromatography-tandem mass spectrometry (LC-MS/MS). To achieve satisfactory enantiomeric separation with a vancomycin stationary phase it was essential to limit sodium chloride in extracted samples to [less than] 1 µg injection-1. This was achieved through a straightforward SPE method using a 50 mL water wash volume and analyte elution in acetonitrile. A Chiral-V enantioselective column (150 x 2.1mm; 2.7µm particle size) operated in polar ionic mode enabled simultaneous drug separations in 30 minutes. Analytes with enantioresolution E$ were the stimulants amphetamine and methamphetamine, the beta-agonist salbutamol, the beta-blockers propranolol, sotalol and acebutolol, the anti-depressants fluoxetine, venlafaxine, desmethylvenlafaxine and citalopram, and the antihistamine chlorpheniramine. Method quantitation limits were [less than] 10 ng L-1 and method trueness was 80-110% for most analytes. The method was applied to samples from the Forth and Clyde estuaries, Scotland. Chiral drugs were present at concentrations in the range 4-159 ng L-1 and several were in non-racemic form (enantiomeric fraction G 0.50) demonstrating enantiomer enrichment. This emphasises the need for further enantiospecific drug exposure and effect studies in the marine environment

Effect of levamisole on immunity to Corynebacterium pseudotuberculosis in mice and sheep

The stimulating effect of levamisole on immunity to Corynebacterium pseudotuberculosis in mice was marginal, while no enhancement of immunity could be detected in sheep. The conclusion was reached that, as levamisole has no potentiating effect on immunity to C. pseudotuberculosis in normal sheep, it is of no practical value as an immunostimulant in this instance.The articles have been scanned in colour with a HP Scanjet 5590; 300dpi. Comparative efficacy of six Brucella vaccines in guinea-pigs

Antibody response of guinea-pigs and cattle to a Campylobacter fetus oil emulsion vaccine

A method is described for the mass production of Campylobacter fetus oil emulsion vaccine, using 2 strains of C. fetus ss intestinalis and a strain of C. fetus ss intermedius as a substitute for C. fetus ss venerialis.
