    Effets d’une partielle substitutionde la farine de poisson par la farine de soja sur la croissance des juvĂ©niles de la perche du Nil (Lates niloticus, Linnaeus 1758)

    La prĂ©sente Ă©tude a Ă©tĂ© menĂ©e pour Ă©valuer les effets de l’incorporation de la farine de soja dans le rĂ©gime alimentaire des juvĂ©niles de la perche du Nil. Ce travail est rĂ©alisĂ© sur des juvĂ©niles de 1g Ă©levĂ©s dans des aquariums de 50 l. Cinq rĂ©gimes alimentaires isoprotĂ©iques (45% de protĂ©ines) contenant diffĂ©rents taux de farine de soja 0%, 26%, 36%, 51% et 60% notĂ© respectivement A, B, C, D et E ont Ă©tĂ© prĂ©parĂ©s. Chaque rĂ©gime a Ă©tĂ© assignĂ© de façon alĂ©atoire Ă  trois groupes de poissons nourris Ă  satiĂ©tĂ© durant 8 semaines. Les rĂ©sultats obtenus montrent que les meilleures performances de croissance et d’efficacitĂ© alimentaire sont obtenues avec les rĂ©gimes nourris avec la farine de poisson (A), suivis dans l’ordre des rĂ©gimes B, C, D et E. Les taux de croissance spĂ©cifiques (TCS) varient entre 3,26% et 3,77% respectivement chez les poissons nourris avec les rĂ©gimes A, B, C, D et E. Le coefficient d’efficacitĂ© protĂ©ique (CEP) a variĂ© de 2,07 Ă  1,6. Les rĂ©sultats statistiques montrent que le meilleur CEP est obtenu avec le rĂ©gime A (P< 0.05) et le plus faible avec le rĂ©gime E. Les rĂ©sultats de la prĂ©sente Ă©tude indiquent qu’il est possible d’incorporer 51% de la farine de soja dans l’alimentation des juvĂ©niles de Lates niloticus sans compromettre leur croissance.Mots clĂ©s: Perche du Nil, Lates niloticus, farine de soja, grossissement, alimentation

    Evaluating extreme rainfall changes over Taiwan using a standardized index

    The annual daily maximum precipitation (rx1day) is widely used to represent extreme events and is an important parameter in climate change studies. However, the climate variability in rx1day is sensitive to outliers and has difficulty representing the characteristics of large areas. We propose to use the probability index (PI), based on the cumulative density function (CDF) of a generalized extreme value (GEV) distribution to fit and standardize the rx1day to represent extreme event records in this study. A good correlation between the area-averaged PIs of the observed stations and those of the gridded dataset can be found over Taiwan. From the past PI records, there is no distinct trend in western Taiwan before the end of the 20th century, but a climate regime change happened during 2002 - 2003. The dual change effects from both the variance and linear trend of extreme events are identified over the northeastern and southern parts of Taiwan, along with the island's central and southern regions, showing different abrupt changing trends and intensity. The PI can also be calculated using climate projection data to represent the characteristics of future extreme changes. The climate variability of PIs on the present (ALL) and future (RCP4.5 and RCP8.5) scenarios were evaluated using the 16 Couple Model Intercomparison Projects Phase-5 models (CMIP5). The simulated present fluctuations in PIs are smaller than those of actual observations.     Defects in ErbB-Dependent Establishment of Adult Melanocyte Stem Cells Reveal Independent Origins for Embryonic and Regeneration Melanocytes

    Adult stem cells are responsible for maintaining and repairing tissues during the life of an organism. Tissue repair in humans, however, is limited compared to the regenerative capabilities of other vertebrates, such as the zebrafish (Danio rerio). An understanding of stem cell mechanisms, such as how they are established, their self-renewal properties, and their recruitment to produce new cells is therefore important for the application of regenerative medicine. We use larval melanocyte regeneration following treatment with the melanocytotoxic drug MoTP to investigate these mechanisms in Melanocyte Stem Cell (MSC) regulation. In this paper, we show that the receptor tyrosine kinase, erbb3b, is required for establishing the adult MSC responsible for regenerating the larval melanocyte population. Both the erbb3b mutant and wild-type fish treated with the ErbB inhibitor, AG1478, develop normal embryonic melanocytes but fail to regenerate melanocytes after MoTP-induced melanocyte ablation. By administering AG1478 at different time points, we show that ErbB signaling is only required for regeneration prior to MoTP treatment and before 48 hours of development, consistent with a role in establishing MSCs. We then show that overexpression of kitla, the Kit ligand, in transgenic larvae leads to recruitment of MSCs, resulting in overproliferation of melanocytes. Furthermore, kitla overexpression can rescue AG1478-blocked regeneration, suggesting that ErbB signaling is required to promote the progression and specification of the MSC from a pre–MSC state. This study provides evidence that ErbB signaling is required for the establishment of adult MSCs during embryonic development. That this requirement is not shared with the embryonic melanocytes suggests that embryonic melanocytes develop directly, without proceeding through the ErbB-dependent MSC. Moreover, the shared requirement of larval melanocyte regeneration and metamorphic melanocytes that develops at the larval-to-adult transition suggests that these post-embryonic melanocytes develop from the same adult MSC population. Lastly, that kitla overexpression can recruit the MSC to develop excess melanocytes raises the possibility that Kit signaling may be involved in MSC recruitment during regeneration

    Going Coastal: Shared Evolutionary History between Coastal British Columbia and Southeast Alaska Wolves (Canis lupus)

    Many coastal species occupying the temperate rainforests of the Pacific Northwest in North America comprise endemic populations genetically and ecologically distinct from interior continental conspecifics. Morphological variation previously identified among wolf populations resulted in recognition of multiple subspecies of wolves in the Pacific Northwest. Recently, separate genetic studies have identified diverged populations of wolves in coastal British Columbia and coastal Southeast Alaska, providing support for hypotheses of distinct coastal subspecies. These two regions are geographically and ecologically contiguous, however, there is no comprehensive analysis across all wolf populations in this coastal rainforest.By combining mitochondrial DNA datasets from throughout the Pacific Northwest, we examined the genetic relationship between coastal British Columbia and Southeast Alaska wolf populations and compared them with adjacent continental populations. Phylogenetic analysis indicates complete overlap in the genetic diversity of coastal British Columbia and Southeast Alaska wolves, but these populations are distinct from interior continental wolves. Analyses of molecular variation support the separation of all coastal wolves in a group divergent from continental populations, as predicted based on hypothesized subspecies designations. Two novel haplotypes also were uncovered in a newly assayed continental population of interior Alaska wolves.We found evidence that coastal wolves endemic to these temperate rainforests are diverged from neighbouring, interior continental wolves; a finding that necessitates new international strategies associated with the management of this species

    Hypermutable Non-Synonymous Sites Are under Stronger Negative Selection

    Mutation rate varies greatly between nucleotide sites of the human genome and depends both on the global genomic location and the local sequence context of a site. In particular, CpG context elevates the mutation rate by an order of magnitude. Mutations also vary widely in their effect on the molecular function, phenotype, and fitness. Independence of the probability of occurrence of a new mutation's effect has been a fundamental premise in genetics. However, highly mutable contexts may be preserved by negative selection at important sites but destroyed by mutation at sites under no selection. Thus, there may be a positive correlation between the rate of mutations at a nucleotide site and the magnitude of their effect on fitness. We studied the impact of CpG context on the rate of human–chimpanzee divergence and on intrahuman nucleotide diversity at non-synonymous coding sites. We compared nucleotides that occupy identical positions within codons of identical amino acids and only differ by being within versus outside CpG context. Nucleotides within CpG context are under a stronger negative selection, as revealed by their lower, proportionally to the mutation rate, rate of evolution and nucleotide diversity. In particular, the probability of fixation of a non-synonymous transition at a CpG site is two times lower than at a CpG site. Thus, sites with different mutation rates are not necessarily selectively equivalent. This suggests that the mutation rate may complement sequence conservation as a characteristic predictive of functional importance of nucleotide sites

    COX inhibitors and breast cancer

    There is considerable evidence to suggest that prostaglandins play an important role in the development and growth of cancer. The enzyme cyclo-oxygenase (COX) catalyses the conversion of arachidonic acid to prostaglandins. In recent years, there has been interest in a possible role for COX inhibitors in the prevention and treatment of malignancy. Cyclo-oxygenase-2 (COX-2) is overexpressed in several epithelial tumours, including breast cancer. Preclinical evidence favours an antitumour role for COX inhibitors in breast cancer. However, the epidemiological evidence for an association is conflicting. Trials are being conducted to study the use of COX inhibitors alone and in combination with other agents in the chemoprevention of breast cancer, and in the neo-adjuvant, adjuvant, and metastatic treatment settings. In evaluating the potential use of these agents particularly in cancer chemoprophylaxis, the safety profile is as important as their efficacy. Concern over the cardiovascular safety of both selective and nonselective COX-inhibitors has recently been highlighted
