    Listeriolysin S, a Novel Peptide Haemolysin Associated with a Subset of Lineage I Listeria monocytogenes

    peer-reviewedStreptolysin S (SLS) is a bacteriocin-like haemolytic and cytotoxic virulence factor that plays a key role in the virulence of Group A Streptococcus (GAS), the causative agent of pharyngitis, impetigo, necrotizing fasciitis and streptococcal toxic shock syndrome. Although it has long been thought that SLS and related peptides are produced by GAS and related streptococci only, there is evidence to suggest that a number of the most notorious Gram-positive pathogenic bacteria, including Listeria monocytogenes, Clostridium botulinum and Staphylococcus aureus, produce related peptides. The distribution of the L. monocytogenes cluster is particularly noteworthy in that it is found exclusively among a subset of lineage I strains; i.e., those responsible for the majority of outbreaks of listeriosis. Expression of these genes results in the production of a haemolytic and cytotoxic factor, designated Listeriolysin S, which contributes to virulence of the pathogen as assessed by murine- and human polymorphonuclear neutrophil–based studies. Thus, in the process of establishing the existence of an extended family of SLS-like modified virulence peptides (MVPs), the genetic basis for the enhanced virulence of a proportion of lineage I L. monocytogenes may have been revealed.Work is funded by the Irish Government under the National Development Plan, through a Science Foundation Ireland Investigator award to CH, PR and PC (06/IN.1/B98)

    ABSTRACT The U.S. Solar Rating and Certification Corporation (SRCC) issues ratings for the thermal performance of solar collectors and solar water heaters (SWH). A bias favoring unglazed collectors currently exists because unglazed collectors have been tested under ASHRAE96 which specifies low wind during testing, whereas glazed collectors are tested under ASHRAE93 which specifies high winds. Wind is mostly negligible for glazed collectors, but it significantly affects unglazed collector efficiency. This bias didn't matter until ~2007 when unglazed SWH began to directly compete against glazed SWH under SRCC/OG300. It is suggested here that wind bias be mitigated by using a calibrated collector model to derive a wind correction to the measured efficiency curve. The resulting models depend explicitly on wind velocity, and could provide unbiased ratings. For unglazed collectors, the forced convection correlation has to be fit to wind-dependent collector data. The calculated rating increase for glazed systems is small, depending on collector insulation levels. The corresponding decrease for typical unglazed systems is ~20%, depending on unglazed collector type and wind coupling

    Parent-Reported Provider Recommendation of HPV Vaccination Among Minority Adolescents Before and During the Covid-19 Pandemic: Findings From the National Immunization Survey-Teen, 2019-2021

    Provider recommendation of human papillomavirus (HPV) vaccination among adolescents has steadily improved over the years, however, limited research has been conducted to examine if the COVID-19 pandemic disrupted this positive trend in parent-reported provider recommendation among minority adolescents. Therefore, we conducted the present study to determine if there is an association between the pandemic and parent-reported provider recommendation of HPV vaccine among non-Hispanic black and Hispanic adolescents. We also examined whether any changes in parent-reported provider recommendation in the years 2019, 2020, and 2021 differed by race or ethnicity. Using a cross-sectional design to examine data from the National Immunization Survey-Teen (2019-2021), a moderation analysis and logistic regression analysis were performed to model race-specific variation in parent-reported provider recommendation (n = 50,739). We found that Hispanic parents had lower odds (aOR = 0.80 [0.71, 0.91]) of reporting receiving a recommendation compared to non-Hispanic white parents. We also found that the odds of parent-reported provider recommendation were higher in 2020 (aOR = 1.15 [1.03-1.29]) than in 2019. Other variables-age, region, sex, health insurance status, and poverty status-were all associated with parent-reported provider recommendation. These findings demonstrated that the pandemic may not have triggered any race-related gap in the recommendation of HPV vaccines, however, more pandemic-resilient public health efforts are needed to improve parent and provider communication regarding HPV vaccination of adolescents

    Analyzing the Relationship Between Perception of Safety and Reported Crime in n Urban Neighborhood Using GIS and Sketch Maps

    This study analyzes the perception of safety among residents of Main South neighborhood in Worcester, MA, USA and compares it to reported crimes. This neighborhood is the focus of a community-based crime reduction project funded by the Bureau of Justice Assistance, the policy development arm of the U.S. Department of Justice. We collected social disorder and violent crime data from the Worcester Police Department and conducted 129 household surveys to understand residents’ perception of safety in the neighborhood and trust in community institutions. The surveys included a map on which residents indicated where they felt unsafe. The goal of this research was twofold: (1) to use geographic information systems (GIS) to analyze the differences in perception of neighborhood safety by gender and length of residency in the neighborhood and (2) to explore the relationship between reported crime and perception of safety in the community. Findings indicate that the strength of the correlation between perceived safety and reported crime varies and that gender and length of residency are significant factors that shape perceptions of safety. Implications of this research suggest the need for comprehensive community-based development initiatives to offer differentiated strategies that address a broad range of safety perceptions and crime experiences among a diverse group of residents

    The Prevalence of Human Papillomavirus Vaccination Among Racial and Ethnic Minority Adolescents During the Covid-19 Pandemic

    BACKGROUND: Human papillomavirus (HPV) vaccination among adolescents has steadily improved over the past several years. However, research conducted to determine whether the COVID-19 pandemic disrupted this positive trend in HPV vaccine initiation among racial and ethnic minority adolescents is limited. Therefore, this study was conducted to determine if the COVID-19 pandemic and the resulting changes in the US health-care sector affected the increasing HPV vaccine initiation among non-Hispanic Black and Hispanic adolescents aged 13-17 years. METHODS: Using a cross-sectional design to examine data from the National Immunization Survey-Teen (2019-2021), logistic regression and moderation analysis were used to model race-specific variations in HPV vaccine initiation (n = 49 031). Two-sided P values of up to .05 were considered statistically significant. RESULTS: Hispanic (adjusted odds ratio [AOR] = 1.35, 95% confidence interval [CI] = 1.16 to 1.57) and non-Hispanic Black (AOR = 1.29, 95% CI = 1.10 to 1.51) adolescents had higher odds of HPV vaccine initiation than did non-Hispanic White adolescents. Additionally, the odds of HPV vaccine initiation were higher in 2021 (AOR = 1.22, 95% CI = 1.08 to 1.38) than in 2019. Other variables-age, region, sex, insurance status, and poverty status-were also associated with HPV vaccine initiation. CONCLUSION: These findings demonstrate that during the COVID-19 pandemic, racial and ethnic minorities had higher odds of receiving the HPV vaccine. Therefore, more research of the impact of the pandemic on HPV vaccine initiation among non-Hispanic White and racial and ethnic minority adolescents is needed

    Recycled arc mantle recovered from the Mid-Atlantic Ridge

Nature Communications, 11(1), (2020): 3887, doi:10.1038/s41467-020-17604-8.Plate tectonics and mantle dynamics necessitate mantle recycling throughout Earth’s history, yet direct geochemical evidence for mantle reprocessing remains elusive. Here we present evidence of recycled supra-subduction zone mantle wedge peridotite dredged from the Mid-Atlantic Ridge near 16°30′N. Peridotite trace-element characteristics are inconsistent with fractional anhydrous melting typically associated with a mid-ocean ridge setting. Instead, the samples are best explained by hydrous flux melting which changed the melting reactions such that clinopyroxene was not exhausted at high degrees of melting and was retained in the residuum. Based on along-axis ridge depth variations, this buoyant refractory arc mantle is likely compensated at depth by denser, likely garnet-rich, lithologies within the mantle column. Our results suggest that highly refractory arc mantle relicts are entrained in the upper mantle and may constitute >60% of the upper mantle by volume. These highly refractory mantle domains, which contribute little to mantle melting, are under-represented in compilations of mantle composition that rely on inverted basalt compositions alone.We thank the science party for their dutiful collection and description of dredge samples, and in particular chief scientist Dr. Deborah K.     The Lantibiotic Lacticin 3147 Prevents Systemic Spread of Staphylococcus aureus in a Murine Infection Model

    The objective of this study was to investigate the in vivo activity of the lantibiotic lacticin 3147 against the luminescent Staphylococcus aureus strain Xen 29 using a murine model. Female BALB/c mice (7 weeks old, 17 g) were divided into groups (n = 5) and infected with the Xen 29 strain via the intraperitoneal route at a dose of 1 × 106 cfu/animal. After 1.5 hr, the animals were treated subcutaneously with doses of phosphate-buffered saline (PBS; negative control) or lacticin 3147. Luminescent imaging was carried 3 and 5 hours postinfection. Mice were then sacrificed, and the levels of S. aureus Xen 29 in the liver, spleen, and kidneys were quantified. Notably, photoluminescence and culture-based analysis both revealed that lacticin 3147 successfully controlled the systemic spread of S. aureus in mice thus indicating that lacticin 3147 has potential as a chemotherapeutic agent for in vivo applications
