    Mermithid Nematodes: SEM Observations Comparing Hexamethyldisilazane and Critical Point Drying Methods

    Morphological features of mermithid nematodes (Mermithidae) were studied with scanning electron microscopy, using hexamethyldisilazane-air drying in comparison with critical point drying via liquid carbon dioxide. Although general morphologic preservation of both HMDS-dried and CP-dried specimens was similar, structural features of the complex cuticle and internal organization were more easily resolved at higher magnifications in the HMDS-dried nematodes. These features include the superficial cuticular annulations, the fibrillar inner cuticle and peg-like microtrabeculae. The previously undescribed microtrabeculae are of special interest since they may facilitate an interaction of the mermithid (and perhaps nameatodes in general) musculature with its body wall that, at least in part, may account for the unique thrashing locomotion so characteristic of these organisms

    While mindfulness has demonstrated many advantages in the workplace, this paper addresses important issues around the future of mindfulness at work. We begin by clarifying our understanding of mindfulness in the workplace. This is followed by a discussion on the problematic nature of mindfulness-based interventions in workplaces and potential guidance is provided for those who intend to undertake interventions. Finally, we examine how workplaces are naturalistic settings that differ in how they can nurture mindfulness in employees. Ultimately this paper provides organizations and practitioners insight into potential issues in navigating mindfulness at work, while also providing cautionary optimism around the future of mindfulness in the workplace

    Comparing Model Representations of Physiological Limits on Transpiration at a Semi-arid Ponderosa Pine Site

    Mechanistic representations of biogeochemical processes in ecosystem models are rapidly advancing, requiring advancements in model evaluation approaches. Here we quantify multiple aspects of model functional performance to evaluate improved process representations in ecosystem models. We compare semi-empirical stomatal models with hydraulic constraints against more mechanistic representations of stomatal and hydraulic functioning at a semi-arid pine site using a suite of metrics and analytical tools. We find that models generally perform similarly under unstressed conditions, but performance diverges under atmospheric and soil drought. The more empirical models better capture synergistic information flows between soil water potential and vapor pressure deficit to transpiration, while the more mechanistic models are overly deterministic. Although models can be parameterized to yield similar functional performance, alternate parameterizations could not overcome structural model constraints that underestimate the unique information contained in soil water potential about transpiration. Additionally, both multilayer canopy and big-leaf models were unable to capture the magnitude of canopy temperature divergence from air temperature, and we demonstrate that errors in leaf temperature can propagate to considerable error in simulated transpiration. This study demonstrates the value of merging underutilized observational data streams with emerging analytical tools to characterize ecosystem function and discriminate among model process representations

    A case study in adaptable and reusable infrastructure at the Keck Observatory Archive: VO interfaces, moving targets, and more

    The Keck Observatory Archive (KOA) (https://koa.ipac.caltech.edu) curates all observations acquired at the W. M. Keck Observatory (WMKO) since it began operations in 1994, including data from eight active instruments and two decommissioned instruments. The archive is a collaboration between WMKO and the NASA Exoplanet Science Institute (NExScI). Since its inception in 2004, the science information system used at KOA has adopted an architectural approach that emphasizes software re-use and adaptability. This paper describes how KOA is currently leveraging and extending open source software components to develop new services and to support delivery of a complete set of instrument metadata, which will enable more sophisticated and extensive queries than currently possible. In August 2015, KOA deployed a program interface to discover public data from all instruments equipped with an imaging mode. The interface complies with version 2 of the Simple Imaging Access Protocol (SIAP), under development by the International Virtual Observatory Alliance (IVOA), which defines a standard mechanism for discovering images through spatial queries. The heart of the KOA service is an R-tree-based, database-indexing mechanism prototyped by the Virtual Astronomical Observatory (VAO) and further developed by the Montage Image Mosaic project, designed to provide fast access to large imaging data sets as a first step in creating wide-area image mosaics (such as mosaics of subsets of the 4.7 million images of the SDSS DR9 release). The KOA service uses the results of the spatial R-tree search to create an SQLite data database for further relational filtering. The service uses a JSON configuration file to describe the association between instrument parameters and the service query parameters, and to make it applicable beyond the Keck instruments. The images generated at the Keck telescope usually do not encode the image footprints as WCS fields in the FITS file headers. Because SIAP searches are spatial, much of the effort in developing the program interface involved processing the instrument and telescope parameters to understand how accurately we can derive the WCS information for each instrument. This knowledge is now being fed back into the KOA databases as part of a program to include complete metadata information for all imaging observations. The R-tree program was itself extended to support temporal (in addition to spatial) indexing, in response to requests from the planetary science community for a search engine to discover observations of Solar System objects. With this 3D-indexing scheme, the service performs very fast time and spatial matches between the target ephemerides, obtained from the JPL SPICE service. Our experiments indicate these matches can be more than 100 times faster than when separating temporal and spatial searches. Images of the tracks of the moving targets, overlaid with the image footprints, are computed with a new command-line visualization tool, mViewer, released with the Montage distribution. The service is currently in test and will be released in late summer 2016

    Minimally Invasive Posterior Facet Decortication and Fusion Using Navigated Robotic Guidance: Feasibility and Workflow Optimization

    Minimally invasive spine surgery reduces tissue dissection and retraction, decreasing the morbidity associated with traditional open spine surgery by decreasing blood loss, blood transfusion, complications, and pain. One of the key challenges with a minimally invasive approach is achieving consistent posterior fusion. Although advantageous in all fusion surgeries, solid posterior fusion is particularly important in spinal deformity, revisions, and fusions without anterior column support. A minimally invasive surgical approach accomplished without sacrificing the quality of the posterior fusion has the potential to decrease both short- and long-term complications compared to the traditional open techniques. Innovations in navigated and robotic-assisted spine surgery continue to address this need.     Striking the right balance in right whale conservation

There is a crucial gap in our understanding of their habitat use in the migratory corridor along the eastern seaboard of the United States. Here, we characterize habitat suitability in migrating right whales in relation to depth, distance to shore, and the recently enacted ship speed regulations near major ports. We find that the range of suitable habitat exceeds previous estimates and that, as compared with the enacted 20 nautical mile buffer, the originally proposed 30 nautical mile buffer would protect more habitat for this critically endangered species.This work was supported in part by SERDP/DoD grant W912HQ-04-C-0011 to A.J.     Discovery and Validation of a New Class of Small Molecule Toll-Like Receptor 4 (TLR4) Inhibitors

    Many inflammatory diseases may be linked to pathologically elevated signaling via the receptor for lipopolysaccharide (LPS), toll-like receptor 4 (TLR4). There has thus been great interest in the discovery of TLR4 inhibitors as potential anti-inflammatory agents. Recently, the structure of TLR4 bound to the inhibitor E5564 was solved, raising the possibility that novel TLR4 inhibitors that target the E5564-binding domain could be designed. We utilized a similarity search algorithm in conjunction with a limited screening approach of small molecule libraries to identify compounds that bind to the E5564 site and inhibit TLR4. Our lead compound, C34, is a 2-acetamidopyranoside (MW 389) with the formula C17H27NO9, which inhibited TLR4 in enterocytes and macrophages in vitro, and reduced systemic inflammation in mouse models of endotoxemia and necrotizing enterocolitis. Molecular docking of C34 to the hydrophobic internal pocket of the TLR4 co-receptor MD-2 demonstrated a tight fit, embedding the pyran ring deep inside the pocket. Strikingly, C34 inhibited LPS signaling ex-vivo in human ileum that was resected from infants with necrotizing enterocolitis. These findings identify C34 and the β-anomeric cyclohexyl analog C35 as novel leads for small molecule TLR4 inhibitors that have potential therapeutic benefit for TLR4-mediated inflammatory diseases. © 2013 Neal et al

    Netrin-1 Peptide Is a Chemorepellent in \u3cem\u3eTetrahymena thermophila\u3c/em\u3e

    Netrin-1 is a highly conserved, pleiotropic signaling molecule that can serve as a neuronal chemorepellent during vertebrate development. In vertebrates, chemorepellent signaling is mediated through the tyrosine kinase, src-1, and the tyrosine phosphatase, shp-2. Tetrahymena thermophila has been used as a model system for chemorepellent signaling because its avoidance response is easily characterized under a light microscope. Our experiments showed that netrin-1 peptide is a chemorepellent in T. thermophila at micromolar concentrations. T. thermophila adapts to netrin-1 over a time course of about 10 minutes. Netrin-adapted cells still avoid GTP, PACAP-38, and nociceptin, suggesting that netrin does not use the same signaling machinery as any of these other repellents. Avoidance of netrin-1 peptide was effectively eliminated by the addition of the tyrosine kinase inhibitor, genistein, to the assay buffer; however, immunostaining using an anti-phosphotyrosine antibody showed similar fluorescence levels in control and netrin-1 exposed cells, suggesting that tyrosine phosphorylation i s not required for signaling to occur. In addition, ELISA indicates that a netrin-like peptide is present in both whole cell extract and secreted protein obtained from Tetrahymena thermophila. Further study will be required in order to fully elucidate the signaling mechanism of netrin-1 peptide in this organism
