    Forecast UTI: aplicativo para previsĂŁo de leitos de unidades de terapia intensiva no contexto da pandemia de covid-19

    In view of the need to manage and forecast the number of beds in the Intensive Care Units for critically ill patients in Covid-19, the Forecast UTI was developed: an open access application that allows the monitoring of hospital indicators based on historical data from the service health and the temporal dynamics of the epidemic. It is also possible to make short-term forecasts of the number of beds occupied daily by the disease and to establish possible care scenarios. This article presents the functions, mode of access and examples of use of Forecast UTI, a computational tool capable of assisting managers of public and private hospitals in the Unified Health System, since they support decision-making quickly, strategically and efficiently.Frente à necessidade de gerenciamento e previsão do número de leitos de unidades de terapia intensiva (UTI) para pacientes graves de covid-19, foi desenvolvido o Forecast UTI, um aplicativo de livre acesso e que permite o monitoramento de indicadores hospitalares com base em dados históricos do serviço de saúde e na dinâmica temporal dessa epidemia por coronavírus. O Forecast UTI também possibilita realizar previsões de curto prazo sobre número de leitos ocupados pela doença diariamente, e estabelecer possíveis cenários de atendimento. Este artigo apresenta as funções, modo de acesso e exemplos de uso do Forecast UTI, uma ferramenta computacional destinada a auxiliar gestores de hospitais da rede pública e privada do Sistema Único de Saúde no subsídio à tomada de decisão, de forma rápida, estratégica e eficiente.

    Transcriptional profiling of a fungal granuloma reveals a low metabolic activity of Paracoccidioides brasiliensis yeasts and an actively regulated host immune response

    Granulomas are important immunological structures in the host defense against the fungus Paracoccidioides brasiliensis, the main etiologic agent of Paracoccidioidomycosis (PCM), a granulomatous systemic mycosis endemic in Latin America. We have performed transcriptional and proteomic studies of yeasts present in the pulmonary granulomas of PCM aiming to identify relevant genes and proteins that act under stressing conditions. C57BL/6 mice were infected with 1x106 yeasts and after 8- and 12-weeks of infection, granulomatous lesions were obtained for extraction of fungal and murine RNAs and fungal proteins. Dual transcriptional profiling was done comparing lung cells and P. brasiliensis yeasts from granulomas with uninfected lung cells and the original yeast suspension used in the infection, respectively. Mouse transcripts indicated a lung malfunction, with low expression of genes related to muscle contraction and organization. In addition, an increased expression of transcripts related to the activity of neutrophils, eosinophils, macrophages, lymphocytes as well as an elevated expression of IL-1β, TNF-α, IFN-γ, IL-17 transcripts were observed. The increased expression of transcripts for CTLA-4, PD-1 and arginase-1, provided evidence of immune regulatory mechanisms within the granulomatous lesions. Also, our results indicate iron as a key element for the granuloma to function, where a high number of transcripts related to fungal siderophores for iron uptake was observed, a mechanism of fungal virulence not previously described in granulomas. Furthermore, transcriptomics and proteomics analyzes indicated a low fungal activity within the granuloma, as demonstrated by the decreased expression of genes and proteins related to energy metabolism and cell cycle

    Calreticulin expression levels and endoplasmic reticulum during late oogenesis and early embryogenesis of rhodnius prolixus stahl

    11 p. : il.This study reports the cloning, expression analysis and localization of calreticulin (CRT) in the endoplasmic reticulum (ER) during late oogenesis and early embryogenesis of the insect Rhodnius prolixus. CRT was cloned and sequenced from cDNA extracted from unfertilized eggs. Real-time PCR showed that CRT expression remains at lower levels during late oogenesis when compared to vitellogenic oocytes or day 0 laid fertilized eggs. Immunofluorescence microscopy showed that this protein is located in the periphery of the egg, in a differential peripheral ooplasm surrounding the yolk-rich internal ooplasm, only identified by transmission electron microscopy (TEM) of thin sections. Using immunogold electron microscopy, the ER ultrastructure (CRT labeled) was identified in the peripheral ooplasm as dispersed lamellae, randomly distributed in the peripheral ooplasm. No massive alterations of ER ultrastructure were found before or right after (30 min) fertilization, but an increase in CRT expression levels and assembly of typical rough ER (parallel cisternae with associated ribosomes) were observed 18–24 h after oviposition. The lack of ER assembly at fertilization and the later formation of rough ER together with the increase in CRT expression levels, suggest that the major functions of ER might be of great importance during the early events of development. The possible involvement of ER in the early steps of embryogenesis will be discussed
