    Mist interception by three species of mountain fynbos

    Bibliography: leaves 92-96.The objective of the present study was to investigate mist interception by Mountain Fynbos under non-rainfall conditions. Quantitative measurements and qualitative observations of plant-drip and stem-flow from selected plant species were recorded at five experimental sites on the Back Table of Table Mountain, Cape Province. Plant-drip and stem-flow catching devices were designed and installed in order to collect the moisture intercepted by plants. The results indicated that mist interception by the vegetation was influenced by wind speed, density of mist and by the morphological characteristics of the plants. Greater interception occurred when dense mist was accompanied by high wind speeds. The interception ability was found to increase with plant size. Interception efficiency depended on the leaf morphology of the plants. Narrow-sclerophylls (e.g. Psoralea pinnata) were found to be more effective in intercepting mist droplets than broad-clerophylls (e.g. Leucadendron salignum). The portioning of intercepted water into plant-drip and stem-flow was largely influenced by the shape of the plant. The mean rate of water yield per plant observed during five periods of rainfree, misty conditions, was 71 ml hr-1 . Rainfall is, therefore, not the sole source of moisture contributing to the water balance. The additional moisture intercepted by vegetation should, therefore, be taken into consideration in studies of water balance and hydrology

    Evolutionary History of a Specialized P450 Propane Monooxygenase

    The evolutionary pressures that shaped the specificity and catalytic efficiency of enzymes can only be speculated. While directed evolution experiments show that new functions can be acquired under positive selection with few mutations, the role of negative selection in eliminating undesired activities and achieving high specificity remains unclear. Here we examine intermediates along the ‘lineage’ from a naturally occurring C12–C20 fatty acid hydroxylase (P450BM3) to a laboratory-evolved P450 propane monooxygenase (P450PMO) having 20 heme domain substitutions compared to P450BM3. Biochemical, crystallographic, and computational analyses show that a minimal perturbation of the P450BM3 fold and substrate-binding pocket accompanies a significant broadening of enzyme substrate range and the emergence of propane activity. In contrast, refinement of the enzyme catalytic efficiency for propane oxidation (not, vert, similar 9000-fold increase in kcat/Km) involves profound reshaping and partitioning of the substrate access pathway. Remodeling of the substrate-recognition mechanisms ultimately results in remarkable narrowing of the substrate profile around propane and enables the acquisition of a basal iodomethane dehalogenase activity as yet unknown in natural alkane monooxygenases. A highly destabilizing L188P substitution in a region of the enzyme that undergoes a large conformational change during catalysis plays an important role in adaptation to the gaseous alkane. This work demonstrates that positive selection alone is sufficient to completely respecialize the cytochrome P450 for function on a nonnative substrate

    Establishing the extent of malaria transmission and challenges facing pre-elimination in the Republic of Djibouti.

    BACKGROUND: Countries aiming for malaria elimination require a detailed understanding of the current intensity of malaria transmission within their national borders. National household sample surveys are now being used to define infection prevalence but these are less efficient in areas of exceptionally low endemicity. Here we present the results of a national malaria indicator survey in the Republic of Djibouti, the first in sub-Saharan Africa to combine parasitological and serological markers of malaria, to evaluate the extent of transmission in the country and explore the potential for elimination. METHODS: A national cross-sectional household survey was undertaken from December 2008 to January 2009. A finger prick blood sample was taken from randomly selected participants of all ages to examine for parasitaemia using rapid diagnostic tests (RDTs) and confirmed using Polymerase Chain Reaction (PCR). Blood spots were also collected on filter paper and subsequently used to evaluate the presence of serological markers (combined AMA-1 and MSP-119) of Plasmodium falciparum exposure. Multivariate regression analysis was used to determine the risk factors for P. falciparum infection and/or exposure. The Getis-Ord G-statistic was used to assess spatial heterogeneity of combined infections and serological markers. RESULTS: A total of 7151 individuals were tested using RDTs of which only 42 (0.5%) were positive for P. falciparum infections and confirmed by PCR. Filter paper blood spots were collected for 5605 individuals. Of these 4769 showed concordant optical density results and were retained in subsequent analysis. Overall P. falciparum sero-prevalence was 9.9% (517/4769) for all ages; 6.9% (46/649) in children under the age of five years; and 14.2% (76/510) in the oldest age group (≥50 years). The combined infection and/or antibody prevalence was 10.5% (550/4769) and varied from 8.1% to 14.1% but overall regional differences were not statistically significant (χ2=33.98, p=0.3144). Increasing age (p<0.001) and decreasing household wealth status (p<0.001) were significantly associated with increasing combined P. falciparum infection and/or antibody prevalence. Significant P. falciparum hot spots were observed in Dikhil region. CONCLUSION: Malaria transmission in the Republic of Djibouti is very low across all regions with evidence of micro-epidemiological heterogeneity and limited recent transmission. It would seem that the Republic of Djibouti has a biologically feasible set of pre-conditions for elimination, however, the operational feasibility and the potential risks to elimination posed by P. vivax and human population movement across the sub-region remain to be properly established

    A Search For Organic Molecules in Intermediate Redshift DLAs

    There has been a renewed interest in searching for diffuse interstellar bands (DIBs) due to their probable connection to organic molecules and, thus, their possible link to life in the Universe. Our group is undertaking an extensive search for DIBs in DLAs via QSO absorption-line systems. Six of our DLA targets are presented here. Our equivalent width (EW) limits for the 5780 DIB line strongly suggests that DIB abundance is below the Milky Way expected value or that metallicity plays a large role in DIB strengths.Comment: 3 pages; 2 figures; presented at IAU 199; Probing Galaxies through Quasar Absorption Line
