    Serotonin-mediated modulation of Na+/K+ pump current in rat hippocampal CA1 pyramidal neurons

    <p>Abstract</p> <p>Background</p> <p>The aim of this study was to investigate whether serotonin (5-hydroxytryptamine, 5-HT) can modulate Na<sup>+</sup>/K<sup>+ </sup>pump in rat hippocampal CA1 pyramidal neurons.</p> <p>Results</p> <p>5-HT (0.1, 1 mM) showed Na<sup>+</sup>/K<sup>+ </sup>pump current (Ip) densities of 0.40 ± 0.04, 0.34 ± 0.03 pA/pF contrast to 0.63 ± 0.04 pA/pF of the control of 0.5 mM strophanthidin (Str), demonstrating 5-HT-induced inhibition of Ip in a dose-dependent manner in hippocampal CA1 pyramidal neurons. The effect was partly attenuated by ondasetron, a 5-HT<sub>3 </sub>receptor (5-HT<sub>3</sub>R) antagonist, not by WAY100635, a 5-HT<sub>1A</sub>R antagonist, while 1-(3-Chlorophenyl) biguanide hydrochloride (m-CPBG), a 5-HT<sub>3</sub>R specific agonist, mimicked the effect of 5-HT on Ip.</p> <p>Conclusion</p> <p>5-HT inhibits neuronal Na<sup>+</sup>/K<sup>+ </sup>pump activity via 5-HT<sub>3</sub>R in rat hippocampal CA1 pyramidal neurons. This discloses novel mechanisms for the function of 5-HT in learning and memory, which may be a useful target to benefit these patients with cognitive disorder.</p

    Datamator : an authoring tool for creating datamations via data query decomposition

    Datamation is designed to animate an analysis pipeline step by step, serving as an intuitive and efficient method for interpreting data analysis outcomes and facilitating easy sharing with others. However, the creation of a datamation is a difficult task that demands expertise in diverse skills. To simplify this task, we introduce Datamator, a language-oriented authoring tool developed to support datamation generation. In this system, we develop a data query analyzer that enables users to generate an initial datamation effortlessly by inputting a data question in natural language. Then, the datamation is displayed in an interactive editor that affords users the ability to both edit the analysis progression and delve into the specifics of each step undertaken. Notably, the Datamator incorporates a novel calibration network that is able to optimize the outputs of the query decomposition network using a small amount of user feedback. We have previously established a robust serial dilution protocol for calibration of plate reader measurements of absorbance to estimated bacterial cell count and for green fluorescence from proteins expressed in bacterial cells to molecules of equivalent fluorescein. We now extend these protocols to calibration of red fluorescence to the sulforhodamine-101 fluorescent dye and blue fluorescence to Cascade Blue.     Integrative epigenome profiling of 47XXY provides insights into whole genomic DNA hypermethylation and active chromatin accessibility

    Klinefelter syndrome (KS, 47XXY) is a disorder characterized by sex chromosomal aneuploidy, which may lead to changes in epigenetic regulations of gene expression. To define epigenetic architectures in 47XXY, we annotated DNA methylation in euploid males (46XY) and females (46XX), and 47XXY individuals using whole genome bisulfite sequencing (WGBS) and integrated chromatin accessbilty, and detected abnormal hypermethylation in 47XXY. Furthermore, we detected altered chromatin accessibility in 47XXY, in particular in chromosome X, using Assay for Transposase-Accessible Chromatin sequencing (ATAC-seq) in cultured amniotic cells. Our results construct the whole genome-wide DNA methylation map in 47XXY, and provide new insights into the early epigenomic dysregulation resulting from an extra chromosome X in 47XXY

    Vector competence evaluation of mosquitoes for Tahyna virus PJ01 strain, a new Orthobunyavirus in China

    IntroductionTahyna virus (TAHV), an arbovirus of the genus Orthobunyavirus, is a cause of human diseases and less studied worldwide. In this study, a new strain of TAHV was isolated from Aedes sp. mosquitoes collected in Panjin city, Liaoning province. However, the competent vector of TAHV in China is still unknown.MethodsThe genome of newly isolated TAHV was sequenced and phylogenetic analysis is performed. Aedes albopictus and Culex pipiens pallens were orally infected with artificial virus blood meals (1:1 of virus suspension and mouse blood), the virus was detected in the midgut, ovary, salivary gland and saliva of the mosquitoes. Then, the transmission and dissemination rates, vertical transmission and horizontal transmission of the virus by the mosquitoes were assessed.ResultsPhylogenetic analysis revealed that the virus shared high similarity with TAHV and was named the TAHV PJ01 strain. After oral infection with virus blood meals, Ae. albopictus showed positive for the virus in all tested tissues with an extrinsic incubation period of 2 days and a fluctuating increasement of transmission and dissemination rates. Whereas no virus was detected in the saliva of Cx. pipiens pallens. Suckling mice bitten by infectious Ae. albopictus developed obvious neurological symptoms, including inactivity, hind-leg paralysis and difficulty turning over, when the virus titer reached 1.70×105 PFU/mL in the brain. Moreover, TAHV was detected in the eggs, larvae and adults of F1 offspring of Ae. albopictus.DiscussionAe. albopictus is an efficient vector to transmit TAHV but Cx. pipiens pallens is not. Ae. albopictus is also a reservoir host that transmits the virus vertically, which further increases the risk of outbreaks. This study has important epidemiological implications for the surveillance of pathogenic viruses in China and guiding comprehensive vector control strategies to counteract potential outbreaks in future

    Repetitive transcranial magnetic stimulation of the dorsolateral prefrontal cortex modulates electroencephalographic functional connectivity in Alzheimer’s disease

    Background: Increasing evidence demonstrates that repetitive transcranial magnetic stimulation (rTMS) treatment of the dorsolateral prefrontal cortex is beneficial for improving cognitive function in patients with Alzheimer’s disease (AD); however, the underlying mechanism of its therapeutic effect remains unclear. Objectives/Hypothesis: The aim of this study was to investigate the impact of rTMS to the dorsolateral prefrontal cortex on functional connectivity along with treatment response in AD patients with different severity of cognitive impairment. Methods: We conducted a 2-week treatment course of 10-Hz rTMS over the left dorsolateral prefrontal cortex in 23 patients with AD who were split into the mild or moderate cognitive impairment subgroup. Resting state electroencephalography and general cognition was assessed before and after rTMS. Power envelope connectivity was used to calculate functional connectivity at the source level. The functional connectivity of AD patients and 11 cognitively normal individuals was compared. Results: Power envelope connectivity was higher in the delta and theta bands but lower in the beta band in the moderate cognitive impairment group, compared to the cognitively normal controls, at baseline (p < 0.05). The mild cognitive impairment group had no significant abnormities. Montreal Cognitive Assessment scores improved after rTMS in the moderate and mild cognitive impairment groups. Power envelope connectivity in the beta band post-rTMS was increased in the moderate group (p < 0.05) but not in the mild group. No significant changes in the delta and theta band were found after rTMS in both the moderate and mild group. Conclusion: High-frequency rTMS to the dorsolateral prefrontal cortex modulates electroencephalographic functional connectivity while improving cognitive function in patients with AD.     Cosmological evolution of interacting phantom (quintessence) model in Loop Quantum Gravity

    The dynamics of interacting dark energy model in loop quantum cosmology (LQC) is studied in this paper. The dark energy has a constant equation of state wxw_x and interacts with dark matter through a form 3cH(ρx+ρm)3cH(\rho_x+\rho_m). We find for quintessence model (wx>1w_x>-1) the cosmological evolution in LQC is the same as that in classical Einstein cosmology; whereas for phantom dark energy (wx<1w_x<-1), although there are the same critical points in LQC and classical Einstein cosmology, loop quantum effect reduces significantly the parameter spacetime (c,wxc, w_x) required by stability. If parameters cc and wxw_x satisfy the conditions that the critical points are existent and stable, the universe will enter an era dominated by dark energy and dark matter with a constant energy ratio between them, and accelerate forever; otherwise it will enter an oscillatory regime. Comparing our results with the observations we find at 1σ1\sigma confidence level the universe will accelerate forever.Comment: 15 pages, 8 figures, to appear in JCA
