    A Novel On-chip Three-dimensional Micromachined Calorimeter with Fully Enclosed and Suspended Thin-film Chamber for Thermal Characterization of Liquid Samples

    A microfabricated calorimeter (μ-calorimeter) with an enclosed reaction chamber is presented. The 3D micromachined reaction chamber is capable of analyzing liquid samples with volume of 200 nl. The thin film low-stress silicon nitride membrane is used to reduce thermal mass of the calorimeter and increase the sensitivity of system. The μ-calorimeter has been designed to perform DC and AC calorimetry, thermal wave analysis, and differential scanning calorimetry. The μ-calorimeter fabricated with an integrated heater and a temperature sensor on opposite sides of the reaction chamber allows to perform thermal diffusivity and specific heat measurements on liquid samples with same device.     R992C (p.R1192C) Substitution in collagen II alters the structure of mutant molecules and induces the unfolded protein response.

    We investigated the molecular bases of spondyloepiphyseal dysplasia (SED) associated with the R992C (p.R1192C) substitution in collagen II. At the protein level, we analyzed the structure and integrity of mutant molecules, and at the cellular level, we specifically studied the effects of the presence of the R992C collagen II on the biological processes taking place in host cells. Our studies demonstrated that mutant collagen II molecules were characterized by altered electrophoretic mobility, relatively low thermostability, the presence of atypical disulfide bonds, and slow rates of secretion into the extracellular space. Analyses of cellular responses to the presence of the mutant molecules showed that excessive accumulation of thermolabile collagen II was associated with the activation of an unfolded protein response and an increase in apoptosis of host cells. Collectively, these data suggest that molecular mechanisms of SED may be driven not only by structural changes in the architecture of extracellular collagenous matrices, but also by intracellular processes activated by the presence of mutant collagen II molecules

    Serum high mobility group box-1 (HMGB1) is closely associated with the clinical and pathologic features of gastric cancer

    <p>Abstract</p> <p>Background</p> <p>High mobility group box-1 (HMGB1) is a newly recognized factor regulating cancer cell tumorigenesis, expansion and invasion. We investigated the correlation between the serum HMGB1 levels and the clinical and pathologic features of gastric cancer and evaluated the validity of HMGB1 as a potential biomarker for the early diagnosis of gastric cancer.</p> <p>Methods</p> <p>A total of 227 subjects were classified into 5 disease groups according to the 'gastritis-dysplasia-carcinoma' sequence of gastric carcinogenesis and their serum levels of HMGB1 were analyzed by an enzyme-linked immunosorbent assay (ELISA) method. Clinical parameters, International Union Against Cancer (UICC) TNM stage, cancer size, differentiation or lymphatic invasion, vascular or perineural invasion and prognosis were used as analysis variables.</p> <p>Results</p> <p>The serum HMGB1 levels were significantly different among disease groups (ANOVA, <it>p < 0.05</it>) and HMGB1 levels tended to increase according to the progression of gastric carcinogenesis. Serum HMGB1 levels were significantly associated with depth of invasion, lymph node metastasis, tumor size, and poor prognosis (<it>p < 0.05</it>). However, HMGB1 levels were not associated with patient gender or age, differentiation of tumor cells, or lymphatic, vascular and perineural invasion, or the existence of distant metastasis in advanced cancer (<it>p > 0.05</it>). The sensitivity and specificity of serum HMGB1 was 71% and 67% (cut-off value of 5 ng/ml) for the diagnosis of early gastric cancer, and 70% and 64% (cut-off value of 4 ng/ml) for the diagnosis of high-risk lesions, respectively. These values were greater than those for carcinoembryonic antigen (CEA) (30–40% of sensitivity).</p> <p>Conclusion</p> <p>HMGB1 appears to be a useful serological biomarker for early diagnosis as well as evaluating the tumorigenesis, stage, and prognosis of gastric cancer.</p

    Vertical ridge augmentation feasibility using unfixed collagen membranes and particulate bone substitutes: A 1- to 7-year retrospective single-cohort observational study

    AIM To determine whether vertical ridge augmentation (VRA) can be obtained through guided bone regeneration (GBR) using exclusively resorbable collagen membranes and particulate bone substitutes without additional stabilization. MATERIALS AND METHODS This study retrospectively examined 22 participants who underwent VRA with staged or simultaneous implant placement. The vertical defects of all participants were filled with particulate bone substitutes and covered with resorbable collagen membranes. The augmented sites were stabilized with unfixed collagen membranes and the flap without any additional fixation. The augmented tissue height was assessed using cone-beam computed tomography at baseline, immediately after surgery, and at annual follow-ups. RESULTS The vertical bone gain of the 22 augmented sites amounted to 6.48 ± 2.19 mm (mean ± SD) immediately after surgery and 5.78 ± 1.72 mm at 1- to 7-year follow-up. Of the 22 augmented sites, 18 exhibited changes of less than 1 mm, while the other 4 showed changes of greater than 1 mm. Histological observation of three representative cases revealed new bone apposition on the remaining material. CONCLUSION The present findings indicate that GBR procedures using exclusively collagen membranes and particulate biomaterials without any additional fixation are feasible options for VRA

    Do basic psychological needs affect student engagement in medical school?

    Purpose The purpose of this study is to verify the effect of basic psychological needs of learners on student engagement in medical school. Methods A total of 91 first-year and second-year medical students participated in this study. Their basic psychological needs were determined. Student engagement scales were utilized to determine their engagement. Correlation and multiple regression analyses were conducted. Results Basic psychological needs showed a total explanatory power of 13% for student engagement (F=5.27, p<0.01). Competence (β=0.295, p<0.01) had statistically significant effect on student engagement. Conclusion Results of the present study verified that student engagement could be determined by learner’s traits. Among psychological traits of learners, student engagement was significantly affected by competence. Thus, medical school should provide various experiences to satisfy competence as a basic psychological need of learners

    Large-scale learning of generalised representations for speaker recognition

    The objective of this work is to develop a speaker recognition model to be used in diverse scenarios. We hypothesise that two components should be adequately configured to build such a model. First, adequate architecture would be required. We explore several recent state-of-the-art models, including ECAPA-TDNN and MFA-Conformer, as well as other baselines. Second, a massive amount of data would be required. We investigate several new training data configurations combining a few existing datasets. The most extensive configuration includes over 87k speakers' 10.22k hours of speech. Four evaluation protocols are adopted to measure how the trained model performs in diverse scenarios. Through experiments, we find that MFA-Conformer with the least inductive bias generalises the best. We also show that training with proposed large data configurations gives better performance. A boost in generalisation is observed, where the average performance on four evaluation protocols improves by more than 20%. In addition, we also demonstrate that these models' performances can improve even further when increasing capacity.Comment: 5pages, 5 tables, submitted to ICASS

    ANTI-INFLAMMATORY AND ANALGESIC ACTIVITIES OF METHANOLIC SEED EXTRACT OF STERCULIA VILLOSA ROXB.

    objective: To scientifically validate the analgesic and anti-inflammatory properties of methanolic seed extract of Sterculia villosa Roxb. (SVME) inanimal models.Methods: Analgesic activity of SVME was evaluated by the hot plate-induced pain model, acetic acid-induced abdominal writhing response, andformalin test in mice at the doses of 250 and 500 mg/kg. A carrageenan-induced paw edema model was also used to evaluate anti-inflammatorypotential of SVME in rats at doses of 250 and 500 mg/kg.Results: Phytochemical analysis revealed the presence of flavonoid, gums and carbohydrates, steroids, alkaloid, reducing sugar, and terpenoids insignificant amounts. The SVME produced significant analgesic activity in the hot-plate test in mice at all the time points measured. Extracts of 250 and500 mg/kg reduced dose-dependent acetic acid-induced writhing by 40.2% (p&lt;0.01) and 59.8% (p&lt;0.001), respectively. Significant inhibitionof formalin-induced pain was also observed, with inhibition of 62.1% (p&lt;0.001) and 66.7% (p&lt;0.001) in the early phase at dosages of 250 and500 mg/kg, respectively, and 64.4% (p&lt;0.01) and 70.3% (p&lt;0.01) in the late stage at these dosages. SVME pretreatments showed significant antiinflammatoryactivityagainstcarrageenan-inducededema at all the time points measured.Conclusion: The results suggest that SVME possesses analgesic and anti-inflammatory activities.     A Case of More Abundant and Dysplastic Adenomas in the Interposed Colon than in the Native Colon

    We report a 60-year-old woman with intramucosal adenocarcinoma arising in the interposed colon, 40 years after the esophageal reconstruction for lye induced esophageal stricture. Although synchronous adenomas were also found in the native colon where the graft was taken, the number of adenomas was greater in the interposed colon and more dysplastic, even progressed to adenocarcinoma, than that of the native colon. The microsatellite instability-testing performed in the intramucosal carcinoma from interposed colon showed absence of microsatellite instability. Changing of location and functional deman]d of colonic segment, and the exposure to different intraluminal contents might have facilitated the adenomacarcinoma transformation in the interposed colon
