Ending Drunk Driving at ECU
Ending Drunk Driving at ECU is a mission to inform, persuade and educate the population of ECU on the dangers of drinking and driving. We hope to make the Greenville community a safer environment by providing a wide array of educational opportunities to the students of East Carolina University. After multiple attempts at redesigning the Alcohol EDU course, Greek life education, and a student led driving service, we decided that education and social media would be the best product. We are currently working with the Health 1000 Education Department to create an informative video that will be incorporated into the new Health 1000 online curriculum. Our twitter page, @EDDECU, has gained a significant following. Our group also handed out red ribbons in April to bring awareness to National Distracted Driving Month
Quantifying engineered nanomaterial toxicity: comparison of common cytotoxicity and gene expression measurements
BACKGROUND: When evaluating the toxicity of engineered nanomaterials (ENMS) it is important to use multiple bioassays based on different mechanisms of action. In this regard we evaluated the use of gene expression and common cytotoxicity measurements using as test materials, two selected nanoparticles with known differences in toxicity, 5 nm mercaptoundecanoic acid (MUA)-capped InP and CdSe quantum dots (QDs). We tested the effects of these QDs at concentrations ranging from 0.5 to 160 µg/mL on cultured normal human bronchial epithelial (NHBE) cells using four common cytotoxicity assays: the dichlorofluorescein assay for reactive oxygen species (ROS), the lactate dehydrogenase assay for membrane viability (LDH), the mitochondrial dehydrogenase assay for mitochondrial function, and the Comet assay for DNA strand breaks. RESULTS: The cytotoxicity assays showed similar trends when exposed to nanoparticles for 24 h at 80 µg/mL with a threefold increase in ROS with exposure to CdSe QDs compared to an insignificant change in ROS levels after exposure to InP QDs, a twofold increase in the LDH necrosis assay in NHBE cells with exposure to CdSe QDs compared to a 50% decrease for InP QDs, a 60% decrease in the mitochondrial function assay upon exposure to CdSe QDs compared to a minimal increase in the case of InP and significant DNA strand breaks after exposure to CdSe QDs compared to no significant DNA strand breaks with InP. High-throughput quantitative real-time polymerase chain reaction (qRT-PCR) data for cells exposed for 6 h at a concentration of 80 µg/mL were consistent with the cytotoxicity assays showing major differences in DNA damage, DNA repair and mitochondrial function gene regulatory responses to the CdSe and InP QDs. The BRCA2, CYP1A1, CYP1B1, CDK1, SFN and VEGFA genes were observed to be upregulated specifically from increased CdSe exposure and suggests their possible utility as biomarkers for toxicity. Reliability and validity of the Healthy Home Survey: A tool to measure factors within homes hypothesized to relate to overweight in children
<p>Abstract</p> <p>Background</p> <p>The contribution of the environment to the obesity epidemic is well recognized. Parents have control over their home environment and can, therefore, support healthy dietary and activity habits in their children by manipulating factors such as access to energy-dense foods, availability of physical activity equipment, and restricting screen time. This paper describes the development of the Healthy Home Survey and its reliability and validity. The Healthy Home Survey was designed to assess characteristics of the home environment that are hypothesized to influence healthy weight behaviors in children including diet and physical activity.</p> <p>Methods</p> <p>We recruited 85 families with at least one child between 3–8 years. The Healthy Home Survey was administered to parents via telephone and repeated in a random sample of 45 families after 7 days. In-home observations were performed within 14 days of the first Healthy Home Survey interview. Percent agreement, Kappa statistics, Intra-class correlation coefficients and sensitivity analyses were used to evaluate reliability and validity evidence.</p> <p>Results</p> <p>Reliability and validity estimates for the Healthy Home Survey were varied, but generally high (0.22–1.00 and 0.07–0.96 respectively), with lower scores noted for perishable foods and policy items. Lower scores were likely related to actual change in the perishable foods present and the subjective nature or clarity of policy questions and response categories.</p> <p>Conclusion</p> <p>Initial testing demonstrated that the Healthy Home Survey is a feasible, reliable, and valid assessment of the home environment; however, it has also highlighted areas that need improvement. The Healthy Home Survey will be useful in future research exploring the relationship between the home environment and child weight.</p
Clover Care: Website and E-Marketing Development
Clover Care is a geographically diverse company with both caregivers and clients spread across multiple counties. Efficient resource management and communication is critical for such a broadly distributed company. Clover Care also expects to continue expanding its geographical service area, making a solution even more critical. Clover Care was interested in updating and expanding their existing website to address this need. The centralization provided by using a website allows caregivers, clients and their families, and managers a way to quickly and effectively communicate.
The primary Clover Care forms that were to be centralized included the caregiver timesheet, client case management form, and client invoice. Four distinct roles were identified as needing access to information: Caregiver, Client, Case Manager, and Administrator. Administrators required read and write capabilities for all objects. Case Managers required read capability for Caregiver timesheets and users, and write capability for case management forms. Caregivers required write capability for timesheets. Clients required read capability for timesheets, case management forms, and invoices.
Access to the secure forms was provided using a central logon page. Users were then taken to a dashboard that allowed them access to forms based on the user’s role. The user would then read from and/or write to a form based on the permissions associated with their role.
The website interface was developed using the PrimeFaces JSF library. Back-end functionality and security was provided using Spring and Hibernate. MySQL was used to store forms and user information for the purposes of development. This website is expected to be hosted using an Amazon hosting service (AWS), using their database service.
Once the web site is publicly hosted on AWS, statistics gathering and analysis will be performed to assist Clover Care in focusing on the geographical locations with the most usage. Mapping the Learning Pathways of Larval Zebrafish through Positively Stimulating Their Reward Pathways Using Paramecium
Zebrafish rely on visual acuity to hunt for prey all of their lives, for this reason, their eyesight develops when they are embryos. The zebrafish in this experiment are between 5 and 20 days old. Once their egg yolks are completely reabsorbed the zebrafish have a need to eat, and only then will they have an interest in eating Paramecium. The zebrafishes’ eyes will be able to see clearly by 5 days post fertilization as well, so when food is introduced to them, a certain colored light will be simultaneously shined in the same direction as the one the food is coming from.
The purpose of this experiment is to use different colored visual cues to train larval zebrafish into expecting food whenever they are shone. The usage of lights also tests their extraordinary visual abilities. Red lights will be associated with a food reward in one group of larvae, and in the second group, green lights will be associated with a food reward, demonstrating associative learning. The zebrafishes’ unconditioned response (eating Paramecium when they appear) will be trained into a conditioned response (looking to eat Paramecium when the light is shined). After they have been conditioned to respond to these signals, the zebrafishes’ brains will be studied to find changes in their neural pathways.

Contrast Enhanced-Magnetic Resonance Imaging as a Surrogate to Map Verteporfin Delivery in Photodynamic Therapy
The use of in vivo contrast-enhanced magnetic resonance (MR) imaging as a surrogate for photosensitizer (verteporfin) dosimetry in photodynamic therapy of pancreas cancer is demonstrated by correlating MR contrast uptake to ex vivo fluorescence images on excised tissue. An orthotopic pancreatic xenograft mouse model was used for the study. A strong correlation ([i]r=0.57 ) was found for bulk intensity measurements of T1-weighted gadolinium enhancement and verteporfin fluorescence in the tumor region of interest. The use of contrast-enhanced MR imaging shows promise as a method for treatment planning and photosensitizer dosimetry in human photodynamic therapy (PDT) of pancreas cancer

Combination Therapies with CDK4/6 Inhibitors to Treat KRAS-Mutant Pancreatic Cancer.
Mutational loss of CDKN2A (encoding p16INK4A) tumor-suppressor function is a key genetic step that complements activation of KRAS in promoting the development and malignant growth of pancreatic ductal adenocarcinoma (PDAC). However, pharmacologic restoration of p16INK4A function with inhibitors of CDK4 and CDK6 (CDK4/6) has shown limited clinical efficacy in PDAC. Here, we found that concurrent treatment with both a CDK4/6 inhibitor (CDK4/6i) and an ERK-MAPK inhibitor (ERKi) synergistically suppresses the growth of PDAC cell lines and organoids by cooperatively blocking CDK4/6i-induced compensatory upregulation of ERK, PI3K, antiapoptotic signaling, and MYC expression. On the basis of these findings, a Phase I clinical trial was initiated to evaluate the ERKi ulixertinib in combination with the CDK4/6i palbociclib in patients with advanced PDAC (NCT03454035). As inhibition of other proteins might also counter CDK4/6i-mediated signaling changes to increase cellular CDK4/6i sensitivity, a CRISPR-Cas9 loss-of-function screen was conducted that revealed a spectrum of functionally diverse genes whose loss enhanced CDK4/6i growth inhibitory activity. These genes were enriched around diverse signaling nodes, including cell-cycle regulatory proteins centered on CDK2 activation, PI3K-AKT-mTOR signaling, SRC family kinases, HDAC proteins, autophagy-activating pathways, chromosome regulation and maintenance, and DNA damage and repair pathways. Novel therapeutic combinations were validated using siRNA and small-molecule inhibitor-based approaches. In addition, genes whose loss imparts a survival advantage were identified (e.g., RB1, PTEN, FBXW7), suggesting possible resistance mechanisms to CDK4/6 inhibition. In summary, this study has identified novel combinations with CDK4/6i that may have clinical benefit to patients with PDAC.
SIGNIFICANCE: CRISPR-Cas9 screening and protein activity mapping reveal combinations that increase potency of CDK4/6 inhibitors and overcome drug-induced compensations in pancreatic cancer
CHK1 protects oncogenic KRAS-expressing cells from DNA damage and is a target for pancreatic cancer treatment
We apply genetic screens to delineate modulators of KRAS mutant pancreatic ductal adenocarcinoma (PDAC) sensitivity to ERK inhibitor treatment, and we identify components of the ATR-CHK1 DNA damage repair (DDR) pathway. Pharmacologic inhibition of CHK1 alone causes apoptotic growth suppression of both PDAC cell lines and organoids, which correlates with loss of MYC expression. CHK1 inhibition also activates ERK and AMPK and increases autophagy, providing a mechanistic basis for increased efficacy of concurrent CHK1 and ERK inhibition and/or autophagy inhibition with chloroquine. To assess how CHK1 inhibition-induced ERK activation promotes PDAC survival, we perform a CRISPR-Cas9 loss-of-function screen targeting direct/indirect ERK substrates and identify RIF1. A key component of non-homologous end joining repair, RIF1 suppression sensitizes PDAC cells to CHK1 inhibition-mediated apoptotic growth suppression. Furthermore, ERK inhibition alone decreases RIF1 expression and phenocopies RIF1 depletion. We conclude that concurrent DDR suppression enhances the efficacy of ERK and/or autophagy inhibitors in KRAS mutant PDAC
