    Using Impedance for Mechanical Conditioning of Tomato Transplants to Control Excessive Stem Elongation

    Mechanical stimulation is known to control excessive stem elongation in highdensity tomato (Lycopersicon esculentum Mill.) transplants. Mechanical stimulation using physical impedance provided height control equivalent to that obtained using brushing. Low-cost materials can be used to apply the impedance. Mylar film in a plastic frame was equivalent to expensive acrylic sheets in its effect on plant height (40 mm shorter than nontreated, a 40% reduction in the elongation rate during the treatment period), stem diameter (18% thicker), and biomass (14% lighter) when they applied a pressure of 66 N·m-2• Stem elongation was not reduced ifless pressure was applied (25 or50 N·m-2). Height control was equally effective with a solid material (mylar film) and a permeable material (fiberglass insect screen), indicating that restricting air movement is not an important mechanism for the growth response. Overnight treatments resulted in the desired growth response (27 mm shorter than nontreated, a 30% reduction in elongation rate), but 0.5-h treatments had insufficient effect for commercial use (11 mm shorter, 10% reduction in elongation rate). These experiments demonstrate that impedance can be used in commercial production conditions to control tomato transplant height with inexpensive materials. However, satisfactory height control requires a large applied force and a long daily treatment period

    Transparent yttrium hydride thin films prepared by reactive sputtering

    Metal hydrides have earlier been suggested for utilization in solar cells. With this as a motivation we have prepared thin films of yttrium hydride by reactive magnetron sputter deposition. The resulting films are metallic for low partial pressure of hydrogen during the deposition, and black or yellow-transparent for higher partial pressure of hydrogen. Both metallic and semiconducting transparent YHx films have been prepared directly in-situ without the need of capping layers and post-deposition hydrogenation. Optically the films are similar to what is found for YHx films prepared by other techniques, but the crystal structure of the transparent films differ from the well-known YH3 phase, as they have an fcc lattice instead of hcp

    Mechanical Conditioning for Controlling Excessive Elongation in Tomato Transplants: Sensitivity to Dose, Frequency, and Timing of Brushing

    During production of plug transplants, the high plant density results in rapid stem elongation as plants compete for light. The resulting tall, weak-stemmed plants are difficult to transplant and are easily damaged. One technique that can prevent excessive elongation is mechanical stimulation by brushing. Wide adoption of brushing is limited by a lack of information on how plants respond to variations in applying the technique. Our investigation examined how tomato (Lycopersicon esculentum cv. Oh8245) seedling growth responded to varying doses of mechanical stimulation, varying intervals between brush strokes during stimulation, time of day that stimulation was applied, and growth stage at which application started. Seedlings were grown in 288-cell flats at 2100 plants/m2. Daily doses from 0 to 40 brush strokes were applied from canopy closure until the nontreated plants reached a canopy height of 15 cm. The final height was reduced by ≈20% for all brushed treatments, with little further effect with \u3e10 strokes/d. Intervals between strokes as long as 10 minutes resulted in the same reduction in the rate of stem elongation as the same daily dose applied in one continuous treatment. Treatments were similarly effective whether applied in the morning or late afternoon. Treatments begun at a canopy height of 6 (canopy closure), 8, or 10 cm gave similar reductions in the rate of stem elongation. Plants grew 6 mm·d–1 when they were not treated and 3 mm·d–1 when treated. Therefore, the final height was directly related to the number of treatment days. Stimulation appears to be sensed and integrated over at least half an hour and the reduction in the rate of stem elongation expressed over the subsequent daily cycle of growth. All results indicate that there is substantial flexibility in applying brushing for controlling elongation in tomato transplants

    Mechanical Conditioning of Tomato Seedlings Improves Transplant Quality without Deleterious Effects on Field Performance

    Excessive stem elongation reduces plant survival in the field and hinders mechanical transplanting. Mechanical conditioning is an effective method for reducing stem elongation during transplant production. This investigation examined the consequences of mechanical conditioning, using brushing and impedance, on subsequent field performance of tomatoes (Lycopersicon esculentum Mill.). Mechanically conditioned transplants of processing tomatoes resumed growth after transplant shock as quickly as did untreated plants, and subsequent canopy development was also equal. In 4 years of field trials, yield was not reduced by mechanical conditioning. Transplants for fresh-market tomatoes may be more sensitive to injury than those for processing tomatoes because they flower sooner after the conditioning treatments. Nevertheless, neither earliness nor defects in the fruits of the first cluster were affected by mechanical conditioning. Early and total yields were equal in both years that fresh-market crops were tested. Thus, there were no adverse effects on field performance of either processing or fresh-market tomatoes as a result of reducing stem elongation by mechanical conditioning before transplanting. Improved wind tolerance was tested both in a wind tunnel and in the field. In wind-tunnel tests, brushed and impeded plants resisted stem bending at wind speeds 4 to 12 km·h–1 higher than did untreated plants. A 70 km·h–1 wind after transplanting killed 12% of untreated plants but only 2% of treated plants.     Surface oxide on thin films of yttrium hydride studied by neutron reflectometry

    The applicability of standard methods for compositional analysis is limited for H-containing films. Neutron reflectometry is a powerful, non-destructive method that is especially suitable for these systems due to the large negative scattering length of H. In this work we demonstrate how neutron reflectometry can be used to investigate thin films of yttrium hydride. Neutron reflectometry gives a strong contrast between the film and the surface oxide layer, enabling us to estimate the oxide thickness and oxygen penetration depths. A surface oxide layer of 5-10 nm thickness was found for unprotected yttrium hydride films

    Polymerase chain reaction is superior to serology for the diagnosis of acute Mycoplasma pneumoniae infection and reveals a high rate of persistent infection

    <p>Abstract</p> <p>Background</p> <p>Diagnosis of <it>Mycoplasma pneumoniae </it>(MP) infection is traditionally based on serology, which may require more than two weeks for diagnostic antibodies to develop. PCR-based methods offer earlier diagnosis. During a community outbreak of MP infection, we compared semi-nested and real-time PCR of oropharyngeal swabs with serology for diagnosis of MP infection at different time points after disease onset. PCR-positive individuals were followed longitudinally to assess the persistence of MP DNA in throat secretions. We also studied carriage of MP among household contacts and school children.</p> <p>Results</p> <p>MP infection was diagnosed in 48 of 164 patients with respiratory tract infection. Forty-five (29%) had detectable MP DNA in oropharynx. A significant increase in MP IgG IgG titre or MP IgM antibodies was detected in 44/154 (27%) subjects. Two MP PCR-positive patients lacked antibody responses. Sera were missing from another two patients. The agreement between serology and PCR was good, κ = 0.90.</p> <p>During the first three weeks after disease onset the performance of PCR was excellent and all patients but one were detected. In contrast, only 21% of the patients with confirmed MP infection were positive by serum 1 during the first symptomatic week (56% during the second and 100% during the third week). Only 1/237 (0.4%) school children was positive by PCR. This child had respiratory symptoms. Eighteen of 22 (75%) symptomatic household contacts were MP PCR positive.</p> <p>Persistence of MP DNA in the throat was common. Median time for carriage of MP DNA was 7 weeks after disease onset (range 2 days – 7 months). Adequate antibiotic treatment did not shorten the period of persistence. Bacterial load, measured by quantitative real-time PCR declined gradually, and all followed patients eventually became PCR-negative.</p> <p>Conclusion</p> <p>PCR is superior to serology for diagnosis of MP infection during the early phases of infection. Persistent, sometimes long-term, carriage of MP DNA in the throat is common following acute infection, and is not affected by antibiotic therapy. Asymptomatic carriage of MP even during an outbreak is uncommon.</p

    Cryptosporidium parvum and Giardia intestinalis in Calf Diarrhoea in Sweden

    The objective of this study conducted in 75 herds was to investigate the presence and significance of Cryptosporidium parvum and Giardia intestinalis in Swedish dairy calves in comparison with rotavirus, coronavirus and Escherichia coli K99+. The farmers were asked to collect faecal samples from each heifer calf that had diarrhoea between birth and 90 days of age, and also from a healthy calf of the same age. In total, 270 samples were collected and analysed. C. parvum, either alone or together with G. intestinalis and/or rotavirus, was detected in 16 (11%) and 6 (5%) of the samples from diarrhoeic and healthy calves, respectively. Even though a higher proportion of diarrhoeic calves shed C. parvum, the difference between the groups was not statistically significant (p = 0.067), possibly due to the low number of positive samples. G. intestinalis was found in 42 (29%) of the diarrhoea samples and in 29 (23%) of the samples from healthy calves. Rotavirus and coronavirus were demonstrated in 24% and 3% of the diarrhoea samples, respectively, whereas E. coli K99+ was only found in samples from 2 healthy calves. C. parvum and G. intestinalis were found in samples from calves 7 to 84 days of age and during all seasons. The results confirm that C. parvum is present in Swedish dairy herds and might have clinical significance. G. intestinalis was the most common agent found but the importance of this parasite remains unclear. Both parasites have suggested zoonotic potential and thus warrant further attention. In addition, rotavirus is a major pathogen in neonatal enteritis in Sweden, whereas coronavirus and E. coli K99+ seem to be of less importance

    First ice core records of NO3− stable isotopes from Lomonosovfonna, Svalbard

    Samples from two ice cores drilled at Lomonosovfonna, Svalbard, covering the period 1957–2009, and 1650–1995, respectively, were analyzed for NO3− concentrations, and NO3− stable isotopes (δ15N and δ18O). Post-1950 δ15N has an average of (−6.9 ± 1.9) ‰, which is lower than the isotopic signal known for Summit, Greenland, but agrees with values observed in recent Svalbard snow and aerosol. Pre-1900 δ15N has an average of (4.2 ± 1.6) ‰ suggesting that natural sources, enriched in the 15 N-isotope, dominated before industrialization. The post-1950 δ18O average of (75.1 ± 4.1) ‰ agrees with data from low and polar latitudes, suggesting similar atmospheric NOy (NOy = NO + NO2 + HNO3) processing pathways. The combination of anthropogenic source δ15N and transport isotope effect was estimated as −29.1 ‰ for the last 60 years. This value is below the usual range of NOx (NOx = NO + NO2) anthropogenic sources which is likely the result of a transport isotope effect of –32 ‰. We suggest that the δ15N recorded at Lomonosovfonna is influenced mainly by fossil fuel combustion, soil emissions and forest fires; the first and second being responsible for the marked decrease in δ15N observed in the post-1950s record with soil emissions being associated to the decreasing trend in δ15N observed up to present time, and the third being responsible for the sharp increase of δ15N around 2000

    Reduced Population Control of an Insect Pest in Managed Willow Monocultures

    BACKGROUND: There is a general belief that insect outbreak risk is higher in plant monocultures than in natural and more diverse habitats, although empirical studies investigating this relationship are lacking. In this study, using density data collected over seven years at 40 study sites, we compare the temporal population variability of the leaf beetle Phratora vulgatissima between willow plantations and natural willow habitats. METHODOLOGY/PRINCIPAL FINDINGS: The study was conducted in 1999-2005. The density of adult P. vulgatissima was estimated in the spring every year by a knock-down sampling technique. We used two measures of population variability, CV and PV, to compare temporal variations in leaf beetle density between plantation and natural habitat. Relationships between density and variability were also analyzed to discern potential underlying processes behind stability in the two systems. The results showed that the leaf beetle P. vulgatissima had a greater temporal population variability and outbreak risk in willow plantations than in natural willow habitats. We hypothesize that the greater population stability observed in the natural habitat was due to two separate processes operating at different levels of beetle density. First, stable low population equilibrium can be achieved by the relatively high density of generalist predators observed in natural stands. Second, stable equilibrium can also be imposed at higher beetle density due to competition, which occurs through depletion of resources (plant foliage) in the natural habitat. In willow plantations, competition is reduced mainly because plants grow close enough for beetle larvae to move to another plant when foliage is consumed. CONCLUSION/SIGNIFICANCE: To our knowledge, this is the first empirical study confirming that insect pest outbreak risk is higher in monocultures. The study suggests that comparative studies of insect population dynamics in different habitats may improve our ability to predict insect pest outbreaks and could facilitate the development of sustainable pest control in managed systems

    Willingness and ability to pay for artemisinin-based combination therapy in rural Tanzania

    The aim of this study was to analyse willingness to pay (WTP) and ability to pay (ATP) for ACT for children below five years of age in a rural setting in Tanzania before the introduction of artemisinin-based combination therapy (ACT) as first-line treatment for uncomplicated malaria. Socio-economic factors associated with WTP and expectations on anti-malaria drugs, including ACT, were also explored. Structured interviews and focus group discussions were held with mothers, household heads, health-care workers and village leaders in Ishozi, Gera and Ishunju wards in north-west Tanzania in 2004. Contingent valuation method (CVM) was used with "take-it-or-leave-it" as the eliciting method, expressed as WTP for a full course of ACT for a child and households' opportunity cost of ACT was used to assess ATP. The study included descriptive analyses with multivariate adjustment for potential confounding factors. Among 265 mothers and household heads, 244 (92%, CI = 88%-95%) were willing to pay Tanzanian Shillings (TSh) 500 (US0.46)forachild′sdoseofACT,butonly55 0.46) for a child's dose of ACT, but only 55% (49%-61%) were willing to pay more than TSh 500. Mothers were more often willing to pay than male household heads (adjusted odds ratio = 2.1, CI = 1.2-3.6). Socio-economic status had no significant effect on WTP. The median annual non-subsidized ACT cost for clinical malaria episodes in an average household was calculated as US 6.0, which would represent 0.9% of the average total consumption expenditures as estimated from official data in 2001. The cost of non-subsidized ACT represented 7.0% of reported total annual expenditure on food and 33.0% of total annual expenditure on health care."Rapid effect," "no adverse effect" and "inexpensive" were the most desired features of an anti-malarial drug. WTP for ACT in this study was less than its real cost and a subsidy is, therefore, needed to enable its equitable affordability. The decision taken in Tanzania to subsidize Coartem fully at governmental health care facilities and at a consumer price of TSh 300-500 (US$ 0.28-0.46) at special designated shops through the programme of Accredited Drug Dispensing Outlets (ADDOs) appears to be well founded
