    Interferon-Induced Guanylate Binding Protein-1 (GBP-1) Mediates an Antiviral Effect against Vesicular Stomatitis Virus and Encephalomyocarditis Virus

    AbstractA cDNA encoding the human guanylate binding protein-1 (hGBP-1) was expressed in HeLa cells using a constitutive expression vector. Stably transfected clones expressing hGBP-1 exhibited resistance to the cytopathic effect mediated by both vesicular stomatitis virus (VSV) and encephalomyocarditis virus (EMCV) and produced less viral progeny than control cells following infection with these viruses. To study the role hGBP-1 plays in the IFN-mediated antiviral effect, cells were stably transfected with a construct expressing antisense RNA for hGBP-1. VSV infection of IFN-α-treated antisense RNA-expressing cells produced an amount of virus comparable to that produced in the parental cell line, while EMCV infection of the IFN-α-treated transfected cells and VSV and EMCV infection of the IFN-γ-treated transfected cells produced far more virus than was produced in the parental cell line. These results demonstrate that GBP-1 mediates an antiviral effect against VSV and EMCV and plays a role in the IFN-mediated antiviral response against these viruses

    Multiple causes of interannual sea surface temperature variability in the equatorial Atlantic Ocean

    The eastern equatorial Atlantic Ocean is subject to interannual fluctuations of sea surface temperatures, with climatic impacts on the surrounding continents. The dynamic mechanism underlying Atlantic temperature variability is thought to be similar to that of the El Nino/Southern Oscillation (ENSO) in the equatorial Pacific, where air-sea coupling leads to a positive feedback between surface winds in the western basin, sea surface temperature in the eastern basin, and equatorial oceanic heat content. Here we use a suite of observational data, climate reanalysis products, and general circulation model simulations to reassess the factors driving the interannual variability. We show that some of the warm events can not be explained by previously identified equatorial wind stress forcing and ENSO-like dynamics. Instead, these events are driven by a mechanism in which surface wind forcing just north of the equator induces warm ocean temperature anomalies that are subsequently advected toward the equator. We find the surface wind patterns are associated with long-lived subtropical sea surface temperature anomalies and suggest they therefore reflect a link between equatorial and subtropical Atlantic variability

    Leptopilina heterotoma and L. boulardi: Strategies to avoid cellular defense responses of Drosophila melanogaster

    Eggs of three strains of the cynipid parasitoid Leptopilina heterotoma and a Tunisian strain (G317) of L. boulardi are not encapsulated by hemocytes of Drosophila melanogaster hosts, but the eggs of a Congolese strain (L104) of L. boulardi are encapsulated. To determine the reason for the difference in host response against the parasitoid eggs, lamellocytes (hemocytes that encapsulate foreign objects and form capsules around endogenous tissues in melanotic tumor mutants) were examined in host larvae parasitized by the five Leptopilina strains. Parasitization by the three L. heterotoma strains affected the morphology of host lamellocytes and suppressed endogenous melanotic capsule formation in melanotic tumor hosts. L104 did not alter the morphology of host lamellocytes nor block tumor formation in melanotic tumor mutant hosts. The morphology of some lamellocytes was affected by G317 parasitization but host lamellocytes were still capable of forming melanotic tumors and encapsulating dead supernumerary parasitoid larvae. Therefore, the eggs of strains affecting lamellocyte morphology are protected from encapsulation by the host's blood cells. L. heterotoma eggs float freely in the host hemocoel but L. boulardi eggs are attached to host tissue surfaces. Lamellocytes cannot infiltrate the attachment site so the capsule around the L104 egg remains incomplete.
