Relação entre presença de anticorpos anti-leishmania spp. e carga parasitária em sangue de cães de área endêmica para leishmaniose visceral

Visceral leishmaniasis (VL) control strategies in Brazil are based on the early detection and treatment of the patients, vector control by insecticide and the serological screening and euthanasia of positive dogs. Thus, canine visceral leishmaniasis (CVL) diagnosis must accurately identify canine reservoir hosts to ensure the efficiency of screening. Considering that the humoral immune response of dogs to natural Leishmania spp. infection is dependent on parasite load, our aim was to evaluate if outcomes of the diagnostic kits currently used in canine seroepidemiological surveys in Brazil (DPP-BioManguinhos™ rapid test and EIA-ELISA Indirect ELISA BioManguinhos™) could be related to the blood parasite loads in dogs. Six hundred and 10 dogs from the VL endemic region of Northwest São Paulo State were sampled for this evaluation. Positivity varied from 15.9 to 68.4% between diagnostic techniques, due to differences in test sensitivity and specificity. Overall, positive dogs had higher mean parasite loads than negative dogs. Nevertheless, the potential of transmission of the negative dogs shall be investigate

Raising of horn fly haematobia irritans (L.) (diptera: muscidae) in laboratory by means of egg and larva inoculation

Several studies have required Haematobia irritans (L.) raising in laboratory. The present study assessed two methods of inoculating immature forms of H. irritans to obtain adults. In 2007, 15 Nellore steers (Bos indicus) (L.) were used for the collection of feces free of anthelmintic treatment and flies to produce for eggs and larva. For method I, 30 eggs were incubated in square filter paper (5 × 5 cm) and deposited on bovine feces (500 g) where they were kept until hatching and spontaneous penetration of larvae (L1) into the fecal mass. After 24 h, eggs were analyzed under a stereoscope microscope (40×) for the number of larvae that instinctively penetrated the feces. In method II, larvae were obtained only by natural egg hatching. At birth, 30 larvae were collected and individually inoculated, directly onto the fecal plate by employing a moistened brush. The tests were carried out at controlled temperature (28˚C ± 2˚C) and saturated humidity (80%) until the emergence of flies with both methods. The number of emerged flies was considered in the result. Using method I, 276 (76.7%) flies emerged from 360 inoculated eggs, while using method II, 283 (78.6%) flies emerged from 360 inoculated larvae. There was no significant difference (P = 0.7821) between methods for the number of flies; however, the proportion between males and females by means of larva inoculation was different from 1:1 (P = 0.0146). Results indicated that both methods led to a satisfactory production of flies and egg inoculation provided an easier establishment

Bispectral index in calves anesthetized with xylazine, midazolam, ketamine, isoflurane and subjected to continuous rate infusion of lidocaine

PURPOSE: To assess the bispectral index (BIS) and recovery in calves anesthetized with xylazine, midazolam, ketamine and isoflurane and subjected to CRI of lidocaine. METHODS: Xilazine was administered followed by ketamine and midazolam, orotracheal intubation and maintenance on isoflurane using mechanical ventilation. Lidocaine (2 mg kg-1 bolus) or saline (0.9%) was administered IV followed by a CRI (100 µg kg-1 minute-1) of lidocaine (L) or saline (C). Were recorded BIS, heart rate (HR), mean arterial pressure (MAP) and rectal temperature (RT) before administration of premedication (TB) and 15 minutes after (TX), before administering lidocaine (T0) and 20, 40, 60 and 80 minutes after the start of the CRI . Time do sternal recumbency (SRE) and standing (ST) and plasma lidocaine concentration also evaluated. RESULTS: In both treatments BIS decreased significantly at all times compared to TB. TX was higher than the subsequent times. HR decreased from baseline at all times and decreased from T40 in L compared to C. SRE was higher in L compared to C. CONCLUSIONS: Bispectral index values were consistent with the degree of hypnosis of the animals. Lidocaine did not potentiate isoflurane anesthesia assessed by BIS in unstimulated calves anesthetized with constant E´Iso. Lidocaine increased the time to sternal recumbency.Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP
