Three-Dimensional Limit Analysis of the Vicoforte Elliptical Dome
Limit Analysis provides a conceptually simple and robust method to estimate the safety of structures and has been long applied to the analysis of the ultimate collapse state of two-dimensional masonry structures or structural elements. In revolving symmetric domes, the three-dimensional problem can be reduced to the two-dimensional case under appropriate hypotheses. The Vicoforte dome is the largest elliptical dome in the world, and its complex geometry makes this kind of analysis not straightforward. Starting from some basic assumptions, a method for analyzing the three-dimensional elliptical geometry and understand the behavior at collapse of the drum-dome system using limit analysis is proposed. The three dimensional collapse mechanism is found and the system behavior including the presence of tension rings at different levels is interpreted. The results are compared against a nonlinear finite element model
Identification of novel vascular markers through gene expression profiling of tumor-derived endothelium
<p>Abstract</p> <p>Background</p> <p>Targeting tumor angiogenesis and vasculature is a promising strategy for the inhibition of tumor growth and dissemination. Evidence suggests that tumor vasculature expresses unique markers that distinguish it from normal vasculature. Our efforts focused on the molecular characterization of endothelial cells (EC) in the search for selective markers of tumor vasculature that might be helpful for the development of effective therapeutic approaches.</p> <p>Results</p> <p>We investigated by microarray analysis the gene expression profiles of EC purified and cultured from tumor (ovarian carcinoma [HOC-EC]) and normal (human adrenal gland [HA-EC]) tissue specimens. We found distinct transcriptional features characterizing the EC of different origin, and identified 158 transcripts highly expressed by HOC-EC. We analyzed four of these genes, ADAM23, FAP, GPNMB and PRSS3, which were not previously known to be expressed by endothelium. <it>In vitro </it>experiments confirmed the higher expression of the selected genes in tumor-derived endothelium with no expression in tumor cells. <it>In vivo </it>investigation by <it>in situ </it>hybridization established that ADAM23, GPNMB and PRSS3 expression is localized on blood vessels of human cancer specimens.</p> <p>Conclusion</p> <p>These findings elucidate some of the molecular features of the tumor endothelium. Comparative transcriptomic analysis allowed us to determine molecular differences of tumor and normal tissue-derived endothelium and to identify novel markers that might be exploited to selectively target tumor vasculature.</p
Gene and microRNA modulation upon trabectedin treatment in a human intrahepatic cholangiocarcinoma paired patient derived xenograft and cell line
Establishment of a patient-derived intrahepatic cholangiocarcinoma xenograft model with KRAS mutation.
BACKGROUND: Intrahepatic cholangiocarcinoma (ICC) is an aggressive, highly lethal tumors and lacks of effective chemo and targeted therapies. Cell lines and animal models, even partially reflecting tumor characteristics, have limits to study ICC biology and drug response. In this work, we created and characterized a novel ICC patient-derived xenograft (PDX) model of Italian origin. METHODS: Seventeen primary ICC tumors derived from Italian patients were implanted into NOD (Non-Obese Diabetic)/Shi-SCID (severe combined immunodeficient) mice. To verify if the original tumor characteristics were maintained in PDX, immunohistochemical (cytokeratin 7, 17, 19, and epithelial membrane antigen) molecular (gene and microRNA expression profiling) and genetic analyses (comparative genomic hybridization array, and mutational analysis of the kinase domain of EGFR coding sequence, from exons 18 to 21, exons 2 to 4 of K-RAS, exons 2 to 4 of N-RAS, exons 9 and 20 of PI3KCA, and exon 15 of B-RAF) were performed after tumor stabilization. RESULTS: One out of 17 (5.8 %) tumors successfully engrafted in mice. A high molecular and genetic concordance between primary tumor (PR) and PDX was confirmed by the evaluation of biliary epithelial markers, tissue architecture, genetic aberrations (including K-RAS G12D mutation), and transcriptomic and microRNA profiles. CONCLUSIONS: For the first time, we established a new ICC PDX model which reflects the histology and genetic characteristics of the primary tumor; this model could represent a valuable tool to understand the tumor biology and the progression of ICC as well as to develop novel therapies for ICC patients.
