    Finding a Disappearing Nontimber Forest Resource: Using Grounded Visualization to Explore Urbanization Impacts on Sweetgrass Basketmaking in Greater Mt. Pleasant, South Carolina

    Despite growing interest in urbanization and its social and ecological impacts on formerly rural areas, empirical research remains limited. Extant studies largely focus either on issues of social exclusion and enclosure or ecological change. This article uses the case of sweetgrass basketmaking in Mt. Pleasant, South Carolina, to explore the implications of urbanization, including gentrification, for the distribution and accessibility of sweetgrass, an economically important nontimber forest product (NTFP) for historically African American communities, in this rapidly growing area. We explore the usefulness of grounded visualization for research efforts that are examining the existence of fringe ecologies associated with NTFP. Our findings highlight the importance of integrated qualitative and quantitative analyses for revealing the complex social and ecological changes that accompany both urbanization and rural gentrification

    Robust Weak-lensing Mass Calibration of Planck Galaxy Clusters

    In light of the tension in cosmological constraints reported by the Planck team between their SZ-selected cluster counts and Cosmic Microwave Background (CMB) temperature anisotropies, we compare the Planck cluster mass estimates with robust, weak-lensing mass measurements from the Weighing the Giants (WtG) project. For the 22 clusters in common between the Planck cosmology sample and WtG, we find an overall mass ratio of \left = 0.688 \pm 0.072. Extending the sample to clusters not used in the Planck cosmology analysis yields a consistent value of <MPlanck/MWtG>=0.698±0.062\left< M_{Planck}/M_{\rm WtG} \right> = 0.698 \pm 0.062 from 38 clusters in common. Identifying the weak-lensing masses as proxies for the true cluster mass (on average), these ratios are 1.6σ\sim 1.6\sigma lower than the default mass bias of 0.8 assumed in the Planck cluster analysis. Adopting the WtG weak-lensing-based mass calibration would substantially reduce the tension found between the Planck cluster count cosmology results and those from CMB temperature anisotropies, thereby dispensing of the need for "new physics" such as uncomfortably large neutrino masses (in the context of the measured Planck temperature anisotropies and other data). We also find modest evidence (at 95 per cent confidence) for a mass dependence of the calibration ratio and discuss its potential origin in light of systematic uncertainties in the temperature calibration of the X-ray measurements used to calibrate the Planck cluster masses. Our results exemplify the critical role that robust absolute mass calibration plays in cluster cosmology, and the invaluable role of accurate weak-lensing mass measurements in this regard.Comment: 5 pages, 2 figure

    A comprehensive framework of E2–RING E3 interactions of the human ubiquitin–proteasome system

    Covalent attachment of ubiquitin to substrates is crucial to protein degradation, transcription regulation and cell signalling. Highly specific interactions between ubiquitin-conjugating enzymes (E2) and ubiquitin protein E3 ligases fulfil essential roles in this process. We performed a global yeast-two hybrid screen to study the specificity of interactions between catalytic domains of the 35 human E2s with 250 RING-type E3s. Our analysis showed over 300 high-quality interactions, uncovering a large fraction of new E2–E3 pairs. Both within the E2 and the E3 cohorts, several members were identified that are more versatile in their interaction behaviour than others. We also found that the physical interactions of our screen compare well with reported functional E2–E3 pairs in in vitro ubiquitination experiments. For validation we confirmed the interaction of several versatile E2s with E3s in in vitro protein interaction assays and we used mutagenesis to alter the E3 interactions of the E2 specific for K63 linkages, UBE2N(Ubc13), towards the K48-specific UBE2D2(UbcH5B).     A randomised controlled trial of a physical activity and nutrition program targeting middle-aged adults at risk of metabolic syndrome in a disadvantaged rural community

    Background: Approximately 70% of Australian adults aged over 50 are overweight or obese, with the prevalence significantly higher in regional/remote areas compared to cities. This study aims to determine if a low-cost, accessible lifestyle program targeting insufficiently active adults aged 50-69 y can be successfully implemented in a rural location, and whether its implementation will contribute to the reduction/prevention of metabolic syndrome, or other risk factors for type 2 diabetes, and cardiovascular disease.Methods/Design: This 6-month randomised controlled trial will consist of a nutrition, physical activity, and healthy weight intervention for 50–69 year-olds from a disadvantaged rural community. Five hundred participants with central obesity and at risk of metabolic syndrome will be recruited from Albany and surrounding areas in Western Australia (within a 50 kilometre radius of the town). They will be randomly assigned to either the intervention (n = 250) or wait-listed control group (n = 250). The theoretical concepts in the study utilise the Self-Determination Theory, complemented by Motivational Interviewing. The intervention will include a custom-designed booklet and interactive website that provides information, and encourages physical activity and nutrition goal setting, and healthy weight management. The booklet and website will be supplemented by an exercise chart, calendar, newsletters, resistance bands, accelerometers, and phone and email contact from program staff. Data will be collected at baseline and post-intervention.Discussion: This study aims to contribute to the prevention of metabolic syndrome and inter- related chronic illnesses: type 2 diabetes mellitus, cardiovascular disease, and some cancers; which are associated with overweight/obesity, physical inactivity, and poor diet. This large rural community-based trial will provide guidelines for recruitment, program development, implementation, and evaluation, and has the potential to translate findings into practice by expanding the program to other regional areas in Australia.
