    La frontière Lampedusa. Mises en intrigue du sécuritaire et de l’humanitaire

    Cet article montre que l’île de Lampedusa est un théâtre sur lequel est donné le « spectacle de la frontière ». Sont présentés les acteurs et les spectateurs de ce spectacle, selon leurs places et rôles respectifs dans l’architecture de l’espace scénique et dans la dramaturgie de la représentation. On analyse les cinq actes du spectacle, selon les périodes durant lesquelles ils se déroulent et selon les événements qui les marquent. Chacun des actes est étudié en fonction des rhétoriques qui le caractérisent. La guerre aux migrations irrégulières se fonde et se justifie en recourant à deux registres discursifs différents : le registre sécuritaire et le registre humanitaire. Sur le théâtre de Lampedusa, les deux récits dominent la scène en alternance, mais sont en réalite toujours concomittants, étroitement entremêlés. Ensemble, ils contribuent à former et à renforcer les politiques et les pratiques de contrôle de l’immigration et des frontières.This paper presents the island of Lampedusa as the theatre stage on which the “border play” of immigration control is performed. The paper first introduces the performers and spectators of the play, outlining their roles and places with respect to the architecture of the theatre space as well as the dramaturgy of the play. Next, the paper analyses the five acts of the play, notably examining the time period in which each of them transpires and the most marking or spectacular events. Each act is analysed with regard to its dominant narratives. The war against irregular migration is waged and justified in resorting to two different narratives: one being security, and the other humanitarian. On the Lampedusa stage, while both narratives take turns commanding the scene, they both are in fact always present. The two rhetorics are intertwined with one another, and together they contribute to constituting and strengthening the policies and practices of migration and border control

    Loin de marquer la fin des frontières, l’ère de la mondialisation met plutôt en lumière le processus continu de redéfinition de leurs formes et de leurs modalités opératoires. Cela est particulièrement visible dans le champ des contrôles de l’immigration. Le pouvoir territorial y opère à travers une double modalité. D’un côté, il agit directement sur ses frontières territoriales, en les diversifiant et en délocalisant l’action dans l’espace : les points constitutifs de la ligne de frontière sont clonés, multipliés et projetés en deçà et au-delà de la ligne même, en produisant un effet de flexibilisation introvertie ou extravertie de la frontière. La frontière peut ainsi passer de frontière fixe à frontière mobile, de matérielle à immatérielle, de linéaire à punctiforme ou zonale. D’un autre côté, le pouvoir territorial investit la sphère des frontières représentées par les statuts, par les conditions personnelles de migrants, en mettant en relief non seulement les différents statuts déjà existants, mais en les multipliant, en en remodelant les contours et en en différenciant les contenus.Far from marking the end of borders, the globalization era enlightens us actually more about the continuous process by which borders are redefined in their shape and in the operational modalities. This is most obvious in the field of immigration control where territorial power operates through a double modality. On the one hand, it has a direct effect on territorial borders, diversifying and relocating actions in space: the constitutive points of the borderline are duplicated, multiplicated and projected bellow and beyond the line itself, thus producing an effect of either introvert or extravert flexibilisation of the border. The border is thus transformed from a fixed one to a mobile one, from a material one to an immaterial one and from a linear one to a zonal one. On the other hand, territorial power vests itself in the sphere of the borders, borders represented by the status and personal conditions of migrants, by not only delineating the different status already existing but by multiplying them through the remodelling of their outlines and the differentiation of their contents

    Rapporto tecnico sulla ottimizzazione del processo di biosintesi di una Green Fluorescent Protein ricombinante estratta da A. sulcata

    L’espressione di geni eterologhi in Escherichia coli rappresenta uno dei metodi più veloci, semplici ed economici per la produzione di ampie quantità di proteine target. Tuttavia, meccanismi di folding e le modifiche post traduzionali inducono a volte un non corretto ripiegamento delle proteine nella conformazione nativa, con successiva aggregazione in quelli che vengono definiti corpi di inclusione. Nel nostro caso, l’attenzione è stata focalizzata su una Green Fluorescent Protein (GFP) di 228 aa estratta da Anemonia sulcata, contenente un fluoroforo composto da tre amminoacidi Gln63, Tyr64, Gly65 all'interno di una struttura a barile. Il corretto folding della proteina era correlato strettamente alla funzionalità del fluoroforo. Il nostro obiettivo è stato quello, quindi, di ottimizzare il processo di biosintesi della GFP espressa in E. coli, ovviando alla formazione di corpi di inclusione contenenti la proteina (non funzionale), definendo e standardizzando inoltre, le condizioni che consentivano di produrre la più alta percentuale di GFP correttamente ripiegata (in condizioni non denaturanti) e quindi funzionale

    Viene illustrata in questa nota l’attuale condizione in cui si trova lo stato dell’ambiente marino nell’area del Canale di Sicilia immediatamente a ridosso della costa merdionale delle Sicilia dal Golfo di Gela alla costa iblea. Lo stato dell’ambiente in oggetto viene valutato in base alla distribuzione di alcuni metalli in tracce normalmente ritenuti fra i più significativi per gli scopi prefissati, esaminati lungo la colonna d’acqua e nei sedimenti superficiali di fondale. I risultati ottenuti, illustrati anche attraverso un’indagine statistica multivariata, indicano un arricchimento superficiale della colonna d’acqua in lantanio che può essere ascritto tanto ad un’origine litogenica, quanto ad input antropico. Nei sedimenti al naturale raggruppamento fra elementi in tracce in tracce basato su analogie comportamentali di carattere geochimica sfugge il Ni che mostra di provenire da una fonte differente per la quale non si può escludere la natura antropica. Tale ipotesi di esiti di antropizzazione trova conforto nei raffronti condotti con il chimismo del particolato atmosferico desunto da dati di letteratura

    Development of a biosensor for copper detection in aqueous solutions using an Anemonia sulcata recombinant GFP

    Fluorescent proteins from marine organisms represent potential candidates for biosensor development. In this paper, we described the isolation of a native green fluorescent protein from Anemonia sulcata and the cloning and purification of its equivalent as a recombinant protein in Escherichia coli. Furthermore, the spectroscopic behaviours of the native and recombinant GFPs were investigated as a function of Cu2+, Cd2+, Pb 2+ and Ni2+ concentration. Our results suggest the high selectivity of both proteins at copper than the other metals and, for the recombinant protein, a great sensitivity at a very low concentration (0.1-1 μM). Moreover, starting from these data, using the combination of molecular biology techniques and optical setup, we developed a device for the detection of Cu2+ in water solutions. The quenching effect detected with the device showed that the relative attenuation of the signal (0.46±0.02 AU) was slightly larger than the data measured by fluorescence spectra (0.65±0.03 AU). The good sensitivity in the span of two orders of the magnitude of Cu2+ concentration, the fact that the instrument is made up of low-cost and sturdy parts and the selective quenching of rAsGFP to copper ions make this setup suited as a low cost, on-the-field, copper ion-specific biosensor. © 2013 Springer Science+Business Media

    Partially Purified Extracts of Sea Anemone Anemonia viridis Affect the Growth and Viability of Selected Tumour Cell Lines

    In the last few years, marine species have been investigated for the presence of natural products with anticancer activity. Using reversed phase chromatography, low molecular weight proteins were fractionated from the sea anemone Anemonia viridis. Four different fractions were evaluated for their cytotoxic activity by means of erythrocyte haemolysis test, MTS, and LDH assays. Finally, the antiproliferative activities of three of these fractions were studied on PC3, PLC/PRF/5, and A375 human cancer cell lines. Our analysis revealed that the four fractions showed different protein contents and diverse patterns of activity towards human PBMC and cancer cell lines. Interestingly, fractions III and IV exerted cytotoxic effects on human cells. Conversely, fractions I and II displayed very low toxic effects associated with antiproliferative activities on cancer cell lines
