    Role of Body Mass Index, Waist-to-Height and Waist-to-Hip Ratio in Prediction of Nonalcoholic Fatty Liver Disease

    Objective. To investigate the anthropometric indicators that can effectively predict the nonalcoholic fatty liver disease (NAFLD). Methods. The height, body weight, waist and hip circumference were measured, and body mass index (BMI), waist-to-height (WHtR) and waist-to-hip ratio (WHR) were calculated. M-H chi square test, logistic regression analysis, and receiver-operating characteristic (ROC) curve were employed for the analysis of risk factors. Patients or Materials. 490 patients were recruited, of whom 250 were diagnosed as NAFLD and 240 as non-NAFLD (control group). Results. Compared with the control group, the BMI, WHR, and WHtR were significantly higher in patients with NAFLD. Logistic regression analysis showed that BMI and WHR were effective prognostic factors of NAFLD. In addition, WHR plays a more important role in prediction of NAFLD by the area under curve. Conclusion. WHR is closely related to the occurrence of NAFLD. We assume that WHR is beneficial for the diagnosis NAFLD

The dioxane ring has a half-boat conformation, in which the C atom between the dioxane O atoms is −0.464 (8) Å out of the plane of the other five atoms. In the mol­ecule there is an intra­molecular N—H⋯O hydrogen bond, involving the NH H atom and the adjacent dioxane carbonyl O atom.     Photonic Crystal Fiber Mach-Zehnder Interferometer for Refractive Index Sensing

    We report on a refractive index sensor using a photonic crystal fiber (PCF) interferometer which was realized by fusion splicing a short section of PCF (Blaze Photonics, LMA-10) between two standard single mode fibers. The fully collapsed air holes of the PCF at the spice regions allow the coupling of PCF core and cladding modes that makes a Mach-Zehnder interferometer. The transmission spectrum exhibits sinusoidal interference pattern which shifts differently when the cladding/core surface of the PCF is immersed with different RI of the surrounding medium. Experimental results using wavelength-shift interrogation for sensing different concentrations of sucrose solution show that a resolution of 1.62 × 10−4–8.88 × 10−4 RIU or 1.02 × 10−4–9.04 × 10−4 RIU (sensing length for 3.50 or 5.00 cm, respectively) was achieved for refractive indices in the range of 1.333 to 1.422, suggesting that the PCF interferometer are attractive for chemical, biological, biochemical sensing with aqueous solutions, as well as for civil engineering and environmental monitoring applications

    Clinical and Virological Characteristics of Chronic Hepatitis B Patients with Hepatic Steatosis

    Abstract Objective: This study aimed to explore clinical and virological characteristics of chronic hepatitis B (CHB) patients with hepatic steatosis in order to provide a theoretical basis for the prevention and control of hepatic steatosis. Methods: A total of 360 CHB inpatients were recruited from Affiliated Dongnan Hospital of Xiamen University and divided into hepatic steatosis group and non-hepatic steatosis group. The body mass index (BMI), waist-to-hip ratio (WHR), fasting blood glucose (FBG), triglyceride (TG), total cholesterol (TC), aminotransferase (AST), alanine aminotransferase (ALT), gamma-glutamyl transpeptidase (GGT), hepatitis B e antigen (HBeAg), hepatitis B virus DNA (HBV DNA) and hepatic histological changes were detected and compared between the two groups. The association of these factors with hepatic steatosis was evaluated in CHB patients. Results: BMI, FPG, TG, TC, GGT, AST and HBV DNA showed statistically significant differences between two groups (P<0.01). The patients with hepatic steatosis had markedly higher BMI, FBG, TG and TC than those without steatosis did. No significant differences were found in ALT and HBeAg between two groups (P>0.05). In male patients, there was marked difference in the WHR between two groups (P < 0.01), which was not found in female patients (P > 0.05). The severity of hepatic steatosis increased in patients with hepatic steatosis, compared to those without steatosis (P < 0. 01), but the severities of inflammation and fibrosis in the non-hepatic steatosis group were dramatically higher than those in the hepatic steatosis group (P < 0. 01). Conclusions: BMI, WHR, FBG, TG and TC appeared to be influencing factors of CHB combined with hepatic steatosis. Hepatic steatosis in CHB patients was closely related to changes in anthropometric indices and metabolic factors but not HBV. It is necessary to improve these factors to effectively prevent hepatic steatosis in CHB patients

    A Microfluidic Long-Period Fiber Grating Sensor Platform for Chloride Ion Concentration Measurement

    Optical fiber sensors based on waveguide technology are promising and attractive in chemical, biotechnological, agronomy, and civil engineering applications. A microfluidic system equipped with a long-period fiber grating (LPFG) capable of measuring chloride ion concentrations of several sample materials is presented. The LPFG-based microfluidic platform was shown to be effective in sensing very small quantities of samples and its transmitted light signal could easily be used as a measurand. The investigated sample materials included reverse osmosis (RO) water, tap water, dilute aqueous sample of sea sand soaked in RO water, aqueous sample of sea sand soaked in RO water, dilute seawater, and seawater. By employing additionally a chloride ion-selective electrode sensor for the calibration of chloride-ion concentration, a useful correlation (R2 = 0.975) was found between the separately-measured chloride concentration and the light intensity transmitted through the LPFG at a wavelength of 1,550 nm. Experimental results show that the sensitivity of the LPFG sensor by light intensity interrogation was determined to be 5.0 × 10−6 mW/mg/L for chloride ion concentrations below 2,400 mg/L. The results obtained from the analysis of data variations in time-series measurements for all sample materials show that standard deviations of output power were relatively small and found in the range of 7.413 × 10−5−2.769 × 10−3 mW. In addition, a fairly small coefficients of variations were also obtained, which were in the range of 0.03%–1.29% and decreased with the decrease of chloride ion concentrations of sample materials. Moreover, the analysis of stability performance of the LPFG sensor indicated that the random walk coefficient decreased with the increase of the chloride ion concentration, illustrating that measurement stability using the microfluidic platform was capable of measuring transmitted optical power with accuracy in the range of −0.8569 mW/ h to −0.5169 mW/ h. Furthermore, the bias stability was determined to be in the range of less than 6.134 × 10−8 mW/h with 600 s time cluster to less than 1.412 × 10−6 mW/h with 600 s time cluster. Thus, the proposed LPFG-based microfluidic platform has the potential for civil, chemical, biological, and biochemical sensing with aqueous solutions. The compact (3.5 × 4.2 cm), low-cost, real-time, small-volume (∼70 μL), low-noise, and high-sensitive chloride ion sensing system reported here could hopefully benefit the development and applications in the field of chemical, biotechnical, soil and geotechnical, and civil engineering

    A comparison of the impact on neuronal transcriptome and cognition of rAAV5 transduction with three different doses in the mouse hippocampus

    IntroductionRecombinant adeno-associated viruses (rAAVs) are widely used in genetic therapeutics. AAV5 has shown superior transduction efficiency, targeting neurons and glial cells in primate brains. Nonetheless, the comprehensive impact of AAV5 transduction on molecular and behavioral alterations remains unexplored. This study focuses on evaluating the effects of AAV5 transduction in the hippocampus, a critical region for memory formation and emotional processes.MethodsIn this experiment, fluorescence-activated cell sorting (FACS) was utilized to isolate the mCherry-labeled pyramidal neurons in the hippocampus of CaMkIIα-cre mice following three different doses rAAV5-mCherry infusion after 3 weeks, which were then subjected to RNA sequencing (RNA-seq) to assess gene expression profiles. The cytokines concentration, mRNA expression, and glial response in hippocampi were confirmed by ELASA, digital droplet PCR and immunohistochemistry respectively. Locomotion and anxiety-like behaviors were elevated by Open Field Test and Elevated Plus Maze Test, while the Y-Maze were used to assessed spatial working memory. Recognition memory and fear responses were examined by the Novel Object Recognition Test and Fear Conditioning Test, respectively.ResultsWe found that 2.88 × 1010 v.g rAAV5 transduction significantly upregulated genes related to the immune response and apoptosis, and downregulated genes associated with mitochondrial function and synaptic plasticity in hippocampal pyramidal neurons, while did not induce neuronal loss and gliosis compared with 2.88 × 109 v.g and 2.88 × 108 v.g. Furthermore, the same doses impaired working memory and contextual fear memory, without effects on locomotion and anxiety-related behaviors.DiscussionOur findings highlight the detrimental impact of high-dose administration compared to median-dose or low-dose, resulting in increased neural vulnerability and impaired memory. Therefore, when considering the expression effectiveness of exogenous genes, it is crucial to also take potential side effects into account in clinical settings. However, the precise molecular mechanisms underlying these drawbacks of high-dose rAAV5-mCherry still require further investigation in future studies
