The advent of numerous tools, ease of techniques, and concepts related to nanotechnology, in combination with functionalization via simple chemistry has made gold important for various biomedical applications. In this dissertation, the development and characterization of planar gold surfaces with responsive hydrogel patterns for rapid point of care sensing and the functionalization of gold nanoparticles for drug delivery are highlighted. Biomedical micro- and nanoscale devices that are spatially functionalized with intelligent hydrogels are typically fabricated using conventional UV-lithography. Herein, precise 3-D hydrogel patterns made up of temperature responsive crosslinked poly(N-isopropylacrylamide) over gold were synthesized. The XY control of the hydrogel was achieved using microcontact printing, while thickness control was achieved using atom transfer radical polymerization (ATRP). Atomic force microscopy analysis showed that to the ATRP reaction time governed the pattern growth. The temperature dependent swelling ratio was tailored by tuning the mesh size of the hydrogel. While nanopatterns exhibited a broad lower critical solution temperature (LCST) transition, surface roughness showed a sharp LCST transition. Quartz crystal microbalance with dissipation showed rapid response behavior of the thin films, which makes them applicable as functional components in biomedical devices. The easy synthesis, relative biocompatibility, inertness, and easy functionalization of gold nanoparticles (GNPs) have made them useful for various biomedical applications. Although ATRP can be successfully carried out over GNPs, the yield of stable solution based GNPs for biomedical applications prove to be low. As an alternative approach, a novel method of ISOlating, FUnctionalizing, and REleasing nanoparticles (ISOFURE) was proposed. Biodegradable poly(β-amino ester) hydrogels were used to synthesize ISOFURE-GNP composites. ATRP was performed inside the composite, and the final hydrogel coated GNPs were released via matrix degradation. Response analysis confirmed that the ISOFURE method led to the increased stability and yield of the hydrogel coated ISOFURE-GNPs. The ISOFURE protocol was also utilized in functionalizing GNPs with enzyme catalase in the absence of a stabilizing reagent. Biotin-streptavidin affinity was used as the bioconjugation method. Activity analysis of the conjugated enzyme showed that the ISOFURE-GNPs showed enhanced biomolecular loading relative to solution based stabilizing reagent passivated GNPs

A Recovery Scheme for Cluster Federations Using Sender-based Message Logging

    Get PDF
A cluster federation is a union of clusters and is heterogeneous. Each cluster contains a certain number of processes. An application running in such a computing environment is divided into communicating modules so that these modules can run on different clusters. To achieve fault-tolerance different clusters may employ different check pointing schemes. For example, some may use coordinated schemes, while some other may use communication-induced schemes. It may complicate the recovery process. In this paper, we have addressed the complex problem of recovery for cluster computing environment. The proposed approach handles both inter cluster orphan and lost messages unlike the existing works in this area. We first propose an algorithm to determine a recovery line so that there does not exist any inter cluster orphan message between any pair of the cluster level check points belonging to the recovery line. The main feature of the proposed algorithm is that it can be executed simultaneously by all clusters in the cluster federation. Next we apply the sender-based message logging idea to effectively handle all inter cluster lost messages to ensure correctness of computation

A Survey on Software Cost Estimation Techniques

    Get PDF
The ability to accurately estimate the cost needed to complete a specific project has been a challenge over the past decades. For a successful software project, accurate prediction of the cost, time and effort is a very much essential task. This paper presents a systematic review of different models used for software cost estimation which includes algorithmic methods, non-algorithmic methods and learning-oriented methods. The models considered in this review include both the traditional and the recent approaches for software cost estimation. The main objective of this paper is to provide an overview of software cost estimation models and summarize their strengths, weakness, accuracy, amount of data needed, and validation techniques used. Our findings show, in general, neural network based models outperforms other cost estimation techniques. However, no one technique fits every problem and we recommend practitioners to search for the model that best fit their needs

Giovanni II d'Aragona e la partecipazione del Regno di Sardegna e Corsica nella guerra civile catalana

This dissertation partially makes up for the lack of Sardinian, Italian and Iberian historiography, with particular reference to the role of John II of Aragon in the Kingdom of Sardinia and Corsica (1458-79). Given the considerable amount of documents available on the topic, mostly unpublished and stored in archives, both in Italy and abroad, it has been necessary to perform a scientific analysis of unpublished data; these data have then been integrated with those retrieved from published documents, along with data gathered from other literary sources and chronicles. As a result, our work has been subdivided into four sections, featuring a huge collection of documents. After an introduction on the scientific methodology applied to the research process, we have moved on to examine the kingdom of John II after his accession to the throne of Aragon and the reasons for the break out of the ten-year Catalan Civil War, as well as its repercussions on the crisis on the kingdom of Sardinia and Corsica. The period after the Civil War has been taken into consideration and, and particular attention has been paid to the end of the marquisate of Oristano, which has marked the history of the Kingdom of Sardinia and Corsica in the 1470’s. Finally, in the appendix, 111 documents are to be found, with the summaries and the transcription of the most important texts used during the research, borrowed from both the Archive of the Crown of Aragon in Barcelona and the State Archive of Cagliari

Using nanoscale bioreactors to characterize sub-populations of CHO clones and screen transfected pools

    Get PDF
Traditional means to quantify growth and production rates for antibody-expressing CHO lines involve sampling aliquots and supernatants from well plates that have been seeded with single cells. The number of clones studied is often limited by cloning efficiencies (typically 5-50%) and the inability to handle large numbers of well plates. The speed at which each clone can be measured is limited by the growth rates of cells and the number of cells required to perform each assay. Both of these factors lead to a practical throughput of 100s of clones screened over the course of 2-4 weeks. Furthermore, each readout of a clone offers little to no insight into the behavior of sub-populations within each clone since the aliquot or supernatant is just a small sample representing the entire population.
