    Immunogold Labeling to Enhance Contrast in Optical Coherence Microscopy of Tissue Engineered Corneal Constructs

    Our lab has used an optical coherence microscope (OCM) to assess both the structure of tissue-engineered corneal constructs and their transparency. Currently, we are not able to resolve cells versus collagen matrix material in the images produced. We would like to distinguish cells in order to determine if they are viable while growing in culture and also if they are significantly contributing to the light scattering in the tissue. In order to do this, we are currently investigating the use of immunogold labeling. Gold nanoparticles are high scatterers and can create contrast in images. We have conjugated gold nanoparticles to antibodies specific to the α5 β1 integrins expressed in some corneal cells. This choice of target should allow assessment of the phenotypic behavior of the cells in the tissue, as different integrins are expressed in different phenotypes. This study applies the immunogold technique to study cultured corneal cells using the OCM with the ultimate goal of monitoring cellular behavior in engineered tissue and corroborating results from standard histological methods

    Geology and geochronology of the Tana Basin, Ethiopia: LIP volcanism, super eruptions and Eocene–Oligocene environmental change

    New geological and geochronological data define four episodes of volcanism for the Lake Tana region in the northern Ethiopian portion of the Afro–Arabian Large Igneous Province (LIP): pre-31 Ma flood basalt that yielded a single 40Ar/39Ar age of 34.05±0.54/0.56 Ma; thick and extensive felsic ignimbrites and rhyolites (minimum volume of 2–3×103 km3) erupted between 31.108±0.020/0.041 Ma and 30.844±0.027/0.046 Ma (U–Pb CA-ID-TIMS zircon ages); mafic volcanism bracketed by 40Ar/39Ar ages of 28.90±0.12/0.14 Ma and 23.75±0.02/0.04 Ma; and localised scoraceous basalt with an 40Ar/39Ar age of 0.033±0.005/0.005 Ma. The felsic volcanism was the product of super eruptions that created a 60–80 km diameter caldera marked by km-scale caldera-collapse fault blocks and a steep-sided basin filled with a minimum of 180 m of sediment and the present-day Lake Tana. These new data enable mapping, with a finer resolution than previously possible, Afro–Arabian LIP volcanism onto the timeline of the Eocene–Oligocene transition and show that neither the mafic nor silicic volcanism coincides directly with perturbations in the geochemical records that span that transition. Our results reinforce the view that it is not the development of a LIP alone but its rate of effusion that contributes to inducing global-scale environmental change

    Superconductivity Induced by Bond Breaking in the Triangular Lattice of IrTe2

    IrTe2, a layered compound with a triangular iridium lattice, exhibits a structural phase transition at approximately 250 K. This transition is characterized by the formation of Ir-Ir bonds along the b-axis. We found that the breaking of Ir-Ir bonds that occurs in Ir1-xPtxTe2 results in the appearance of a structural critical point in the T = 0 limit at xc = 0.035. Although both IrTe2 and PtTe2 are paramagnetic metals, superconductivity at Tc = 3.1 K is induced by the bond breaking in a narrow range of x > xc in Ir1-xPtxTe2. This result indicates that structural fluctuations can be involved in the emergence of superconductivity.Comment: 10 pages, 4 figure

    Optical coherence microscopy for the evaluation of a tissue-engineered artificial cornea

    A transparent artificial cornea derived from biological material is the ultimate goal of corneal research. Attempts at artificial corneal constructs produced from synthetic polymers have proved unsuccessful due to lack of biocompatibility and ability to integrate into the tissue. We have designed a corneal model derived from collagenous biological materials that has several advantages: it has low antigenicity and therefore small chance of eliciting an immune reaction, it can be broken down by the body’s own cells and gradually replaced over time by natural materials, and it may contain signaling information for native cells, thereby inducing normal phenotype and behavior. In addition, a transparent corneal model has the potential to be used for testing of novel ophthalmic drugs or gene therapy approaches, eliminating the need for animal testing. We have used an optical coherence microscope (OCM) to evaluate both the structure of our tissue constructs over time in culture and the optical properties of the tissue itself. This imaging technique promises to be an important diagnostic tool in our efforts to understand the influence of mechanical forces, cell phenotype, and soluble factors on the transparency of corneal tissue.     Melting mud in Earth's mantle

    Melting of subducted sediment remains controversial, as direct observation of sediment melt generation at mantle depths is not possible. Geochemical fingerprints provide indirect evidence for subduction delivery of sediment to the mantle; however, sediment abundance in mantle-derived melt is generally low (0%–2%), and difficult to detect. Here we provide evidence for melting of subducted sediment in granite sampled from an exhumed mantle section. Peraluminous granite dikes that intrude peridotite in the Oman–United Arab Emirates ophiolite have U-Pb ages of 99.8 ± 3.3 Ma that predate obduction. The dikes have unusually high oxygen isotope (δ18O) values for whole rock (14–23‰) and quartz (20–22‰), and yield the highest δ18O zircon values known (14–28‰; values relative to Vienna standard mean ocean water [VSMOW]). The extremely high oxygen isotope ratios uniquely identify the melt source as high-δ18O marine sediment (pelitic and/or siliciceous mud), as no other source could produce granite with such anomalously high δ18O. Formation of high-δ18O sediment-derived (S-type) granite within peridotite requires subduction of sediment to the mantle, where it melted and intruded overlying mantle wedge. The granite suite described here contains the highest oxygen isotope ratios reported for igneous rocks, yet intruded mantle peridotite below the Mohorovičić seismic discontinuity, the most primitive oxygen isotope reservoir in the silicate Earth. Identifying the presence and quantifying the extent of sediment melting within the mantle has important implications for understanding subduction recycling of supracrustal material and effects on mantle heterogeneity over time.National Geographi

    Composability in quantum cryptography

    In this article, we review several aspects of composability in the context of quantum cryptography. The first part is devoted to key distribution. We discuss the security criteria that a quantum key distribution protocol must fulfill to allow its safe use within a larger security application (e.g., for secure message transmission). To illustrate the practical use of composability, we show how to generate a continuous key stream by sequentially composing rounds of a quantum key distribution protocol. In a second part, we take a more general point of view, which is necessary for the study of cryptographic situations involving, for example, mutually distrustful parties.     Safety Data: Costs, Quality and Strategies for Improvement- Executive Summary

    FHWA-RD-96-027The objectives of this project were to examine the costs and quality of safety data and to identify improvement strategies. The project's scope was narrowed to three key sources: the crash report, roadway inventories, and medical records. The primary focus was on crash reporting. Within the medical source, the focus was upon data regarding driver condition prior to crash, and injury severity, since these are of high interest to the highway community. Since the same piece of data will have a different quality for different users and uses, it was necessary to identify the variety of users and uses of highway safety data. This executive summary presents an overview of the research results. The complete results are available in the research report (FHWA-RD-96-191) and the final report (FHWA-RD-96-192)
