    In his library at Trinity College, Cambridge University, around the year I860, William Whewell (1794-1866) engages in conversation with a company of thinkers on the province of metaphysics and physics, to form a comprehensive scientific belief. In attendance with him are Lord Francis Bacon (1561-1626), Sir Robert Boyle (1627-1691 ), Sir Isaac Newton (1642-1727) , John Henry Cardinal Newman (1801-1890), Professor Alfred North Whitehead (1861-1947), and Pope John Paul II (b. 1920).     In-situ Apparent Conductivity Measurements and Microbial Population Distribution at a Hydrocarbon-Contaminated Site

    We investigated the bulk electrical conductivity and microbial population distribution in sediments at a site contaminated with light nonaqueous-phase liquid (LNAPL). The bulk conductivity was measured using in-situ vertical resistivity probes; the most probable number method was used to characterize the spatial distribution of aerobic heterotrophic and oil-degrading microbial populations. The purpose of this study was to assess if high conductivity observed at aged LNAPL-impacted sites may be related to microbial degradation of LNAPL. The results show higher bulk conductivity coincident with LNAPL-impacted zones, in contrast to geoelectrical models that predict lower conductivity in such zones. The highest bulk conductivity was observed to be associated with zones impacted by residual and free LNAPL. Data from bacteria enumeration from sediments close to the resistivity probes show that oil-degrading microbes make up a larger percentage (5-55%) of the heterotrophic microbial community at depths coincident with the higher conductivity compared to ∼5% at the uncontaminated location. The coincidence of a higher percentage of oil-degrading microbial populations in zones of higher bulk conductivity suggests that the higher conductivity in these zones may result from increased fluid conductivity related to microbial degradation of LNAPL, consistent with geochemical studies that suggest that intrinsic biodegradation is occurring at the site. The findings from this study point to the fact that biogeochemical processes accompanying biodegradation of contaminants can potentially alter geoelectrical properties of the subsurface impacted media

    Evidence for Microbial Enhanced Electrical Conductivity in Hydrocarbon-Contaminated Sediments

    Bulk electrical conductivity of sediments during microbial mineralization of diesel was investigated in a mesoscale laboratory experiment consisting of biotic contaminated and uncontaminated columns. Population numbers of oil degrading microorganisms increased with a clear pattern of depth zonation within the contaminated column not observed in the uncontaminated column. Microbial community structure determined from ribosomal DNA intergenic spacer analysis showed a highly specialized microbial community in the contaminated column. The contaminated column showed temporal increases in bulk conductivity, dissolved inorganic carbon, and calcium, suggesting that the high bulk conductivity is due to enhanced mineral weathering from microbial activity. The greatest change in bulk conductivity occurred in sediments above the water table saturated with diesel. Variations in electrical conductivity magnitude and microbial populations and their depth distribution in the contaminated column are similar to field observations. The results of this study suggest that geophysical methodologies may potentially be used to investigate microbial activity

    A Cloud-based Deep Learning Framework for Remote Detection of Diabetic Foot Ulcers

    This research proposes a mobile and cloud-based framework for the automatic detection of diabetic foot ulcers and conducts an investigation of its performance. The system uses a cross-platform mobile framework which enables the deployment of mobile apps to multiple platforms using a single TypeScript code base. A deep convolutional neural network was deployed to a cloud-based platform where the mobile app could send photographs of patient's feet for inference to detect the presence of diabetic foot ulcers. The functionality and usability of the system were tested in two clinical settings: Salford Royal NHS Foundation Trust and Lancashire Teaching Hospitals NHS Foundation Trust. The benefits of the system, such as the potential use of the app by patients to identify and monitor their condition are discussed

    Comparing the immune response to a novel intranasal nanoparticle PLGA vaccine and a commercial BPI3V vaccine in dairy calves

    peer-reviewedBackground There is a need to improve vaccination against respiratory pathogens in calves by stimulation of local immunity at the site of pathogen entry at an early stage in life. Ideally such a vaccine preparation would not be inhibited by the maternally derived antibodies. Additionally, localized immune response at the site of infection is also crucial to control infection at the site of entry of virus. The present study investigated the response to an intranasal bovine parainfluenza 3 virus (BPI3V) antigen preparation encapsulated in PLGA (poly dl-lactic-co-glycolide) nanoparticles in the presence of pre-existing anti-BPI3V antibodies in young calves and comparing it to a commercially available BPI3V respiratory vaccine. Results There was a significant (P < 0.05) increase in BPI3V-specific IgA in the nasal mucus of the BPI3V nanoparticle vaccine group alone. Following administration of the nanoparticle vaccine an early immune response was induced that continued to grow until the end of study and was not observed in the other treatment groups. Virus specific serum IgG response to both the nanoparticle vaccine and commercial live attenuated vaccine showed a significant (P < 0.05) rise over the period of study. However, the cell mediated immune response observed didn’t show any significant rise in any of the treatment groups. Conclusion Calves administered the intranasal nanoparticle vaccine induced significantly greater mucosal IgA responses, compared to the other treatment groups. This suggests an enhanced, sustained mucosal-based immunological response to the BPI3V nanoparticle vaccine in the face of pre-existing antibodies to BPI3V, which are encouraging and potentially useful characteristics of a candidate vaccine. However, ability of nanoparticle vaccine in eliciting cell mediated immune response needs further investigation. More sustained local mucosal immunity induced by nanoparticle vaccine has obvious potential if it translates into enhanced protective immunity in the face of virus outbreak

    D-cycloserine augmentation of exposure-based cognitive behavior therapy for anxiety, obsessive-compulsive, and posttraumatic stress disorders: a systematic review and meta-analysis of individual participant data

    Importance: Whether and under which conditions D-cycloserine (DCS) augments the effects of exposure-based cognitive behavior therapy for anxiety, obsessive-compulsive, and posttraumatic stress disorders is unclear. Objective: To clarify whether DCS is superior to placebo in augmenting the effects of cognitive behavior therapy for anxiety, obsessive-compulsive, and posttraumatic stress disorders and to evaluate whether antidepressants interact with DCS and the effect of potential moderating variables. Data Sources: PubMed, EMBASE, and PsycINFO were searched from inception to February 10, 2016. Reference lists of previous reviews and meta-analyses and reports of randomized clinical trials were also checked. Study Selection: Studies were eligible for inclusion if they were (1) double-blind randomized clinical trials of DCS as an augmentation strategy for exposure-based cognitive behavior therapy and (2) conducted in humans diagnosed as having specific phobia, social anxiety disorder, panic disorder with or without agoraphobia, obsessive-compulsive disorder, or posttraumatic stress disorder. Data Extraction and Synthesis: Raw data were obtained from the authors and quality controlled. Data were ranked to ensure a consistent metric across studies (score range, 0-100). We used a 3-level multilevel model nesting repeated measures of outcomes within participants, who were nested within studies. Results: Individual participant data were obtained for 21 of 22 eligible trials, representing 1047 of 1073 eligible participants. When controlling for antidepressant use, participants receiving DCS showed greater improvement from pretreatment to posttreatment (mean difference, -3.62; 95% CI, -0.81 to -6.43; P = .01; d = -0.25) but not from pretreatment to midtreatment (mean difference, -1.66; 95% CI, -4.92 to 1.60; P = .32; d = -0.14) or from pretreatment to follow-up (mean difference, -2.98, 95% CI, -5.99 to 0.03; P = .05; d = -0.19). Additional analyses showed that participants assigned to DCS were associated with lower symptom severity than those assigned to placebo at posttreatment and at follow-up. Antidepressants did not moderate the effects of DCS. None of the prespecified patient-level or study-level moderators was associated with outcomes. Conclusions and Relevance: D-cycloserine is associated with a small augmentation effect on exposure-based therapy. This effect is not moderated by the concurrent use of antidepressants. Further research is needed to identify patient and/or therapy characteristics associated with DCS response.2018-05-0

    Interleukin 10-Mediated Response and Correlated Anemia in a Patient with Advanced Non-Small Cell Lung Carcinoma

    Anemia in cancer patients is associated with poor quality of life, reduced response to therapy, and decreased overall survival. We describe a case of a 56-year old woman with advanced metastatic non-small cell lung carcinoma who demonstrated marked response to a novel combinational immunotherapy approach involving a long-acting PEGylated construct of recombinant human Interleukin-10 with Nivolumab, an anti-PD-L1 checkpoint inhibitor. While on treatment, the patient developed severe anemia and hyper-ferritinemia requiring RBC transfusion support.
