    HBX-Mediated Migration of HBV-Replicating HepG2 Cells: Insights on Development of Hepatocellular Carcinoma

    Hepatitus B virus (HBV) is a major cause of the development of hepatpcellular carcinoma (HCC). One of the significant characteristics of tumor progression is cell migration which is reflective of cytoskeletal dynamics. The Rho GTPases contribute to a multiple cellular processes, including the cellular cytoskeletal reorganization and motility. It has been found that some Rho GTPases have oncogenic activity and can promote cancer cell invasion. Here we discuss one of the Rho GTPases, Rac1 can be activated by HBV replication and such activation results in the high motility of HBV-replicating cells. The enhanced cell motility can be interestingly alleviated by the mutation at the sites of proline rich domain located in HBX. Our findings may provide new insights on the mechanism of HCC development associated with chronic HBV infection

    Development and clinical application of a new testicular prosthesis

    A new type of testicular prosthesis made of silastic with an elliptical shape to mimic a normal testis was developed by our team and submitted for patenting in China. The prosthesis was produced in different sizes to imitate the normal testis of the patient. To investigate the effects and safety of the testicular prosthesis, 20 patients receiving testicular prosthesis implantation were recruited for this study. Follow-up after 6 months revealed no complications in the patients. All the patients answered that they were satisfied with their body image and the position of the implants, 19 patients were satisfied with the size and 16 patients were satisfied with the weight. These results show that the testicular prosthesis used in this study can meet patient's expectations. Patients undergoing orchiectomy should be offered the option to receive a testicular prosthesis implantation. The dimensions and weight of the available prosthetic implants should be further addressed to improve patient satisfaction

    Recurrent lung adenocarcinoma benefits from microwave ablation following multidisciplinary treatments: A case with long-term survival

    Lung cancer has become the leading cause of cancer death all over the world. Nowadays, there is a consensus that the treatment of non-small cell lung cancer (NSCLC) prefers a combination of multidisciplinary comprehensive treatment and individualized treatment, which can significantly improve the prognosis of patients. Here, we report a female patient with recurrence-prone NSCLC. She had a decade-long disease course, during which the lesion recurred twice and finally cured with Multi-Disciplinary Treatment (MDT). An elderly female patient was admitted to the hospital after diagnosis of lung cancer, and treated with surgery and postoperative adjuvant chemotherapy. Five years later, suspicious lesions were found by computed tomography (CT) reexamination, and then confirmed tumor recurrence by puncture biopsy. Based on the genetic test results, gefitinib was used for subsequent targeted therapy, and the lesion gradually shrunk to disappear. However, the lesion appeared again two years later, after consultation the microwave ablation was adopted and the curative effect was excellent. At last, regular reexamination showed no abnormality, the patient has survived so far. The case proves the great benefit of multidisciplinary comprehensive treatment, especially microwave ablation for patient with recurrence-prone NSCLC. And the effect of systemic anti-tumor immune response induced by microwave ablation on lung cancer also needs to be further explored

    How far is brain-inspired artificial intelligence away from brain?

    Fueled by the development of neuroscience and artificial intelligence (AI), recent advances in the brain-inspired AI have manifested a tipping-point in the collaboration of the two fields. AI began with the inspiration of neuroscience, but has evolved to achieve a remarkable performance with little dependence upon neuroscience. However, in a recent collaboration, research into neurobiological explainability of AI models found that these highly accurate models may resemble the neurobiological representation of the same computational processes in the brain, although these models have been developed in the absence of such neuroscientific references. In this perspective, we review the cooperation and separation between neuroscience and AI, and emphasize on the current advance, that is, a new cooperation, the neurobiological explainability of AI. Under the intertwined development of the two fields, we propose a practical framework to evaluate the brain-likeness of AI models, paving the way for their further improvements

    The effects of psychiatric disorders on the risk of chronic heart failure: a univariable and multivariable Mendelian randomization study

    BackgroundSubstantial evidence suggests an association between psychiatric disorders and chronic heart failure. However, further investigation is needed to confirm the causal relationship between these psychiatric disorders and chronic heart failure. To address this, we evaluated the potential effects of five psychiatric disorders on chronic heart failure using two-sample Mendelian Randomization (MR).MethodsWe selected single nucleotide polymorphisms (SNPs) associated with chronic heart failure and five psychiatric disorders (Attention-Deficit Hyperactivity Disorder (ADHD), Autism Spectrum Disorder (ASD), Major Depression, Bipolar Disorder and Schizophrenia (SCZ)). Univariable (UVMR) and multivariable two-sample Mendelian Randomization (MVMR) were employed to assess causality between these conditions. Ever smoked and alcohol consumption were controlled for mediating effects in the multivariable MR. The inverse variance weighting (IVW) and Wald ratio estimator methods served as the primary analytical methods for estimating potential causal effects. MR-Egger and weighted median analyses were also conducted to validate the results. Sensitivity analyses included the funnel plot, leave-one-out, and MR-Egger intercept tests. Additionally, potential mediators were investigated through risk factor analyses.ResultsGenetically predicted heart failure was significantly associated with ADHD (odds ratio (OR), 1.12; 95% CI, 1.04–1.20; p = 0.001), ASD (OR, 1.29; 95% CI, 1.07–1.56; p = 0.008), bipolar disorder (OR, 0.89; 95% CI, 0.83–0.96; p = 0.001), major depression (OR, 1.15; 95% CI, 1.03–1.29; p = 0.015), SCZ (OR, 1.04; 95% CI, 1.00–1.07; p = 0.024). Several risk factors for heart failure are implicated in the above cause-and-effect relationship, including ever smoked and alcohol consumption.ConclusionOur study demonstrated ADHD, ASD, SCZ and major depression may have a causal relationship with an increased risk of heart failure. In contrast, bipolar disorder was associated with a reduced risk of heart failure, which could potentially be mediated by ever smoked and alcohol consumption. Therefore, prevention strategies for heart failure should also incorporate mental health considerations, and vice versa

    Effects of forest spatial types, element compositions and forest stands on restorative potential and aesthetic preference

    IntroductionAs global urbanization intensifies, the physical and mental stressors of modern life have led to the growing prevalence of suboptimal health conditions. Spending time in a forest benefits human health and well-being. In this context, based on the forest spatial types (forest interior and forest edge spaces), landscape elements (architecture, water and roads) and forest stands (coniferous, broadleaf and bamboo forests), this study investigated the effects of different forest spatial landscape characteristics on the restorative potential for college students, aesthetic preference and eye movement behavior (total fixation duration and fixation count).MethodsIn this study, a total of 60 subjects were exposed to 42 photographs depicting typical forest landscapes acquired through field studies. The Short-version Revised Restoration and Preference Scale and eye-tracking technology, were employed to study the recovery efficiency and visual attraction of forest spatial of different forest spatial types, element compositions and forest stands.Results(1) The restorative potential and aesthetic preference score of forest edge spaces were significantly higher than those of forest interior spaces. (2) The restorative potential of bamboo forests was significantly higher than those of coniferous and broadleaf forests. (3) In terms of forest interior space, the restorative potential of “forest + 1 element” composition and “forest + 2 elements” composition was significantly higher than that of pure forest, and the restorative potential of interior space of bamboo forest was significantly higher than those of coniferous and broadleaf forests. (4) In terms of forest edge space, the restorative potential of “forest + 2 elements” composition was significantly higher than that of pure forest, and the restorative potential of pure forests was significantly higher than that of the “forest + 1 element” composition. (5) The restorative potential of forest spatial landscape characteristics positively correlated with aesthetic preference and negatively correlated with total fixation duration and fixation count. These results can provide a reference for future forest landscape research, construction and management

    Influences of 1,2-dichlorobenzene on bacterial community structure in wetland soil

    In order to explore the response of the soil microbial community to 1,2-dichlorobenzene exposure in Wetland soil, a short-term (7 weeks) mesocosm study was conducted at four 1,2-dichlorobenzene concentrations of (100, 400 and 800) μg g -1. Dry soil, sterile and unsterile controls were also compared. The results obtained showed a significant effect of 1,2-dichlorobenzene on the development of bacterial populations in soils contaminated with different concentrations of 1,2-dichlorobenzene at the early time after application. In general, however, the number of populations of the same soil sample treated with the same concentration of 1,2-dichlorobenzene differed significantly with the increasing incubation time within the early 5 weeks. The scale of differences in banding patterns-showed that the microbial community structures of 1,2-dichlorobenzene-treated and non-1,2-dichlorobenzene-treated soils were not significantly different after 7 weeks of incubation. DNA in application-responsive bands from the 1,2-dichlorobenzene treatments was recovered and amplified using the universal primers. PCR products were recovered and cloned into pGEM-T Easy (Promega) and two clones were obtained. The two clones were sequenced using the automated Model 3730 DNA sequencing system. The two cloned sequences had very high similarities to an uncultured bacterium reported previously in the database of NCBI

    DLEC1 Expression Is Modulated by Epigenetic Modifications in Hepatocelluar Carcinoma Cells: Role of HBx Genotypes

    Deleted in Lung and Esophageal Cancer 1 (DLEC1) is a functional tumor suppressor gene (TSG). It has been found to be silenced in a variety of human cancers including hepatocellular carcinoma (HCC). The silencing of DLEC1 can be modulated by epigenetic modifications, such as DNA hypermethylation and histone hypoacetylation. In the case of HCC, hepatitis B virus X protein (HBx) has been implicated in methylation of target promoters resulting in the down-regulation of tumor suppressor genes, which in turn contributes to the development of HCC. In the present study, we first established a cell system in which epigenetic modifications can be modulated using inhibitors of either DNA methylation or histone deacetylation. The cell system was used to reveal that the expression of DLEC1 was upregulated by HBx in a genotype-dependent manner. In particular, HBx genotype A was found to decrease DNA methylation of the DLEC1 promoter. Our results have provided new insights on the impact of HBx in HCC development by epigenetic modifications
