    Shear rheological properties of fresh human faeces with different moisture content

    Dry sanitation requires the handling of faeces, which vary in age and degree of transformation. Rheological data are necessary to support the design of equipment to handle faeces. The rheological properties of fresh human faeces were measured using a variable-speed rotational rheometer. Samples were further tested for moisture content, total solids, volatile content, and ash content. Faecal samples were found to have a yield stress; there was a decrease in apparent viscosity with increasing shear rate. For any given shear rate, higher apparent viscosities are associated with lower moisture contents. Across a range of water contents of 58.5% to 88.7%, apparent viscosities of 27 Pa∙s to 2 014 Pa∙s were measured at a shear rate of 1 s-1. During constant shear tests, the apparent viscosity of all faeces was found to decrease asymptotically, where the minimum apparent viscosity value increased with decreasing moisture content. A structural recovery test indicates that human faeces are thixotropic in behaviour, where the viscosity permanently decreases to 0.5% of the initial value after a 20 s exposure to a shear rate of 10 s-1. A linear relationship between viscosity and temperature was found, with a recorded 30.6% decrease in viscosity for a 35.6 °C increase in temperature from 13.4°C.Keywords: Human stool, mechanical properties, water content, flow curve, viscosity, yield stres

    Laboratory protocols for testing the efficacy of commercial pit latrine additives

    There is considerable national interest in the use of commercial microbially derived products for controlling the rate of accumulation of the contents of pit latrines. Manufacturers claim that some of these products can reduce accumulation rates, prevent the pit from ever filling up, or even result in decreases in pit contents volume. Prior to this research, there have been no scientific publications that have conclusively supported or refuted these claims.This project undertook to perform reproducible laboratory tests that would quantify the effect of commercial pit latrine additive products. Protocols were developed and tested on a range of different commercial products sold for their ability to control the rate of accumulation of pit latrine contents. The effect of commercial additives on mass loss from VIP sludge in 300 g honey jars was compared to mass loss from similar units subjected to no treatment and treatment with water.The purpose of these experiments was to separate and quantify the effect of micro-organisms or enzymes originating from commercial pit latrine additives from the effect of natural processes within the pit latrine sludge (including digestion by micro-organisms in the VIP sludge and dehydration) and the effects of other actions associated with treatment, such as the addition of water.Results indicated that insignificant mass loss occurred in all anaerobic test units, while significant mass loss occurred in all other test units. However, there was no statistically significant difference between any of the different treatments in the aerobic units. Investigation of analytical data from the test units indicated that mass loss in aerobic units was due to a combination of dehydration through evaporation of moisture and biological stabilisation processes, and that the latter were not significantly enhanced by the addition of commercial pit latrine products. It was concluded that there was no evidence to support claims that pit latrine additives could extend the life of a pit latrine

    An investigation of the effect of pit latrine additives on VIP latrine sludge content under laboratory and field trials

    Sludge content in VIP latrines is degraded mainly under anaerobic conditions and the process is relatively slow. At varying stages of digestion within pit latrines, sludge accumulates and odour and fly nuisance may occur which could pose risks to public health and the environment. Management of accumulated sludge in pit latrines has been a major problem facing a number of municipalities in South Africa and is also a global issue. Manufacturers of various commercial pit latrine additives claim that by addition of this product to pit content, accumulation rate and pit content volume can be reduced, thereby preventing the pit from ever reaching capacity. This paper presents a comprehensive study conducted to determine the effects of additives on pit contents under laboratory and field conditions. By conducting both laboratory and field trials, it was possible to identify whether there is any acceleration of mass or volume stabilisation as a result of additive addition, and whether any apparent effect is a result of biodegradation or of compaction. The results indicated that neither laboratory trials nor field trials provided any evidence that the use of pit additives has any beneficial effect on pit contents. The reasons why additives seem to not have any beneficial effects are also discussed.Keywords: additives, digestion, pit content, sludge, public health, VIP latrin

    Variation in VIP latrine sludge contents

    This study investigated variations in the characteristics of the sludge content from different ventilated improved pit (VIP) latrines and variation in these characteristics at specific depths within each pit. Faecal sludge from 16 VIP latrines within the eThekwini Municipality was collected and laboratory characterisation including moisture content, total and volatile solids, chemical oxygen demand, and aerobic biodegradability was performed. Sludge samples were collected from 4 specific depths within each pit investigated. The laboratory characterisation performed showed that none of the VIP latrines investigated had the same sludge characteristics, and that within a pit sludge characteristics varied with increasing depth in the pit. This supports the motivating hypothesis that, depending on household habits and local environmental conditions, there should be considerable variation in the organic contents, moisture content, non-biodegradable content and microbial population between different pits. This variation with increasing depth within a pit is expected, since fresh material is constantly being added to the pit overlaying older material which might have undergone a certain degree of stabilisation

    Simulation of a wastewater treatment plant receiving industrial effluents

    A process model simulating the Mariannridge Wastewater Treatment Plant, located in the eThekwini Municipality, has been developed in the WEST (Worldwide Engine for Simulation, Training and Automation) modelling environment, based on the IWA Activated Sludge Model No. 3 (ASM3). The treatment plant receives a high proportion of industrial effluents.The development of the model involves the characterisation of the influent wastewater and determining model parameters (kinetic and stoichiometric coefficients) by undertaking batch respirometric tests on the wastewater and activated sludge, flocculation filtration and simulation of the batch respirometric experiment. To account for equipment-specific factors, thesimulation model was calibrated against plant data covering a year’s operation. The model is intended to be used as part of a system to evaluate the ability of a receiving wastewater treatment works to adequately treat a particular industrial effluent before granting a permit for it to be discharged to sewer

    The study assessed the suitability of two effluent types, namely anaerobic filtered (AF) and horizontal flow constructed wetland (HFCW) effluent for Moistube irrigation (MTI). Secondary to this, the study determined the plugging coefficients (α) on MTI for the respective effluents. The feed water was supplied from a raised tank (3.5 m), and mass-flow rates were recorded at 15 min intervals using an electronic balance. The effluent feed water concentrations and experimental room temperature (25 °C ± 1 °C) were continuously monitored and kept constant. Hermia's models based on the [Formula: see text] coefficient was used to select the best fitting fouling mechanism model and, consequently, the plugging coefficients. In addition, microbial colony analysis and scanning electron microscopy (SEM) analysis was carried out to assess the composition of the deposited sediment (DS) and adhered bacterial film (ABF) onto the MTI lateral. The study revealed that MTI pore blocking was a complex phenomenon described by complete pore-blocking model ([Formula: see text] ≥ 0.50). Discharge followed an exponential decay with early fouling observed on AF effluent because of a high concentration of total suspended solids (TSS) and dissolved organic matter (DOM). Discharge declined by 50% after 20 and 10 h of intermittent operation for AF and HFCW effluent, respectively. The α for each effluent (foulant) were [Formula: see text] = 0.07 and [Formula: see text] = 0.05, respectively, for AF and HFCW. The microbial analysis revealed bacterial aggregation structures that contributed to pore blocking. SEM imaging revealed complete surface coverage by deposited sediment. It is concluded that water quality determines the operation life span of MTI, and the two effluents promote accelerated MTI pore fouling or blocking. Continuous use without flushing the MTI will promote membrane degradation and reduced discharge efficiency. Additional filtration can potentially mitigate the membrane degradation process

    Water security in South Africa: perceptions on public expectations and municipal obligations, governance and water re-use

    South Africa is a water-scarce country with increasing pressure on its water resources. Urgent interventions are needed to protect water security within this rapidly developing country. This paper reports on an interdisciplinary Water Security Colloquium, convened by the South African Young Academy of Science in 2014. A selected group of water professionals from academia, civil society and local government was brought together to discuss water security under three focus themes: ‘public expectations and municipal obligations’, ‘water security and governance: challenges and advances’, and ‘water re-use: health and infrastructural considerations’. Participant perceptions were generated using a focus group methodology, combined with participatory data collection methods. Under each theme, inputs were categorised as ‘challenges’, ‘gaps in knowledge’, and ‘solutions/recommendations’ and these inputs were thereafter ranked in order of importance via a ‘voting’ process. Major challenges perceived included a lack of both skills and political will in government, a need to restore citizen trust in government intention and capability to deliver water-related services, and a failure to up-scale existing water re-use technology. Participants identified understanding of the process and implications of the Green and Blue Drop Programmes, knowledge transfer to the public, and the role of educators as major knowledge gaps. The top suggestions proposed included creating public awareness around and buy-in to initiatives to improve water security, accessible and user-friendly conversion of research results to implementation, and ensuring an active role for educators in creating awareness around water security. In view of the concerns identified, participants suggested as potential solutions: improving government and public understanding around water issues, incentivising water re-use and conservation, introducing rising block tariffs and improving human capacity development in the water sector. Developing the ecological infrastructure that protects both quantity and quality of water and building strong partnerships among all stakeholders were also recognised as key.Keywords: capacity development, education, governance, water security, water re-use, water conservation, local governmen

    Novel statistical approaches for non-normal censored immunological data: analysis of cytokine and gene expression data

    Background: For several immune-mediated diseases, immunological analysis will become more complex in the future with datasets in which cytokine and gene expression data play a major role. These data have certain characteristics that require sophisticated statistical analysis such as strategies for non-normal distribution and censoring. Additionally, complex and multiple immunological relationships need to be adjusted for potential confounding and interaction effects. Objective: We aimed to introduce and apply different methods for statistical analysis of non-normal censored cytokine and gene expression data. Furthermore, we assessed the performance and accuracy of a novel regression approach in order to allow adjusting for covariates and potential confounding. Methods: For non-normally distributed censored data traditional means such as the Kaplan-Meier method or the generalized Wilcoxon test are described. In order to adjust for covariates the novel approach named Tobit regression on ranks was introduced. Its performance and accuracy for analysis of non-normal censored cytokine/gene expression data was evaluated by a simulation study and a statistical experiment applying permutation and bootstrapping. Results: If adjustment for covariates is not necessary traditional statistical methods are adequate for non-normal censored data. Comparable with these and appropriate if additional adjustment is required, Tobit regression on ranks is a valid method. Its power, type-I error rate and accuracy were comparable to the classical Tobit regression. Conclusion: Non-normally distributed censored immunological data require appropriate statistical methods. Tobit regression on ranks meets these requirements and can be used for adjustment for covariates and potential confounding in large and complex immunological datasets
