    Increasing Biogas Yield from Fodder by Microbial Stimulation of Propionic Acid Synthesis in Grass Silages

    A new direction in the use of lactic acid bacteria inoculants is their application for renewable raw materials ensiling for biogas production. The aim of the study was to demonstrate the possibility of stimulating the synthesis of propionic acid in the process of co-fermentation of selected strains of Lactobacillus buchneri and L. diolivorans as well as L. buchneri and Pediococcus acidilactici. L. buchneri KKP 2047p and P. acidilactici KKP 2065p were characterized by the special capabilities for both synthesis and metabolism of 1,2-propanediol. L. diolivorans KKP 2057p stands out for the ability to metabolize 1,2-propanediol to propionic acid. As a result of the co-fermentation a concentration of propionic acid was obtained at least 1.5 times higher in the final stage of culture in comparison to cultivating individual species of bacteria separately. The results of in vitro experiments were applied in agricultural practice, by application of two lactic acid bacteria inoculants in ensiling of grass silage and improving its suitability for biogas production. Grass silages made with the addition of the inoculant were characterized by the content of 1,2-propanediol, 1-propanol and propionic acid ensured extension of the aerobic stability from 4 to 7 days in comparison to untreated silages. It was found that the use of both inoculants resulted in an approximately 10–30% increase in biogas yield from this raw material

    Natural Cinnamic Acid Derivatives: A Comprehensive Study on Structural, Anti/Pro-Oxidant, and Environmental Impacts

    Cinnamic acid (CA), p-coumaric acid (4-hydroxycinnamic acid, 4-HCA), caffeic acid (3,4-vdihydroxycinnamic acid, 3,4-dHCA), and 3,4,5-trihydroxycinnamic acid (3,4,5-tHCA) were studied for their structural, anti-/pro-oxidant properties and biodegradability. The FT-IR, FT-Raman, UV/Vis, 1H and 13C NMR, and quantum chemical calculations in B3LYP/6-311++G** were performed to study the effect on number and position of hydroxyl group in the ring on the molecular structure of molecules. The antioxidant properties of the derivatives were examined using DPPH● and HO● radicals scavenging assays, ferric ion reducing antioxidant power (FRAP), cupric reducing antioxidant capacity (CUPRAC), inhibition of linoleic acid oxidation, as well as the biological antioxidant assay with Saccharomyces cerevisiae. Moreover, the pro-oxidant activity of compounds in Trolox oxidation assay was estimated. The effect of the derivatives on environment on the basis of increasing the carbon and nitrogen compounds transformation processes occurring in biological wastewater treatment was studied

    Enzymatic Synthesis of Poly(alkylene succinate)s: Influence of Reaction Conditions

    Application of lipases (preferentially Candida antarctica Lipase B, CALB) for melt polycondensation of aliphatic polyesters by transesterification of activated dicarboxylic acids with diols allows to displace toxic metal and metal oxide catalysts. Immobilization of the enzyme enhances the activity and the temperature range of use. The possibility to use enzyme-catalyzed polycondensation in melt is studied and compared to results of polycondensations in solution. The experiments show that CALB successfully catalyzes polycondensation of both, divinyladipate and dimethylsuccinate, respectively, with 1,4-butanediol. NMR spectroscopy, relative molar masses obtained by size exclusion chromatography, MALDI-TOF MS and wide-angle X-ray scattering are employed to compare the influence of synthesis conditions for poly(butylene adipate) (PBA) and poly(butylene succinate) (PBS). It is shown that the enzymatic activity of immobilized CALB deviates and influences the molar mass. CALB-catalyzed polycondensation of PBA in solution for 24 h at 70 °C achieves molar masses of up to Mw~60,000 g/mol, higher than reported previously and comparable to conventional PBA, while melt polycondensation resulted in a moderate decrease of molar mass to Mw~31,000. Enzymatically catalyzed melt polycondensation of PBS yields Mw~23,400 g/mol vs. Mw~40,000 g/mol with titanium(IV)n-butoxide. Melt polycondensation with enzyme catalysis allows to reduce the reaction time from days to 3–4 h

    New Isolated Autochthonous Strains of S. cerevisiae for Fermentation of Two Grape Varieties Grown in Poland

    Many commercial strains of the Saccharomyces cerevisiae species are used around the world in the wine industry, while the use of native yeast strains is highly recommended for their role in shaping specific, terroir-associated wine characteristics. In recent years, in Poland, an increase in the number of registered vineyards has been observed, and Polish wines are becoming more recognizable among consumers. In the fermentation process, apart from ethyl alcohol, numerous microbial metabolites are formed. These compounds shape the wine bouquet or become precursors for the creation of new products that affect the sensory characteristics and quality of the wine. The aim of this work was to study the effect of the grapevine varieties and newly isolated native S. cerevisiae yeast strains on the content of selected wine fermentation metabolites. Two vine varieties—Regent and Seyval blanc were used. A total of 16 different yeast strains of the S. cerevisiae species were used for fermentation: nine newly isolated from vine fruit and seven commercial cultures. The obtained wines differed in terms of the content of analyzed oenological characteristics and the differences depended both on the raw material (vine variety) as well as the source of isolation and origin of the yeast strain used (commercial vs. native). Generally, red wines characterized a higher content of tested analytes than white wines, regardless of the yeast strain used.
