    Shiga toxin-producing Escherichia coli: A single-center, 11-year pediatric experience

    The aim of this study was to identify the best practices for the detection of Shiga toxin-producing Escherichia coli (STEC) in children with diarrheal illness treated at a tertiary care center, i.e., sorbitol-MacConkey (SMAC) agar culture, enzyme immunoassay (EIA) for Shiga toxin, or the simultaneous use of both methods. STEC was detected in 100 of 14,997 stool specimens submitted for enteric culture (0.7%), with 65 cases of E. coli O157. Among E. coli O157 isolates, 57 (88%) were identified by both SMAC agar culture and EIA, 6 (9%) by SMAC agar culture alone, and 2 (3%) by EIA alone. Of the 62 individuals with diarrheal hemolytic uremic syndrome (HUS) seen at our institution during the study period, 16 (26%) had STEC isolated from cultures at our institution and 15 (24%) had STEC isolated at other institutions. No STEC was recovered in 31 cases (50%). Of the HUS cases in which STEC was isolated, 28 (90%) were attributable to E. coli O157 and 3 (10%) were attributable to non-O157 STEC. Consistent with previous studies, we have determined that a subset of E. coli O157 infections will not be detected if an agar-based method is excluded from the enteric culture workup; this has both clinical and public health implications. The best practice would be concomitant use of an agar-based method and a Shiga toxin EIA, but a Shiga toxin EIA should not be considered to be an adequate stand-alone test for detection of E. coli O157 in clinical samples

    A Dynamical Analysis of the Dust Tail of Comet C/1995 O1 (Hale-Bopp) at High Heliocentric Distances

    Comet C/1995 O1 (Hale-Bopp) has provided an unprecedented opportunity to observe a bright comet over a wide range of heliocentric distances. We present here Spitzer Space Telescope observations of Hale-Bopp from 2005 and 2008 that show a distinct coma and tail, the presence of which is uncommon given its large heliocentric distance (21.6 AU and 27.2 AU, respectively). The morphology of the dust is compared to dynamical models to understand the activity of the comet. Our analysis shows that the shape of Hale-Bopp's dust tail in these images cannot be explained using the usual Finson-Probstein (solar gravity + solar radiation pressure) dynamical model. Several alternative explanations are explored. The analysis suggests that the most likely cause of the discrepancy is that the dust is being charged by the solar wind, then being affected by the interplanetary magnetic field via the Lorentz force. Though this effect has been explored previously, if correct, this seems to be the first time that the Lorentz force has been required to model a cometary dust tail. The analysis also suggests that Hale-Bopp was actively emitting particles when these images were taken, and the tail characteristics changed between observations.Comment: 23 pages, 10 figures.     Partially Mixed Selectivity in Human Posterior Parietal Association Cortex

    To clarify the organization of motor representations in posterior parietal cortex, we test how three motor variables (body side, body part, cognitive strategy) are coded in the human anterior intraparietal cortex. All tested movements were encoded, arguing against strict anatomical segregation of effectors. Single units coded for diverse conjunctions of variables, with different dimensions anatomically overlapping. Consistent with recent studies, neurons encoding body parts exhibited mixed selectivity. This mixed selectivity resulted in largely orthogonal coding of body parts, which “functionally segregate” the effector responses despite the high degree of anatomical overlap. Body side and strategy were not coded in a mixed manner as effector determined their organization. Mixed coding of some variables over others, what we term “partially mixed coding,” argues that the type of functional encoding depends on the compared dimensions. This structure is advantageous for neuroprosthetics, allowing a single array to decode movements of a large extent of the body

    Transdisciplinarity Within the Academic Engineering Literature

    Despite increased discourse around transdisciplinary (TD) research, there is a perception it has received less attention within engineering. This is significant if, as generally accepted, TD increases the societal value of research. This paper benchmarks TD engineering research against the broader TD literature, addressing the question: How do the characteristics of the academic engineering TD literature compare to the TD academic literature in general? We analyse the chronology, journals, and text of papers referencing TD within their abstract and compare this to papers that fall within the engineering subject area. The conclusions find that TD research is limited generally, and within engineering specifically. Historically, TD research focuses on sustainability challenges, a persistent trend within the general literature. Within engineering research, the focus of TD is wider and addresses operational and “grand challenge” problems. TD remains poorly defined and future work should focus on clarifying meaning within the engineering discipline

    PER1 rs3027172 genotype interacts with early life stress to predict problematic alcohol use, but not reward-related ventral striatum activity

    Increasing evidence suggests that the circadian and stress regulatory systems contribute to alcohol use disorder (AUD) risk, which may partially arise through effects on reward-related neural function. The C allele of the PER1 rs3027172 single nucleotide polymorphism reduces PER1 expression in cells incubated with cortisol and has been associated with increased risk for adult AUD and problematic drinking among adolescents exposed to high levels of familial psychosocial adversity. Using data from undergraduate students who completed the ongoing Duke Neurogenetics Study (n=665), we tested whether exposure to early life stress (ELS; Childhood Trauma Questionnaire) moderates the association between rs3027172 genotype and later problematic alcohol use (Alcohol Use Disorders Identification Test) as well as ventral striatum (VS) reactivity to reward (card-guessing task while functional magnetic resonance imaging data were acquired). Initial analyses found that PER1 rs3027172 genotype interacted with ELS to predict both problematic drinking and VS reactivity; minor C allele carriers, who were also exposed to elevated ELS reported greater problematic drinking and exhibited greater ventral striatum reactivity to reward-related stimuli. When gene x covariate and environment x covariate interactions were controlled for, the interaction predicting problematic alcohol use remained significant (p<0.05, corrected) while the interaction predicting VS reactivity was no longer significant. These results extend our understanding of relationships between PER1 genotype, early life stress, and problematic alcohol use, and serve as a cautionary tale on the importance of controlling for potential confounders in studies of moderation including gene x environment interactions
