    An immunogenic cell death-related gene expression signature in predicting prognosis of pancreatic ductal adenocarcinoma

    Abstract Background Immunogenic cell death (ICD) has been identified as regulated cell death, which is sufficient to activate the adaptive immune response. This study aimed to research ICD-related genes and create a gene model to predict pancreatic ductal adenocarcinoma (PAAD) patients’ prognosis. Methods The RNA sequencing and clinical data were downloaded from the TGCA and GEO databases. The PAAD samples were classified into two subtypes based on the expression levels of ICD-related genes using consensus clustering. Based on the differentially expressed genes (DEGs), a prognostic scoring model was constructed using LASSO regression and Cox regression, and the scoring model was used to predict the prognosis of PAAD patients. Moreover, colony formation assay was performed to confirm the prognostic value of those genes. Results We identified two ICD cluster by consensus clustering, and found that the the ICD-high group was closely associated with immune-hot phenotype, favorable clinical outcomes. We established an ICD-related prognostic model which can predict the prognosis of pancreatic ductal adenocarcinoma. Moreover, depletion of NT5E, ATG5, FOXP3, and IFNG inhibited the colony formation ability of pancreatic cancer cell. Conclusion We identified a novel classification for PAAD based on the expression of ICD-related genes, which may provide a potential strategy for therapeutics against PAAD

    Superior Communication of Positive Emotions Through Nonverbal Vocalisations Compared to Speech Prosody

    The human voice communicates emotion through two different types of vocalizations: nonverbal vocalizations (brief non-linguistic sounds like laughs) and speech prosody (tone of voice). Research examining recognizability of emotions from the voice has mostly focused on either nonverbal vocalizations or speech prosody, and included few categories of positive emotions. In two preregistered experiments, we compare human listeners' (total n = 400) recognition performance for 22 positive emotions from nonverbal vocalizations (n = 880) to that from speech prosody (n = 880). The results show that listeners were more accurate in recognizing most positive emotions from nonverbal vocalizations compared to prosodic expressions. Furthermore, acoustic classification experiments with machine learning models demonstrated that positive emotions are expressed with more distinctive acoustic patterns for nonverbal vocalizations as compared to speech prosody. Overall, the results suggest that vocal expressions of positive emotions are communicated more successfully when expressed as nonverbal vocalizations compared to speech prosody.ISSN:0191-5886ISSN:1573-365

    Healthspan Improvements in Caenorhabditis elegans with Traditional Chinese Herbal Tea

    Searching for natural and safe herbal tea with health benefits has attracted more and more attention, which is of great significance for reducing disease risk. A Chinese traditional herbal tea (HT) is rich in active ingredients extracted from natural plants. Numerous pharmacological studies showed that HT had the potential to improve health, including antidepression and antioxidant effects. In this study, we proposed a strategy to explore the role and underlying mechanism of HT in improving healthspan of a Caenorhabditis elegans model. First, we found that HT significantly prolonged the lifespan without reducing fertility in worms. Second, stress resistance (oxidative stress and heat stress) was enhanced and Aβ- and polyQ-induced toxicity was relieved significantly by HT treatment. Both fat deposition and age pigment accumulation were found to be significantly reduced in HT-treated worms. The locomotion in mid-late stages was improved, indicating that behavioral mobility was also significantly enhanced. Furthermore, the main components of HT were eighteen polyphenols and two terpenoids. Finally, it was found that this protective mechanism was positively correlated with the insulin/insulin-like growth factor signaling- (IIS-) dependent manner, which went through promoting the nuclear localization of DAF-16 and its downstream SOD-3 expression. These results suggested that HT had an important role in improving health, which might serve as a promising healthy tea

    Regional Homogeneity Abnormalities in Early-Onset and Adolescent-Onset Conduct Disorder in Boys: A Resting-State fMRI Study

    Purpose: Developmental taxonomic theory posits that formation of early-onset conduct disorder (EO-CD), is considered to have a neurodevelopmental etiology and have more severe psychosocial and neuropsychological dysfunction than adolescent-onset CD (AO-CD), which is thought to stem largely from social mimicry of deviant peers. The purpose of the current study was to investigate whether regional homogeneity (ReHo), denoting the spontaneous brain activity, supports developmental taxonomic theory in a resting state (rs).Materials and Methods: Rs-functional magnetic resonance imaging (fMRI) examinations were administered to 36 EO-CD patients, 32 AO-CD patients, and 30 healthy controls (HCs). All participants were male adolescents, aged between 12 and 17 years old. A one-way analysis of covariance (ANCOVA), with age and IQ as covariates, was performed to identify regions with significant group differences in ReHo values, followed by a post hoc analyses.Results: Compared with the AO-CD groups, EO-CD had higher ReHo values in the right middle/inferior frontal gyrus. Compared with the HCs, the EO-CD group exhibited lower ReHo values in the left precuneus, left middle occipital gyrus, left cerebellum posterior lobe and the right inferior parietal lobule, as well as higher ReHo values in the right middle frontal gyrus, left insula/inferior frontal gyrus, right postcentral gyrus, and the left anterior cingulate gyrus. Compared with the HCs, the AO-CD group showed lower ReHo values in the bilateral precuneus, left cerebellum posterior lobe, and the right inferior parietal lobule.Conclusion: Significant differences in ReHo were observed between the EO-CD and AO-CD groups, implying distinct neuropathological mechanisms of the two CD subtypes, consistent with developmental taxonomic theory. CD-associated abnormalities in ReHo may be related to high-order cognitive and low-level perceptual system impairments in CD
