    A HIPERGLICEMIA E OS MECANISMOS ENVOLVIDOS NAS DISFUNÇÕES VASCULARES DO DIABETES MELLITUS

    O Diabetes Mellitus (DM) é uma síndrome de etiologia múltipla, decorrente da falta de insulina e/ou da incapacidade de a insulina exercer adequadamente seus efeitos. Caracteriza-se por hiperglicemia crônica, com distúrbios do metabolismo dos carboidratos, lipídios e proteínas. Constitui-se um importante problema de saúde pública, devido à sua elevada prevalência, ao desenvolvimento de complicações crônicas incapacitantes e ao alto custo econômico gerado pelo seu tratamento. A hiperglicemia é um dos fatores mais importantes para o aparecimento e progressão das complicações vasculares do DM. A elevação persistente nas concentrações da glicose provoca mudanças na estrutura e na função de proteínas plasmáticas e tissulares, com efeitos indesejáveis sobre a saúde do portador de diabetes. O aumento da via dos polióis, glicosilação não-enzimática, com a produção dos produtos finais da glicação avançada (AGEs); estresse oxidativo, gerando o aumento de radicais livres e alteração da proteína kinase C, levando à disfunção endotelial e à aterogênese, são alguns dos mecanismos que tentam explicar o dano vascular induzido pela hiperglicemia. O objetivo desta revisão bibliográfica é discutir os mecanismos pelos quais a hiperglicemia pode desencadear as disfunções vasculares associadas com o DM

    Spermatogenesis is a complex, highly organized and coordinated process that results in the production of male gametes. This process is influenced by a number of drugs that enhance physical performance, which have been used mainly by young people, athletes or not, seeking to gain athletic performance or only a prominent social position. The objective of this study was to evaluate the effects of the anabolic steroid nandrolone decanoate on the efficiency of spermatogenesis in sedentary rats and rats subjected to physical training. Twenty-four male rats were divided into four experimental groups: sedentary control, sedentary treated, trained control and trained treated. Treated animals received intramuscular injection of nandrolone decanoate (0.5 mg kg-1 body weight) during the last four weeks of physical training. The training program consisted of running on a programmable ergometer treadmill, tailored to train eight rats simultaneously, for nine weeks. We evaluated the morphological and morphoquantitative profiles of the male reproductive system. The results showed that the treatment causes a reduction in the efficiency of spermatogenesis; however, when associated with physical training, this compensates for the anabolic action, keeping the process of spermatogenesis within normality.<br><br>A espermatogênese é um processo complexo, altamente organizado e coordenado, que resulta na produção dos gametas masculinos. Este processo é influenciado por uma série de drogas potencializadoras do desempenho físico, as quais têm sido utilizadas principalmente, por jovens, atletas ou não, que buscam adquirir maior desempenho esportivo ou mesmo apenas uma posição de destaque na sociedade. O objetivo deste estudo foi avaliar os efeitos do anabolizante esteroide decanoato de nandrolona na eficiência da espermatogênese de ratos sedentários e submetidos ao treinamento físico. Foram utilizados 24 ratos machos divididos em quatro grupos experimentais: sedentário-controle, sedentário tratado, treinado-controle e treinado tratado. Os animais tratados receberam injeção intramuscular de decanoato de nandrolona (0,5 mg kg-1) durante as quatro últimas semanas de treinamento físico. O programa de treinamento consistiu de corrida em esteira ergométrica programável, adaptada para treinar oito ratos simultaneamente, durante nove semanas. Foi avaliado o perfil morfológico e morfoquantitativo do sistema reprodutor masculino. Os resultados demonstraram que o tratamento causa redução da eficiência da espermatogênese, no entanto, o treinamento físico quando associado compensa a ação do anabolizante, mantendo o processo de espermatogênese normal

    The mammalian testis is a complex organ with endocrine and exocrine functions. It consists of seminiferous tubules where the production of the male gametes called spermatogenesis occurs. This process is influenced by a number of factors including the use of physical performance-enhancing drugs. The objective of this study was to evaluate the effects of the anabolic steroid nandrolone decanoate on the morphofunctional structure of testes in sedentary rats and rats subjected to moderate aerobic exercise training. Twenty-four male rats were divided into four experimental groups: sedentary control, sedentary treated, trained control and trained treated.The training lasted eight weeks and consisted of running on a programmable ergometer treadmill, tailored to train eight rats simultaneously. Treated animals received intramuscular injections of nandrolone decanoate (0.5 mg kg-1 body weight) during the last four weeks of physical training, while the control groups received intramuscular injections of vehicle (vegetable oil).The male reproductive system morphology showed that treatment with nandrolone decanoate, in both sedentary and trained rats, promoted morphological and functional changes that result in reduced efficiency of spermatogenesis

    Supplementation of cow milk naturally enriched in polyunsaturated fatty acids and polyphenols to growing rats.

    This study investigated whether intake of cow milk, naturally enriched with polyunsaturated fatty acids (PUFA, omega-3) and polyphenols (from propolis extract and vitamin E), from manipulation of cow's diet, would result in positive metabolic effects in rats from weaning until adulthood. Male Wistar rats were fed a standard chow diet or a hypercaloric diet (metabolically disturbed rats, obese) which was supplemented with either whole common milk, milk enriched with PUFA (PUFA-M) or milk enriched with PUFA and polyphenols (PUFA/P-M), at 5mL/kg body weight,having water as control. Whole milk supplementation increased initial weight gain and reduced gain in the adulthood of rats. Intake of common milk reduced cholesterol levels in non-obese rats and reduced insulin resistance in obese rats. PUFA-milk showed a decreasing effect on plasma triacylglycerol and VLDL concentrations, increasing plasma HDL concentration and reducing adipocyte size of non-obese rats, but no effect was observed in obese rats. PUFA/P-milk in obese rats resulted in greater deposition of muscle mass and mesenteric fat, with a tendency to lower LDL levels, and resulted a visceral fat accumulation in non-obese rats. Thus, whole common milk and PUFA-rich milk have shown to be beneficial in a normal metabolic condition, whereas common milk and milk enriched with PUFA and polyphenols improve metabolic effects of obesity

    Sweeteners based on stevia extract contain a series of diterpene glycosides derivatives from steviol, standing out the rebaudioside-A. There is no tabletop sweeteners in the market formulated purely with rebaudioside-A yet, so its use in foods depends on the development of new products followed by physicochemical and sensory evaluations. This work presents the formulation of a diet strawberry jam dyed with cranberry juice and sweetened with rebaudioside-A purified from stevia plants of the lineage UEM-320 developed in the Centro de Estudos de Produtos Naturais da Universidade Estadual de Maringá. Evaluations of physicochemical properties, microbiological and sensory characteristics were carried out for the product in comparison with a control sweetened with equal amount of sucralose. The results showed that the physicochemical characteristics of the sample and the control are not significantly different and the supplementation with cranberry juice increased both color and total phenolic content in both samples. The sensory acceptability indicated a significant preference for the formulation sweetened with 100% of rebaudioside-A, only in the items flavor and purchase intent. We concluded that rebaudioside-A has a better sensory performance than sucralose, even this last one being 1.33 fold sweeter than rebaudioside-A

    The increasing incidence of diabetes mellitus is becoming a serious threat to human health in various parts of the world. Studies with dairy products have shown a potential beneficial effect against diabetes. This experiment evaluated the supplementation of milk naturally enriched with polyunsaturated fatty acids (PUFA) and polyphenols in rats with streptozotocin-induced diabetes. Forty male 28-day-old Wistar rats were distributed in four experimental treatments of diabetic animals (streptozotocin induction) and a normal group (non-induced). Experimental treatments were: control (water), whole common milk (COM-M), milk enriched with PUFA (PUFA-M), milk enriched with PUFA and polyphenols (PUFA/P-M) through a special diet offered to dairy cows. Milk supplementation at a dose 5 mL/kg body weight was performed for 77 days, 42 days before and 35 days after diabetes induction. The COM-M supplementation increased brown fat deposits, reduced post-induction glucose levels, reduced blood fructosamine levels, and improved glucose tolerance. Milk enriched with PUFA reduced final fasting glucose, LDL levels, and improved blood antioxidant capacity. Milk enriched with PUFA and polyphenols promoted an increase in gastrocnemius muscle mass, and a reduction in mesenteric fat and LDL levels. Milk intake, with an emphasis on milk enriched with PUFA and polyphenols, attenuated the metabolic disorders of streptozotocin-induced diabetes in rats
