    An Adaptation To Life In Acid Through A Novel Mevalonate Pathway.

    Extreme acidophiles are capable of growth at pH values near zero. Sustaining life in acidic environments requires extensive adaptations of membranes, proton pumps, and DNA repair mechanisms. Here we describe an adaptation of a core biochemical pathway, the mevalonate pathway, in extreme acidophiles. Two previously known mevalonate pathways involve ATP dependent decarboxylation of either mevalonate 5-phosphate or mevalonate 5-pyrophosphate, in which a single enzyme carries out two essential steps: (1) phosphorylation of the mevalonate moiety at the 3-OH position and (2) subsequent decarboxylation. We now demonstrate that in extreme acidophiles, decarboxylation is carried out by two separate steps: previously identified enzymes generate mevalonate 3,5-bisphosphate and a new decarboxylase we describe here, mevalonate 3,5-bisphosphate decarboxylase, produces isopentenyl phosphate. Why use two enzymes in acidophiles when one enzyme provides both functionalities in all other organisms examined to date? We find that at low pH, the dual function enzyme, mevalonate 5-phosphate decarboxylase is unable to carry out the first phosphorylation step, yet retains its ability to perform decarboxylation. We therefore propose that extreme acidophiles had to replace the dual-purpose enzyme with two specialized enzymes to efficiently produce isoprenoids in extremely acidic environments

    Generating-function method for fusion rules

    This is the second of two articles devoted to an exposition of the generating-function method for computing fusion rules in affine Lie algebras. The present paper focuses on fusion rules, using the machinery developed for tensor products in the companion article. Although the Kac-Walton algorithm provides a method for constructing a fusion generating function from the corresponding tensor-product generating function, we describe a more powerful approach which starts by first defining the set of fusion elementary couplings from a natural extension of the set of tensor-product elementary couplings. A set of inequalities involving the level are derived from this set using Farkas' lemma. These inequalities, taken in conjunction with the inequalities defining the tensor products, define what we call the fusion basis. Given this basis, the machinery of our previous paper may be applied to construct the fusion generating function.     Move, Adapt, or Die: Lagopus Leucura Changes in Distribution, Habitat and Number at Glacier National Park, Montana

    ABSTRACT.—Because of the “montane island” effect and relative rates of climate change, alpine species may be particularly affected by changing climate. White-tailed Ptarmigan (Lagopus leucura) are adapted to life in the alpine cold, but may be less well-adapted to hot summer temperatures. In 1996 and 1997, ptarmigan living in Glacier National Park (GNP) were closely associated with remnant snow and free water in the late summer, a resource that is rapidly changing in distribution and extent as climate changes. We examined White-tailed Ptarmigan distribution, habitat, and numbers in 2009-2010 for comparison with 1996-1997 data. Briefly, five areas within GNP were searched for ptarmigan during August. Habitat data including microclimate were collected at flock locations. Microclimate at ptarmigan flock locations differed from other areas nearby. Flocking ptarmigan appeared to be less numerous than described in 1996 and 1997, and mean flock locations on Logan Pass moved 335 m upslope between the 1990s and 2009 and 2010. Ptarmigan in 2009 and 2010 chose habitat that had the same coverage of vegetation and rock as 1996 and 1997, but contained less snow and water, was farther from snow and water, contained lower soil moisture content, was higher in elevation, and steeper in slope. Our study demonstrates that White-tailed Ptarmigan in GNP have changed distribution, altered habitat preferences, and perhaps on a local scale, experienced declining population numbers in late summer

    Generating-function method for tensor products

    This is the first of two articles devoted to a exposition of the generating-function method for computing fusion rules in affine Lie algebras. The present paper is entirely devoted to the study of the tensor-product (infinite-level) limit of fusions rules. We start by reviewing Sharp's character method. An alternative approach to the construction of tensor-product generating functions is then presented which overcomes most of the technical difficulties associated with the character method. It is based on the reformulation of the problem of calculating tensor products in terms of the solution of a set of linear and homogeneous Diophantine equations whose elementary solutions represent ``elementary couplings''.     Hypoxia. Regulation of NFÎșB signalling during inflammation: the role of hydroxylases

    NFÎșB is a master regulator of innate immunity and inflammatory signalling. Microenvironmental hypoxia has long been identified as being coincident with chronic inflammation. The contribution of microenvironmental hypoxia to NFÎșB-induced inflammation has more recently been appreciated. Identification of the co-regulation of NFÎșB and hypoxia inducible factor (HIF) pathways by 2-oxo-glutarate-dependent hydroxylase family members has highlighted an intimate relationship between NFÎșB inflammatory signalling and HIF-mediated hypoxic signalling pathways. Adding another layer of complexity to our understanding of the role of NFÎșB inflammatory signalling by hypoxia is the recent recognition of the contribution of basal NFÎșB activity to HIF-1α transcription. This observation implicates an important and previously unappreciated role for NFÎșB in inflammatory disease where HIF-1α is activated. The present review will discuss recent literature pertaining to the regulation of NFÎșB inflammatory signalling by hypoxia and some of the inflammatory diseases where this may play an important role. Furthermore, we will discuss the potential for prolylhydroxylase inhibitors in inflammatory disease

    Equivalent qubit dynamics under classical and quantum noise

    We study the dynamics of quantum systems under classical and quantum noise, focusing on decoherence in qubit systems. Classical noise is described by a random process leading to a stochastic temporal evolution of a closed quantum system, whereas quantum noise originates from the coupling of the microscopic quantum system to its macroscopic environment. We derive deterministic master equations describing the average evolution of the quantum system under classical continuous-time Markovian noise and two sets of master equations under quantum noise. Strikingly, these three equations of motion are shown to be equivalent in the case of classical random telegraph noise and proper quantum environments. Hence fully quantum-mechanical models within the Born approximation can be mapped to a quantum system under classical noise. Furthermore, we apply the derived equations together with pulse optimization techniques to achieve high-fidelity one-qubit operations under random telegraph noise, and hence fight decoherence in these systems of great practical interest.Comment: 5 pages, 2 figures; converted to PRA format, added Fig. 2, corrected typo

    A precise CNOT gate in the presence of large fabrication induced variations of the exchange interaction strength

    We demonstrate how using two-qubit composite rotations a high fidelity controlled-NOT (CNOT) gate can be constructed, even when the strength of the interaction between qubits is not accurately known. We focus on the exchange interaction oscillation in silicon based solid-state architectures with a Heisenberg Hamiltonian. This method easily applies to a general two-qubit Hamiltonian. We show how the robust CNOT gate can achieve a very high fidelity when a single application of the composite rotations is combined with a modest level of Hamiltonian characterisation. Operating the robust CNOT gate in a suitably characterised system means concatenation of the composite pulse is unnecessary, hence reducing operation time, and ensuring the gate operates below the threshold required for fault-tolerant quantum computation.Comment: 9 pages, 8 figure

    Belimumab : a technological advance for systemic lupus erythematosus patients? Report of a systematic review and meta-analysis

    Objectives: To undertake a systematic review and meta-analysis to investigate clinical effectiveness of belimumab for patients with systemic lupus erythematosus (SLE) and antinuclear and/or anti-double-stranded DNA (dsDNA) autoantibodies. Methods: We searched eight electronic databases and reference lists for randomised controlled trials (RCTs) of belimumab against placebo or best supportive care. Quality assessment and random effects meta-analysis were undertaken. Design: A meta-analysis of RCTs. Participants: 2133 SLE patients. Primary and secondary outcome measures: SLE Responder Index (SRI) at week 52. Results: Three double-blind placebo-controlled RCTs (L02, BLISS-52 BLISS-76) investigated 2133 SLE patients. BLISS-52 and BLISS-76 trials recruited patients with antinuclear and/or anti-dsDNA autoantibodies and demonstrated belimumab effectiveness for the SRI at week 52. Ethnicity and geographical location of participants varied considerably between BLISS trials. Although tests for statistical heterogeneity were negative, BLISS-52 results were systematically more favourable for all measured outcomes. Meta-analysis of pooled 52-week SRI BLISS results showed benefit for belimumab (OR 1.63, 95% CI 1.27 to 2.09). By week 76, the primary SRI outcome in BLISS-76 was not statistically significant (OR 1.31, 95% CI 0.919 to 1.855)
