    Toxicity study in blood and tumor cells of laser produced medicines for application in fabrics

    Phenothiazine derivatives are non-antibiotics with antimicrobial, fungistatic and fungicidal effects. We exposed to a high energy UV laser beam phenothiazines solutions in water at 20mg/mL concentration to increase antibacterial activity of resulting mixtures. Compared to previous results obtained on bacteria, more research is needed about UV laser irradiated phenothiazines applications on cancer cell cultures to evidence possible anticancerous properties. Evaluation of the safety of the newly obtained photoproducts in view of use on humans is also needed. Due to expensive animal testing in toxicology and pressure from general public and governments to develop alternatives to in vivo testing, in vitro cell-based models are attractive for preliminary testing of new materials. Cytotoxicity screening reported here shows that laser irradiated (4h exposure time length) chlorpromazine and promazine are more efficient against some cell cultures. Interaction of laser irradiated phenothiazines with fabrics show that promethazine and chlorpromazine have improved wetting properties. Correlation of these two groups of properties shows that chlorpromazine appears to be more recommended for applications on tissues using fabrics as transport vectors. The reported results concern stability study of phenothiazines water solutions to know the time limits within which they are stable and may be used.     Hiperplazia congenitală suprarenală reprezintă o tulburare în sinteza steroizilor, cu transmitere autozomal recesivă şi cu prevalenţă mare în cazul copiilor proveniţi din relaţii de consangvinitate. Prezentăm cazul unui sugar de sex masculin, diagnosticat la vârsta de 2 luni cu sindrom de pierdere de sare şi hiperplazie congenitală suprarenală forma clasică fără semne de virilizare, alături de un review al datelor din literatura recentă legată de acest subiect. Diagnosticul a fost pus pe baza istoricului familial, a datelor clinice şi paraclinice. Prognosticul poate fi unul favorabil atât timp cât există complianţă la tratamentul de substituţie cu glucocorticoizi. Totodată, riscul de deces este ridicat prin crizele suprarenale şi dezechilibrul metabolic major

    Extracellular Electrophysiology in the Prostate Cancer Cell Model PC-3

    Although prostate cancer is one of the most common cancers in the male population, its basic biological function at a cellular level remains to be fully understood. This lack of in depth understanding of its physiology significantly hinders the development of new, targeted and more effective treatment strategies. Whilst electrophysiological studies can provide in depth analysis, the possibility of recording electrical activity in large populations of non-neuronal cells remains a significant challenge, even harder to address in the picoAmpere-range, which is typical of cellular level electrical activities. In this paper, we present the measurement and characterization of electrical activity of populations of prostate cancer cells PC-3, demonstrating for the first time a meaningful electrical pattern. The low noise system used comprises a multi-electrode array (MEA) with circular gold electrodes on silicon oxide substrates. The extracellular capacitive currents present two standard patterns: an asynchronous sporadic pattern and a synchronous quasi-periodic biphasic spike pattern. An amplitude of ±150 pA, a width between 50–300 ms and an inter-spike interval around 0.5 Hz characterize the quasi-periodic spikes. Our experiments using treatment of cells with Gd3+, known as an inhibitor for the Ca2+ exchanges, suggest that the quasi-periodic signals originate from Ca2 channels. After adding the Gd3+ to a population of living PC-3 cells, their electrical activity considerably decreased; once the culture was washed, thus eliminating the Gd3+ containing medium and addition of fresh cellular growth medium, the PC-3 cells recovered their normal electrical activity. Cellular viability plots have been carried out, demonstrating that the PC-3 cells remain viable after the use of Gd3+, on the timescale of this experiment. Hence, this experimental work suggests that Ca2+ is significantly affecting the electrophysiological communication pattern among PC-3 cell populations.     Impact of safety-related dose reductions or discontinuations on sustained virologic response in HCV-infected patients: Results from the GUARD-C Cohort

    BACKGROUND: Despite the introduction of direct-acting antiviral agents for chronic hepatitis C virus (HCV) infection, peginterferon alfa/ribavirin remains relevant in many resource-constrained settings. The non-randomized GUARD-C cohort investigated baseline predictors of safety-related dose reductions or discontinuations (sr-RD) and their impact on sustained virologic response (SVR) in patients receiving peginterferon alfa/ribavirin in routine practice. METHODS: A total of 3181 HCV-mono-infected treatment-naive patients were assigned to 24 or 48 weeks of peginterferon alfa/ribavirin by their physician. Patients were categorized by time-to-first sr-RD (Week 4/12). Detailed analyses of the impact of sr-RD on SVR24 (HCV RNA <50 IU/mL) were conducted in 951 Caucasian, noncirrhotic genotype (G)1 patients assigned to peginterferon alfa-2a/ribavirin for 48 weeks. The probability of SVR24 was identified by a baseline scoring system (range: 0-9 points) on which scores of 5 to 9 and <5 represent high and low probability of SVR24, respectively. RESULTS: SVR24 rates were 46.1% (754/1634), 77.1% (279/362), 68.0% (514/756), and 51.3% (203/396), respectively, in G1, 2, 3, and 4 patients. Overall, 16.9% and 21.8% patients experienced 651 sr-RD for peginterferon alfa and ribavirin, respectively. Among Caucasian noncirrhotic G1 patients: female sex, lower body mass index, pre-existing cardiovascular/pulmonary disease, and low hematological indices were prognostic factors of sr-RD; SVR24 was lower in patients with 651 vs. no sr-RD by Week 4 (37.9% vs. 54.4%; P = 0.0046) and Week 12 (41.7% vs. 55.3%; P = 0.0016); sr-RD by Week 4/12 significantly reduced SVR24 in patients with scores <5 but not 655. CONCLUSIONS: In conclusion, sr-RD to peginterferon alfa-2a/ribavirin significantly impacts on SVR24 rates in treatment-naive G1 noncirrhotic Caucasian patients. Baseline characteristics can help select patients with a high probability of SVR24 and a low probability of sr-RD with peginterferon alfa-2a/ribavirin

    Laser-Irradiated Chlorpromazine as a Potent Anti-Biofilm Agent for Coating of Biomedical Devices

    Nowadays, antibiotic resistance has become increasingly common, triggering a global health crisis, immediately needing alternative, including repurposed drugs with potent bactericidal effects. We demonstrated that chlorpromazine aqueous solutions exposed to laser radiation exhibited visible activity against various microorganisms. The aim of this study was to investigate the quantitative antimicrobial activity of chlorpromazine in non-irradiated and 4-h laser irradiated form. Also, we examined the effect of both solutions impregnated on a cotton patch, cannula, and urinary catheter against Gram-positive Staphylococcus aureus and Gram-negative Pseudomonas aeruginosa and Escherichia coli. In all experimental versions, the chlorpromazine antimicrobial activity was enhanced by laser exposure. Besides the experimental results, the in silico analyses using molecular docking proved that the improved antimicrobial activity of the irradiated compound was a result of the combined action of the photoproducts on the biological target (s). Our results show that laser radiation could alter the molecular structure of various drugs and their effects, proving to be a promising strategy to halt antibiotic resistance, by repurposing current medicines for new antimicrobial strategies, thereby decreasing the costs and time for the development of more efficient drugs

    Applications of TLS and ALS in Evaluating Forest Ecosystem Services: A Southern Carpathians Case Study

    Forests play an important role in biodiversity conservation, being one of the main providers of ecosystem services, according to the Economics of Ecosystems and Biodiversity. The functions and ecosystem services provided by forests are various concerning the natural capital and the socio-economic systems. Past decades of remote-sensing advances make it possible to address a large set of variables, including both biophysical parameters and ecological indicators, that characterize forest ecosystems and their capacity to supply services. This research aims to identify and implement existing methods that can be used for evaluating ecosystem services by employing airborne and terrestrial stationary laser scanning on plots from the Southern Carpathian mountains. Moreover, this paper discusses the adaptation of field-based approaches for evaluating ecological indicators to automated processing techniques based on airborne and terrestrial stationary laser scanning (ALS and TLS). Forest ecosystem functions, such as provisioning, regulation, and support, and the overall forest condition were assessed through the measurement and analysis of stand-based biomass characteristics (e.g., trees’ heights, wood volume), horizontal structure indices (e.g., canopy cover), and recruitment-mortality processes as well as overall health status assessment (e.g., dead trees identification, deadwood volume). The paper, through the implementation of the above-mentioned analyses, facilitates the development of a complex multi-source monitoring approach as a potential solution for assessing ecosystem services provided by the forest, as well as a basis for further monetization approaches
