    Oscillatory behavior of two nonlinear microbial models of soil carbon decomposition

    A number of nonlinear models have recently been proposed for simulating soil carbon decomposition. Their predictions of soil carbon responses to fresh litter input and warming differ significantly from conventional linear models. Using both stability analysis and numerical simulations, we showed that two of those nonlinear models (a two-pool model and a three-pool model) exhibit damped oscillatory responses to small perturbations. Stability analysis showed the frequency of oscillation is proportional to √(ε⁻¹-1) Ks/Vs in the two-pool model, and to √(ε⁻¹-1) Kl/Vl in the three-pool model, where ε is microbial growth efficiency, Ks and Kl are the half saturation constants of soil and litter carbon, respectively, and /Vs and /Vl are the maximal rates of carbon decomposition per unit of microbial biomass for soil and litter carbon, respectively. For both models, the oscillation has a period of between 5 and 15 years depending on other parameter values, and has smaller amplitude at soil temperatures between 0 and 15°C. In addition, the equilibrium pool sizes of litter or soil carbon are insensitive to carbon inputs in the nonlinear model, but are proportional to carbon input in the conventional linear model. Under warming, the microbial biomass and litter carbon pools simulated by the nonlinear models can increase or decrease, depending whether ε varies with temperature. In contrast, the conventional linear models always simulate a decrease in both microbial and litter carbon pools with warming. Based on the evidence available, we concluded that the oscillatory behavior and insensitivity of soil carbon to carbon input are notable features in these nonlinear models that are somewhat unrealistic. We recommend that a better model for capturing the soil carbon dynamics over decadal to centennial timescales would combine the sensitivity of the conventional models to carbon influx with the flexible response to warming of the nonlinear model.15 page(s

    Precipitation chemistry of Lhasa and other remote towns, Tibet

    Precipitation event samples during 1987-1988 field expedition periods and 1997, 1998, 1999 and 2000 have been collected at Lhasa, Dingri, Dangxiong and Amdo, Tibet. The sampling and analysis were based on WMO recommendations for a background network with some modifications according to local conditions and environmental characteristics. The following precipitation constituents and related parameters were measured: pH, conductivity, CO2 partial pressure, total suspended particles, and the content of K+, Na+, Ca2+, Mg2+, Fe, Mn, NH4 +, Cl-, NO2 -, NO3 -, SO4 2-Br-, HCO3 - and HPO4 2-. Some atmospheric dust samples have also been collected. Over 300 precipitation events have been measured for pH and conductivity. Among these, 60 have been analysed for their chemical components. The results show that Lhasa's precipitation events were constantly alkaline with weighted averages of pH 8.36 in the 1987-1988 period, and 7.5 for 1997 to 1999. Only one event was weakly acidic during 1997-1999. Although CO2 partial pressure, a major producer of acidity in natural water on the Plateau, falls with increasing elevation, the lowest measured CO2 partial pressure can only raise pH value by 0.1 units in the sampling areas. Chemical analysis indicates that the major contributor to alkaline precipitation is the continental dust, which is rich in calcium. The analysis also shows that Tibet is still one of the cleanest areas in the world with little air pollution. However, the decline of pH from the 1980s to 1990s, which was reflected by an increase of NO3 - and SO4 2- in precipitation, alerts us to the urgency of environmental protection in this fragile paradise. © 2002 Elsevier Science Ltd.     Charging mechanism in a SiO 2 matrix embedded with Si nanocrystals

    One of the applications of a Si nanocrystals (nc-Si) embedded in a Si O2 matrix is in the area of nonvolatile memory devices based on the charge storage in the material system. However, whether the charge trapping mainly occurs at the nc-SiSi O2 interface or in the nc-Si is still unclear. In this work, by x-ray photoemission spectroscopy analysis of the Si 2p peaks, the concentrations of both the nc-Si and the Si suboxides that exist at the nc-SiSi O2 interface are determined as a function of thermal annealing, and the charging effect is also measured by monitoring the shift of the surface C 1s peak. It is observed that the annealing-caused reduction of the total concentration of the interfacial suboxides is much faster than that of both the C 1s shift and the nc-Si concentration. In addition, the trend of the C 1s shift coincides with that of the nc-Si concentration. The results suggest that the Si nanocrystal, rather than the nc-SiSi O2 interface, plays the dominant role in the charging effect. © 2006 American Institute of Physics.published_or_final_versio

    Molecular cloning and characterization of a novel expressed sequence tag (EST) associated with fecundity in goats

    To screen the genes controlling the fecundity traits in goats, a DDRT-PCR technique was applied. We found a new EST which highly expressed in Chinese native prolific goat breed, Haimen goats. There exists a difference of EST expression level between the prolific and non-prolific goat breed, indicating EST might associate to fecundity in goats. A full-length cDNA with 2253 base pairs was obtained by the 3’- and 5’-RACE method based on the EST sequence encoding a protein segment of 201 amino acid residues. Tissue specific distribution and sequence analysis implicated the likely involvement of EST in the regulation of the hormones related to fecundity

    Biological impacts of 'hot-spot' mutations of hepatitis B virus X proteins are genotype B and C differentiated

    AIM: To investigate the biological impacts of “hot-spot” mutations on genotype B and C HBV X proteins (HBx). METHODS: Five types of “hot-spot” mutations of genotype B or C HBV X genes, which sequentially lead to the amino acid substitutions of HBx as I127T, F132Y, K130M+V131I, I127T+K130M+V131I, or K130M+V131I+F132Y, respectively, were generated by means of site-directed mutagenesis. To evaluate the anti-proliferative effects, HBx or related mutants’ expression vectors were transfected separately to the Chang cells by lipofectamine, and the cells were cultured in hygromycin selective medium for 14 d, drug-resistant colonies were fixed with cold methanol, stained with Giemsa dyes and scored (increase of the colonies indicated the reduction of the anti-proliferation activity, and vice versa). Different types of HBx expression vectors were co-transfected separately with the reporter plasmid pCMVβ to Chang cells, which were lysed 48 h post-transfection and the intra-cellular β-galactosidase activities were monitored (increase of the β-galactosidase activities indicated the reduction of the transactivation activity, and vice versa). All data obtained were calculated by paired-samples t-test. RESULTS: As compared to standard genotype B HBx, mutants of I127T and I127T+K130M+V131I showed higher transactivation and anti-proliferative activities, while the mutants of F132Y, K130M+V131I, and K130M+V131I+F132Y showed lower activities. As compared to standard genotype C HBx, I127T mutant showed higher transactivation activity, while the other four types of mutants showed no differences. With regard to anti-proliferative activity, compared to standard genotype C HBx, F132Y and K130M+V131I mutants showed lower activities, and K130M+V131I +F132Y mutant, on the other hand, showed higher activity, while the mutants of I127T and I127T+K130M+V131I showed no differences. CONCLUSION: “Hot-spot” mutations affect the anti-proliferation and transactivation activities of genotype B and/or C HBx, and the biological impacts of most “hot-spot” mutations on HBx are genotype B and C differentiated.published_or_final_versio
