    Implementing strategies to overcome social-ecological traps: the role of community brokers and institutional bricolage in a locally managed marine area

    We examine collaborative arrangements for resource management between communities and external agencies, with a particular interest in how community-based interventions are integrated into local contexts. This timely inquiry comes in a period when participatory resource management approaches are increasingly being applied to overcome the perpetual cycles of poverty in which poor resource-dependent people find themselves, i.e., social-ecological traps. Much of the literature on social-ecological traps has focused on identifying conditions, factors, and responses that are important in, for example, alleviating systemic poverty or developing sustainable resource management systems. However, insufficient focus has been placed on understanding the practical processes by which strategies are implemented, and how these can reflexively affect the system itself. Drawing from a case study of a locally managed marine area in Indonesia, we examine the interactions between a nongovernment organization and a target community during the implementation of a fisheries management plan. Applying insights from rural development studies, we show how external interventions, designed to pull people out of social-ecological traps, are operationalized into forms that make them locally familiar and appropriate through actions of community brokers and processes of institutional bricolage. We argue that as a consequence of implementing processes, such interventions should be expected to diverge from their initial science-based justification

    The performance of a graded index multimode fiber optic low coherence Mach-Zehnder interferometer with phase modulation is analyzed. Investigated aspects were its ability to measure path length distributions and to perform path length resolved Doppler measurements of multiple scattered photons in a turbid suspension of particles undergoing Brownian and translational motion. The path length resolution of this instrument is compared with a system using single mode fibers for illumination and detection. The optical path lengths are determined from the zero order moment of the phase modulation peak in the power spectrum.     Scoping review: quality of life of siblings of children who are deaf and hard of hearing, have a vision or motor impairment

    This study identified the current state of knowledge about the Quality of Life (QoL) of siblings of children who are deaf and hard of hearing (DHH), have a vision impairment (VI) or motor impairment (MI). Additionally, factors associated with individual variation in QoL were examined. A scoping review was performed using PsycInfo, ERIC, Web of Science, and CINAHL. Empirical studies with siblings (age 0-18 years) of children who are DHH, have a VI or MI that investigate the QoL of siblings were included. A total of 1746 studies were identified of which 11 met the inclusion criteria. The results showed that QoL has different interpretations and various measurement tools are used. The findings showed both positive and negative outcomes for the QoL of siblings. For example, family cohesion was found as a positive consequence. A negative consequence could be a higher score on problem behavior. Personal and parental characteristics such as age and parental availability were two main factors related to individual variation in QoL. Insight in the factors related to individual variation may help researchers to consider the research perspective. In addition, healthcare providers can use the information to be either aware or intervene on specific factors that are related to the QoL of the children who are DHH, have a VI or MI and their siblings

    T Cell Maturation Stage Prior to and During GMP Processing Informs on CAR T Cell Expansion in Patients

    textabstractAutologous T cells were genetically modified to express a chimeric antigen receptor (CAR) directed toward carboxy-anhydrase-IX (CAIX) and used to treat patients with CAIX-positive metastatic renal cell carcinoma. In this study, we questioned whether the T cell maturation stage in the pre-infusion product affected CAIX CAR expression and function in vitro as well as in vivo CAR T cell numbers and expansion. During the 14 days expansion of CAR T cells prior to administration, we observed shifts from a predominant CD4 to a CD8 T cell phenotype and from a significant fraction of naĂŻve to central effector T cells. Surface expression of the CAR was equally distributed among different T cell subsets and T cell maturation stages. During T cell culture days 14-18 (which covered patient treatment days 1-5), T cells demonstrated a decline in CAR expression level per cell irrespective of T cell maturation stage, although the proportion of CAR-positive T cells and CAR-mediated T cell effector functions remained similar for both CD4 and CD8 T cell populations. Notably, patients with a higher fraction of naĂŻve CD8 T cells at baseline (prior to genetic modification) or central effector CD8 T cells at 2 weeks of CAR T cell culture demonstrated a higher fold expansion and absolute numbers of circulating CAR T cells at 1 month after start of therapy. We conclude that the T cell maturation stage prior to and during CAR T cell expansion culture is related to in vivo CAR T cell expansion

    Gene conversion in human rearranged immunoglobulin genes

    Over the past 20 years, many DNA sequences have been published suggesting that all or part of the V<sub>H</sub> segment of a rearranged immunoglobulin gene may be replaced in vivo. Two different mechanisms appear to be operating. One of these is very similar to primary V(D)J recombination, involving the RAG proteins acting upon recombination signal sequences, and this has recently been proven to occur. Other sequences, many of which show partial V<sub>H</sub> replacements with no addition of untemplated nucleotides at the V<sub>H</sub>–V<sub>H</sub> joint, have been proposed to occur by an unusual RAG-mediated recombination with the formation of hybrid (coding-to-signal) joints. These appear to occur in cells already undergoing somatic hypermutation in which, some authors are convinced, RAG genes are silenced. We recently proposed that the latter type of V<sub>H</sub> replacement might occur by homologous recombination initiated by the activity of AID (activation-induced cytidine deaminase), which is essential for somatic hypermutation and gene conversion. The latter has been observed in other species, but not in human Ig genes, so far. In this paper, we present a new analysis of sequences published as examples of the second type of rearrangement. This not only shows that AID recognition motifs occur in recombination regions but also that some sequences show replacement of central sections by a sequence from another gene, similar to gene conversion in the immunoglobulin genes of other species. These observations support the proposal that this type of rearrangement is likely to be AID-mediated rather than RAG-mediated and is consistent with gene conversion

    System performance of a 4-channel PHASAR WDM receiver operating at 1.2 Gbit/s

    Phased arrays are important key components in wavelength-division multiplexing (WDM) systems. We have realized a 4-channel WDM receiver combining a phased array with photodetectors on InP with a Si bipolar transimpedance amplifier. The channels are spaced at 2.0 nm with a 1.0-nm flat passband. On chip loss was 6-7 dB and detector efficiency was better than 90%. The optical cross talk remains below -20 dB. The electrical bandwidth per channel was 1 GHz. The electrical cross talk at 1 GHz after detection is below -25 dB. We have tested this receiver in a full 4-channel syste
