    Diluting the inflationary axion fluctuation by a stronger QCD in the early Universe

    We propose a new mechanism to suppress the axion isocurvature perturbation, while producing the right amount of axion dark matter, within the framework of supersymmetric axion models with the axion scale induced by supersymmetry breaking. The mechanism involves an intermediate phase transition to generate the Higgs \mu-parameter, before which the weak scale is comparable to the axion scale and the resulting stronger QCD yields an axion mass heavier than the Hubble scale over a certain period. Combined with that the Hubble-induced axion scale during the primordial inflation is well above the intermediate axion scale at present, the stronger QCD in the early Universe suppresses the axion fluctuation to be small enough even when the inflationary Hubble scale saturates the current upper bound, while generating an axion misalignment angle of order unity.Comment: 5 pages, 1 figure; v2: discussion extended, references added, typos correcte

    Submerzni uzgoj i karakterizacija polisaharida iz gljive Grifola frondosa – primjena u kozmetičkoj industriji

    Grifola frondosa (maitake) is traditionally called \u27the king of mushrooms\u27 and \u27the hen of the woods\u27. Both the fruiting bodies and the mycelium of maitake have been reported to have antitumor and antiviral activities. Recently, submerged culture processes have been developed, with the intention of providing opportunities for increased economic exploitation of maitake. Commonly the aim of these processes is to produce extracellular polysaccharides (EPS), mostly glucans, and to explore their applications, particularly in the cosmetic industry. A wide variety of EPS with different molecular chain length and chemical compositions are produced under different culture conditions. In this article, various biological and physicochemical properties of the EPS of G. frondosa (GF-EPS) are described, with a view to applications in the area of functional cosmeceuticals. The GF-EPS, together with GF mycelial extract (GF-MPS), showed antioxidative activity, stimulation of collagen biosynthetic activity, cell proliferation activity, and inhibitory activity of melanogenesis, without significant cytotoxicity. These diverse functionalities suggest that both GF-EPS and GF-MPS can be promising cosmetic ingredients.Gljivu Grifola frondosa (maitake) u Koreji tradicionalno zovu kraljicom gljiva i šumskim bogatstvom. Dokazano je da plodište i micelij gljiva maitake imaju antitumorski i antivirusni učinak. Ekonomsko iskorištenje te gljive omogućeno je submerznim uzgojem. Uobičajeni je cilj uzgoja proizvodnja ekstracelularnih polisaharida (EPS), uglavnom glukana i istraživanje mogućnosti njihove primjene u kozmetičkoj industriji. Pri raznim uvjetima proizvodnje dobivaju se ekstracelularni polisaharidi različitih duljina lanaca i kemijskoga sastava. U ovom su radu opisana razna biološka i fizikalno-kemijska svojstva ekstracelularnih polisaharida gljive Grifola frondosa (GF-EPS), te primjena u proizvodnji kozmetičkih preparata s povoljnim utjecajem na ljudsko zdravlje. Ekstracelularni polisaharidi i ekstrakt micelija gljive Grifola frondosa (GFMPS) imaju antioksidativni učinak, stimuliraju biosintezu kolagena, bujanje stanica, sprečavaju nastanak melanoma i nemaju jače citotoksično djelovanje, zbog čega su prikladni za primjenu u kozmetičkoj industriji

    Anti-oxidant and anti-inflammatory effects of rice bran and green tea fermentation mixture on lipopolysaccharideinduced RAW 264.7 macrophages

    Purpose: To investigate the anti-inflammatory and anti-oxidant properties of an enzyme bath of Oryza sativa (rice bran) and Camellia sinensis O. Kuntz (green tea) fermented with Bacillus subtilis (OCB). Methods: The anti-oxidant effects of OCB were assessed by 2,2-Diphenyl-1-picrylhydrazyl (DPPH) assay and flow cytometry. The anti-inflammatory effects of OCB were assessed by a nitric oxide (NO) assay. Enzyme-linked immunosorbent assay and real-time polymerase chain reaction were used to quantify expression of pro-inflammatory cytokines in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. The major compounds of OCB were identified using high performance liquid chromatography (HPLC) analysis. Results: OCB had no cytotoxic effect on LPS-stimulated macrophages or peripheral blood mononuclear cells up to 1 mg/mL. OCB displayed anti-oxidant effects comparable to those of ascorbic acid and reduced reactive oxygen species (ROS) levels in target cells. OCB treatment of LPSstimulated mavrophages decreased nitric oxide (NO), NO synthase (iNOS), cyclooxygenase-2 (COX-2), prostaglandin E2 (PGE2) and key pro-inflammatory cytokine expressions, suggesting that OCB acts as an anti-oxidant and anti-inflammatory agent by reducing ROS levels and inhibiting pro-inflammatory mediators. The main effector compounds in OCB were epicatechin gallate, cathechin, synigrin acid, epicathechin, epigallocatechin gallate, rutin, and isoquercetin, which are known anti-oxidants. Conclusion: OCB fermentation product may be used as synergistic adjuvant therapy for inflammatory diseases.     Impact of vancomycin-induced changes in the intestinal microbiota on the pharmacokinetics of simvastatin

    The pharmacokinetic (PK) properties of drugs are affected in several ways by interactions with microbiota. The aim of this study was to investigate the effects of oral vancomycin on the gut microbiota and, consequently, on the PKs of simvastatin. An open-label, single arm, sequential crossover study was conducted in six healthy Korean male subjects. After 6 days on a control diet, simvastatin 40 mg was orally administered to the subjects before and after 1 week of oral vancomycin treatment. Blood samples for PK analysis and fecal samples for metagenomic and metabolomic analyses were collected. After vancomycin treatment, the richness of microbiota considerably decreased, and the composition was altered. In particular, the relative abundance of Bacteroidetes decreased, whereas that of proteobacteria increased. In addition, changes in fecal metabolites, including D-glucuronic acid, were observed. However, systemic exposure of simvastatin was not changed whereas that of hydroxysimvastatin showed a tendency to increase. The relationship between the change of PKs of simvastatin and the change of gut microbiota and fecal metabolites were not clearly observed

    An efficient strategy for cell-based antibody library selection using an integrated vector system

    BACKGROUND: Cell panning of phage-displayed antibody library is a powerful tool for the development of therapeutic and imaging agents since disease-related cell surface proteins in native complex conformation can be directly targeted. Here, we employed a strategy taking advantage of an integrated vector system which allows rapid conversion of scFv-displaying phage into scFv-Fc format for efficient cell-based scFv library selection on a tetraspanin protein, CD9. RESULTS: A mouse scFv library constructed by using a phagemid vector, pDR-D1 was subjected to cell panning against stable CD9 transfectant, and the scFv repertoire from the enriched phage pool was directly transferred to a mammalian cassette vector, pDR-OriP-Fc1. The resulting constructs enabled transient expression of enough amounts of scFv-Fcs in HEK293E cells, and flow cytometric screening of binders for CD9 transfectant could be performed simply by using the culture supernatants. All three clones selected from the screening showed correct CD9-specificity. They could immunoprecipitate CD9 molecules out of the transfectant cell lysate and correctly stain endogenous CD9 expression on cancer cell membrane. Furthermore, competition assay with a known anti-CD9 monoclonal antibody (mAb) suggested that the binding epitopes of some of them overlap with that of the mAb which resides within the large extracellular loop of CD9. CONCLUSIONS: This study demonstrates that scFv-Fc from mammalian transient expression can be chosen as a reliable format for rapid screening and validation in cell-based scFv library selection, and the strategy described here will be applicable to efficient discovery of antibodies to diverse cell-surface targets

    Dispersion of Vascular Plant in Kumo-do, Korea

    AbstractThe vascular plants observed in the area were composed of a total of 228 taxa; 72 families, 172 genus, 201 species, 25 varieties, 1 sub-species and 1 cross species. The only endangered plants found in the area were Milletia japonica (Siebold & Zucc.) A.Gray. The endemic plants growing in the Geumodo except transplanted plants were Lespedeza x maritima Nakai and Carpinus coreana Nakai. which accounted for 0.8% of the vascular plants in Geumodo, 228 taxa. Specialized plants of Geumodo were a total of 41 species; 30 taxa in Grade I, 1 taxon in Grade II, 9 taxa in Grade III and 1 taxon in Grade V. Milletia japonica (Siebold & Zucc.) A.Gray was the only species found in important Grade IV to V. Currently, ferries ply to the island, attracting many tourists. This poses a threat to the rare plants living in the island and presses down the island to develop. Therefore, in the long-term perspective, the conservation plan such as comprehensive research and monitoring on the ecosystem shall be established to protect evergreen broad-leaved forests

    Satellite radiance data assimilation for binary tropical cyclone cases over the western North Pacific

    A total of three binary tropical cyclone (TC) cases over the Western North Pacific are selected to investigate the effects of satellite radiance data assimilation on analyses and forecasts of binary TCs. Two parallel cycling experiments with a 6 h interval are performed for each binary TC case, and the difference between the two experiments is whether satellite radiance observations are assimilated. Satellite radiance observations are assimilated using the Weather Research and Forecasting Data Assimilation (WRFDA)'s three-dimensional variational (3D-Var) system, which includes the observation operator, quality control procedures, and bias correction algorithm for radiance observations. On average, radiance assimilation results in slight improvements of environmental fields and track forecasts of binary TC cases, but the detailed effects vary with the case. When there is no direct interaction between binary TCs, radiance assimilation leads to better depictions of environmental fields, and finally it results in improved track forecasts. However, positive effects of radiance assimilation on track forecasts can be reduced when there exists a direct interaction between binary TCs and intensities/structures of binary TCs are not represented well. An initialization method (e.g., dynamic initialization) combined with radiance assimilation and/or more advanced DA techniques (e.g., hybrid method) can be considered to overcome these limitations

    Enhancement of radiation response in human cervical cancer cells in vitro and in vivo by arsenic trioxide (As2O3)

    AbstractArsenic trioxide (As2O3) inhibits cell growth and induces apoptosis in certain types of cancer cells including acute promyelocytic leukemia, prostate and ovarian carcinomas, but its effect on response of tumor cells to ionizing radiation has never been explored before. Here we demonstrate that As2O3 can sensitize human cervical cancer cells to ionizing radiation both in vitro and in vivo. As2O3 in combination with ionizing radiation have a synergistic effect in decreasing clonogenic survival and in the regression of established human cervical tumor xenografts. Pretreatment of the cells with As2O3 also synergistically enhanced radiation-induced apoptosis. Apoptosis of the cells by combined treatment of As2O3 and radiation was associated with reactive oxygen species generation and loss of mitochondrial membrane potential, resulting in the activation of caspase-9 and caspase-3. The combined treatment also resulted in an increased G2/M cell cycle distribution at the concentration of As2O3 which did not alter cell cycle when applied alone. These results indicate that As2O3 can synergistically enhance radiosensitivity of human cervix carcinoma cells in vitro and in vivo, suggesting a potential clinical applicability of combination treatment of As2O3 and ionizing radiation in cancer therapies
