    A Comparative Transcriptome and Proteome Analysis in Rat Models Reveals Effects of Aging and Diabetes on Expression of Neuronal Genes

    SummaryBackgroundTo understand neuronal molecular changes in senile diabetes we established a rat senile diabetes model and analyzed transcriptome and proteome changes.MethodsWistar rats were fed a high sugar, high fat diet for 16 months to induce diabetes. Non-diabetic aged rats and young rats were used as controls. Transcript and protein levels in the liver were then analyzed by microarray and antibody arrays, respectively.ResultsNeuronal genes that were differentially expressed between senile diabetic rats, non-diabetic aged rats, and young rats were distributed across 12 pathways and 23 Gene Ontology (GO) clusters. Among them, 2267 genes were aging-related, 1230 genes were diabetes-associated, and 9 proteins might be associated with neurological disorders.ConclusionIn this study, we investigated transcriptome and proteome changes in animal models, analyzed the impact of aging and diabetes on neuronal molecules, and confirmed the correlations. Our study provides support for further studies on mechanisms of neuronal diseases

    Aurora-A contributes to cisplatin resistance and lymphatic metastasis in non-small cell lung cancer and predicts poor prognosis

    BACKGROUND: Platinum-based chemotherapy improves survival among patients with non-small cell lung cancer (NSCLC), but the efficiency is limited due to resistance. In this study, we aimed to identify the expression of Aurora-A and its correlation with cisplatin resistance and prognosis in NSCLC. METHODS: We used immunohistochemical analysis to determine the expression of Aurora-A protein in 102 NSCLC patients treated by surgery and adjuvant cisplatin-based chemotherapy. The prognostic significances were assessed by Kaplan-Meier survival estimates and Cox models. The potential role of Aurora-A in the regulation of cisplatin resistance in NSCLC cells was examined by transfections using expression vector and small interfering RNA or using small-molecule inhibitors. RESULTS: Aurora-A expression was significantly associated with clinical stage (p = 0.018), lymph node metastasis (p = 0.038) and recurrence (p = 0.005), and was an independent prognostic parameter in multivariate analysis. High level of Aurora-A expression predicted poorer overall survival (OS) and progression-free survival (PFS). In vitro data showed that Aurora-A expression was elevated in cisplatin-resistant lung cancer cells, and overexpression or knockdown of Aurora-A resulted in increased or decreased cellular resistance to cisplatin. Furthermore, inhibition of Aurora-A reversed the migration ability of cisplatin-resistant cells. CONCLUSIONS: The current findings suggest that high Aurora-A expression is correlated with cisplatin-based chemotherapeutic resistance and predicts poor patient survival in NSCLC. Aurora-A might serve as a predictive biomarker of drug response and therapeutic target to reverse chemotherapy resistance

    Magnetic monopole induced polarons in atomic superlattices

    Magnetic monopoles have been realized as emergent quasiparticles in both condensed matter and ultracold atomic platforms, with growing interests in the coupling effects between the monopole and different magnetic quasiparticles. In this work, interaction effects between monopoles and magnons are investigated for an atomic pseudospin chain. We reveal that the monopole can excite a virtual magnon cloud in the paramagnetic chain, thereby giving rise to a new type of polaron, the monopole-cored polaron (McP). The McP is composed of the monopole as the impurity core and the virtual magnon excitation as the dressing cloud. The magnon dressing facilitates the Dirac string excitation and impacts the monopole hopping. This induces an anti-trapping effect of the McP, which refers to the fact that the dressing enhances the mobility of the McP, in contrast to the self-trapping of the common polarons. Moreover, heterogeneous bipolarons are shown to exist under the simultaneous doping of a north and a south monopole. The heterogeneous bipolaron possesses an inner degree of freedom composed of two identical impurities.     Antibacterial Bisabolane-Type Sesquiterpenoids from the Sponge-Derived Fungus Aspergillus sp.

    Four new bisabolane-type sesquiterpenoids, aspergiterpenoid A (1), (−)-sydonol (2), (−)-sydonic acid (3), and (−)-5-(hydroxymethyl)-2-(2′,6′,6′-trimethyltetrahydro-2H- pyran-2-yl)phenol (4) together with one known fungal metabolite (5) were isolated from the fermentation broth of a marine-derived fungus Aspergillus sp., which was isolated from the sponge Xestospongia testudinaria collected from the South China Sea. Four of them (1–4) are optically active compounds. Their structures and absolute configurations were elucidated by using NMR spectroscopic techniques and mass spectrometric analysis, and by comparing their optical rotations with those related known analogues. Compounds 1–5 showed selective antibacterial activity against eight bacterial strains with the MIC (minimum inhibiting concentrations) values between 1.25 and 20.0 µM. The cytotoxic, antifouling, and acetylcholinesterase inhibitory activities of these compounds were also examined

    The First Data Release of the Beijing-Arizona Sky Survey

    The Beijing-Arizona Sky Survey (BASS) is a new wide-field legacy imaging survey in the northern Galactic cap using the 2.3m Bok telescope. The survey will cover about 5400 deg2^2 in the gg and rr bands, and the expected 5σ\sigma depths (corrected for the Galactic extinction) in the two bands are 24.0 and 23.4 mag, respectively. BASS started observations in January 2015, and has completed about 41% of the whole area as of July 2016. The first data release contains both calibrated images and photometric catalogs obtained in 2015 and 2016. The depths of single-epoch images in the two bands are 23.4 and 22.9 mag, and the full depths of three epochs are about 24.1 and 23.5 mag, respectively.Comment: 16 pages, published by A

    Detection of quantitative trait loci affecting haematological traits in swine via genome scanning

    <p>Abstract</p> <p>Background</p> <p>Haematological traits, which consist of mainly three components: leukocyte traits, erythrocyte traits and platelet traits, play extremely important role in animal immune function and disease resistance. But knowledge of the genetic background controlling variability of these traits is very limited, especially in swine.</p> <p>Results</p> <p>In the present study, 18 haematological traits (7 leukocyte traits, 7 erythrocyte traits and 4 platelet traits) were measured in a pig resource population consisting of 368 purebred piglets of three breeds (Landrace, Large White and Songliao Black Pig), after inoculation with the swine fever vaccine when the pigs were 21 days old. A whole-genome scan of QTL for these traits was performed using 206 microsatellite markers covering all 18 autosomes and the X chromosome. Using variance component analysis based on a linear mixed model and the false discovery rate (FDR) test, 35 QTL with FDR < 0.10 were identified: 3 for the leukocyte traits, 28 for the erythrocyte traits, and 4 for the platelet traits. Of the 35 QTL, 25 were significant at <it>FDR </it>< 0.05 level, including 9 significant at <it>FDR </it>< 0.01 level.</p> <p>Conclusions</p> <p>Very few QTL were previously identified for hematological traits of pigs and never in purebred populations. Most of the QTL detected here, in particular the QTL for the platelet traits, have not been reported before. Our results lay important foundation for identifying the causal genes underlying the hematological trait variations in pigs.</p

    Sexually dimorphic distribution of kiss1 and kiss2 in the brain of yellowtail clownfish, Amphiprion clarkii

    Kisspeptin system was shown to be a key factor in mediating social stress and reproduction. Yellowtail clownfish, Amphiprion clarkii, is a hermaphrodite fish, whose sex determination and gonadal development are affected by the social status of individuals. The yellowtail clownfish is a fantastic animal model to explore sex determination, but the social status and precise distribution of kiss mRNAs in the brain of this species are unknown. Hererin, a novel in situ hybridization technique, RNAscope, was used to investigate the distribution of kiss1 and kiss2 expressions in the brain of yellowtail clownfish. The coronal planes of brain showed that the kiss1 signal was mainly present in dorsal habenular nucleus (NHd) and kiss2 mRNA was widely expressed in telencephalon, midbrain, and hypothalamus, especially in dorsal part of the nucleus of the lateral recess (NRLd). Additionally, kiss1 and kiss2 signals have sexually dimorphic distribution. The kiss1 mRNA was distributed in NHd, the telencephalon, and lateral part of the diffuse nucleus of the inferior lobe (NDLIl) of females but in NHd and NDLIl of males. kiss2 signals were stronger in females than that in males. The distribution of kiss1 and kiss2 neurons in NHd of habenula and NRLd of hypothalamus may suggest that kiss genes associate environmental signaling and reproductive function in yellowtail clownfish

    Expression of Human Leukocyte Antigen G is associated with Prognosis in Nasopharyngeal Carcinoma

    Human leukocyte antigen G (HLA-G) has multiple immune regulatory functions including the induction of immune tolerance in malignancies. The roles of HLA-G have not been investigated in nasopharyngeal carcinoma (NPC). This study is aimed to evaluate the role of HLA-G as prognostic factor for NPC patients as well as its role in the immune regulation. Western assays showed high HLA-G expression in NPC cell lines, but low in the immortalized nasopharyngeal epithelial cell line NP69. HLA-G protein was further detected in 79.2% of 552 NPC specimens with immunohistochemistry (IHC), but not in normal nasopharyngeal epithelium tissue. Moreover, high expression of HLA-G predicted poor survival of NPC patients and positively correlated with tumor N classification and recurrence or metastasis. Multivariate analysis indicated that HLA-G was an independent and unfavorable prognostic factor. Furthermore, the presence of CD68+macrophages and IL-10 were also examined, which are two prognostic markers of NPC and important factors for regulating immune surveillance. The correlations of HLA-G with these two immune factors were revealed in NPC tissues. Taken together, our results suggest that HLA-G is an independent biomarker for NPC prognosis, and HLA-G might contribute to NPC progression, which might jointly regulate immune surveillance in NPC together with macrophages and IL-10
