Exponential increase of data has motivated advances of data storage technologies. As a promising storage media, DeoxyriboNucleic Acid (DNA) storage provides a much higher data density and superior durability, compared with state-of-the-art media. In this paper, we provide a tutorial on DNA storage and its role in molecular communications. Firstly, we introduce fundamentals of DNA-based molecular communications and storage (MCS), discussing the basic process of performing DNA storage in MCS. Furthermore, we provide tutorials on how conventional coding schemes that are used in wireless communications can be applied to DNA-based MCS, along with numerical results. Finally, promising research directions on DNA-based data storage in molecular communications are introduced and discussed in this paper

A novel cascade machine learning pipeline for Alzheimer’s disease identification and prediction

IntroductionAlzheimer’s disease (AD) is a progressive and irreversible brain degenerative disorder early. Among all diagnostic strategies, hippocampal atrophy is considered a promising diagnostic method. In order to proactively detect patients with early Alzheimer’s disease, we built an Alzheimer’s segmentation and classification (AL-SCF) pipeline based on machine learning.MethodsIn our study, we collected coronal T1 weighted images that include 187 patients with AD and 230 normal controls (NCs). Our pipeline began with the segmentation of the hippocampus by using a modified U2-net. Subsequently, we extracted 851 radiomics features and selected 37 features most relevant to AD by the Hierarchical clustering method and Least Absolute Shrinkage and Selection Operator (LASSO) algorithm. At last, four classifiers were implemented to distinguish AD from NCs, and the performance of the models was evaluated by accuracy, specificity, sensitivity, and area under the curve.ResultsOur proposed pipeline showed excellent discriminative performance of classification with AD vs NC in the training set (AUC=0.97, 95% CI: (0.96-0.98)). The model was also verified in the validation set with Dice=0.93 for segmentation and accuracy=0.95 for classification.DiscussionThe AL-SCF pipeline can automate the process from segmentation to classification, which may assist doctors with AD diagnosis and develop individualized medical plans for AD in clinical practice

Off-label use of Baricitinib improves moderate and severe atopic dermatitis in China through inhibiting MAPK and PI3K/Akt/mTOR pathway via targeting JAK-STAT signaling of CD4+ cells

As an inflammatory disease with a disrupted immune system, cytokine disorders in atopic dermatitis (AD) are closely related to the abnormal activation of JAK-STAT signal pathway. The critical relevance of the JAK-STAT signaling pathway to the pathogenesis of AD provides a strong rationale for JAK inhibitor research. Baricitinib, a small-molecule oral JAK inhibitor, has been proven to inhibit JAK-STAT signaling in a variety of diseases, including AD. It is currently available in China for off-label use. However, its efficacy in China and its mechanism are rarely reported. In our study, we found that the immune status of patients with moderate and severe AD was hyperactive. Among the 49 known immunotherapy targets, JAK1 and JAK2 genes on lymphocytes of AD patients were significantly upregulated, which was closely related to the symptom severity in moderate and severe AD patients. Baricitinib can improve immune hyperresponsiveness and clinical symptoms in moderate and severe AD by inhibiting the activation of Th2 cell subsets and the secretion of Th2-type cytokines through MAPK, mTOR and PI3K-Akt signaling pathways, providing an important theoretical basis for clinical off-label use of Baricitinib to treat moderate and severe AD

Legalizing Sex Industries or Not? A Comparison of the Growth Rate of Sexual Crimes and STDs Among China Mainland, Hong Kong and Taiwan

Existing research rarely establishes the relationship between the legitimacy of sex industry and the growth rate of sexual crimes and STDs. I conducted a research on the subject of China mainland, Hong Kong and Taiwan, because the laws towards sex industry in these three places are different: illegal in China mainland, partly legal in Hong Kong (prostitution is legal, but organizing prostitution is illegal) and legal in Taiwan after 2011; while other variables, such as culture and demographic structure, are similar. First, I compared the growth rate of sexual crimes and STDs in Taiwan before and after 2011. Then, I compared the difference in rate between Taiwan and China mainland before and after 2011. At last, I compared the difference in rate between Taiwan and Hong Kong before and after 2011. Partially legalizing sex industries is not significantly different from illegalizing sex industries in the average growth rate of sexual crimes but is significantly lower than illegalizing sex industries in the growth rate of STDs incidence. On the other hand, generally legalizing sex industries is significantly lower than illegalizing sex industries in the growth rate of sexual crimes but is not significantly from illegalizing sex industries in the growth rate of STDs incidence. In conclusion, legalizing sex industries is significantly lower than illegalizing sex industries in the growth rate of both sexual crimes and STDs incidence. Further research is needed to figure out how to design policies about sex industries to reach the lowest possible growth rate of sexual crimes and STDs incidence

Research Progress on Molecular Mechanism of Salt Tolerance in Rice

Rice is one of the important grain crops in the world and is sensitive to salt stress. The increasingly serious salinization of paddy soils is becoming a potential risk to the safe production of rice. Salt stress can cause osmotic stress, ion toxicity and oxidative stress in rice plant, ultimately leading to a decrease in rice quality and yield. Due to the ability of rice roots to absorb salt and secrete organic acids as well as the growth characteristics of water holding in the early stage and drainage in rice paddies in the later stage, rice is also an excellent crop for improving saline soil. Therefore, cultivating new rice varieties of salt tolerant and improving rice salt tolerance can effectively enhance the production potential of saline farmland, which is important to food security in China and the world. In recent years, quantitative genetics and molecular marker technology have been continuously developed. Through genetic, biochemical and molecular biology methods, a large number of salt-tolerant related QTLs and genes have been excavated, which is of great benefit to analyze the molecular mechanism of salt tolerance in rice and improve the breeding efficiency of salt-tolerant rice through molecular marker-assisted selection, gene editing and other molecular means. Nevertheless, many cloned genes related to salt tolerance are difficult to be applied in rice breeding currently as most of them are obtained through reverse genetics methods and exhibit salt tolerance only under overexpression conditions, or these genes are recessive genes. The study summarized the recent progress in the identification and mining of salt-tolerant genes in rice, and reviewed the research progress on molecular mechanisms of salt tolerance in rice from four aspects: osmotic regulation of organic matter, regulation of ion absorption and transport, regulation of reactive oxygen removal by antioxidant system and regulation of hormone. The challenges of future researches on salt tolerance of rice were also discussed in order to provide some suggestions for molecular breeding of salt tolerance of rice in the future
