    A Functional Analysis of 2013 Australian Member of Parliament and Prime Minister Debates

    Kevin Rudd (Labor) engaged in two election debates in 2013: first for his seat in Parliament (Griffith) and then for Prime Minister. In seeking re-election to his Griffith seat he was challenged by Bill Glasson (Liberal), Geoff Ebbs (Green), and Karen Hunter (Palmer United). He faced Tony Abbott (Liberal) in the PM Debate. This study content analyses these two debates: an August 6, 2013 Member of Parliament debate and an August 11, 2013 Prime Minister debate. Acclaims were more common in these two debates than attacks; defenses were the least common function. Statements about policy outnumbered those on policy. In each debate, when the candidates discussed record in office (past deeds), the incumbent acclaimed more and attacked less than the challenger. More acclaims and fewer attacks addressed general goals and ideals

    Reaction of Group III Biheterocyclic Complexes

    Group III alkyl complexes supported by a ferrocene diamide ligand (1,1′-fc(NSitBuMe_2)_2) have been found to be reactive toward aromatic N-heterocycles such as 1-methylimidazole and pyridines. These reactions were investigated experimentally and computationally. An initial C−H activation event is followed by a coupling reaction to form biheterocyclic complexes, in which one of the rings is dearomatized. In the case of 1-methylimidazole, the biheterocyclic compound could not be isolated and further led to an imidazole ring-opened product; in the case of pyridines, it transformed into an isomer with extended conjugation of double bonds. Mechanisms for both reactions are proposed on the basis of experimental and computational results. DFT calculations were also used to show that an energetically accessible pathway for the ring-opening of pyridines exists

    Re-evaluating the role of natural killer cells in innate resistance to herpes simplex virus type 1

    BACKGROUND: Interferon-γ acts to multiply the potency with which innate interferons (α/β) suppress herpes simplex virus type 1 (HSV-1) replication. Recent evidence suggests that this interaction is functionally relevant in host defense against HSV-1. However, it is not clear which WBCs of the innate immune system, if any, limit HSV-1 spread in an IFN-γ dependent manner. The current study was initiated to determine if natural killer (NK) cells provide innate resistance to HSV-1 infection, and if so to determine if this resistance is IFN-γ-dependent. RESULTS: Lymphocyte-deficient scid or rag2(-/- )mice were used to test four predictions of the central hypothesis, and thus determine if innate resistance to HSV-1 is dependent on 1. NK cell cytotoxicity, 2. NK cells, 3. WBCs, or 4. the IFN-activated transcription factor, Stat 1. Loss of NK cell cytotoxic function or depletion of NK cells had no effect on the progression of HSV-1 infection in scid mice. In contrast, viral spread and pathogenesis developed much more rapidly in scid mice depleted of WBCs. Likewise, loss of Stat 1 function profoundly impaired the innate resistance of rag2(-/- )mice to HSV-1. CONCLUSION: Lymphocyte-deficient mice possess a very tangible innate resistance to HSV-1 infection, but this resistance is not dependent upon NK cells

    The comparative cytotoxicity of riddelliine in primary mouse, rat and chick hepatocytes

    Dehydropyrrolizidine alkaloid (DHPA) producing plants commonly poison livestock, wildlife and humans. Poisoning occurs when DHPAs are ingested as feed or food, or when they contaminate medicinal or herbal products. Direct toxicologic comparison of individual DHPAs is essential to estimate their actual health risks. This has been problematic due to varying models and difficulties in DHPA isolation or synthesis. In contrast, the macrocyclic DHPA riddelliine is readily isolated and it has been used as a benchmark to characterize different models of toxicity and carcinogenicity. Following earlier work with immortalized cell lines, the objective of this study was to characterize the effect of riddelliine on primary mouse, rat and chick hepatocyte cultures with the aim of developing a suitable, sensitive model for assessing DHPA-related cytotoxicity. After establishing viable cultures, the hepatocytes were exposed for 24 hours to riddelliine (from 0.1µM to 1.2mM) and cytotoxicity (CT­­50) was estimated using a mitochondrial function assay (MTT). Despite a biphasic response, possibly attributable to a sub-population of resistant chick hepatocytes, chick hepatocyte cultures were highly sensitive (CT50 0.9 µM) to riddelliine cytotoxicity relative to rat (CT50 289 µM) and mouse (CT50 627 µM) hepatocytes. Chick, mouse and rat hepatocyte cytochrome P450 3A4 activities did not correlate with riddelliine-induced cytotoxicity. With further development to utilize the highly sensitive primary chick hepatocytes, this model may be useful to directly compare panels of DHPAs, including rare or difficult to isolate alkaloids

    Distance Education of Pennsylvania Pond Owners

    Pennsylvania pond owners represent a large audience with a great need for educational programming and assistance. Penn State Extension attempted to reach this audience via a pond management program delivered by satellite. Evaluations by both program attendees and Extension professionals demonstrated that the program was successful. However, attendee respondents indicated that they wanted more in-depth information over a shorter time frame. A relatively small proportion (10%) of attendees did not favor satellite delivery. This level of dissatisfaction is quite low and may be improved by focusing on shorter, more focused satellite programs and by providing videotapes of satellite programs

    Secondary succession in an experimentally fragmented landscape: community patterns across space and time

    Secondary succession reflects, at least in part, community assembly—the sequences of colonizations and extinctions. These processes in turn are expected to be sensitive to the size of the site undergoing assembly and its location relative to source pools. In this paper we describe patterns of succession over 18 years in an experimentally fragmented landscape created in eastern Kansas, USA, in 1984. The design of the experiment permits one to assess the influence of patch size and landscape position on successional dynamics. The general trajectory of succession follows that typical of succession in much of the eastern United States. In the initial years of the study, there was relatively little effect of patch size or distance to sources. Here we show that spatial effects in this system have become increasingly evident with time, as gauged both by repeated-measures ANOVA and ordination techniques. Woody plants have colonized more rapidly (per unit area) on large and nearby patches. Species richness at a local (within-quadrat) scale in general has increased, with slightly greater richness in large than in small patches later in the study. Temporal stability in community composition has generally been greater in large patches. Spatial heterogeneity in community composition has increased during succession, but with different patterns in large and small patches. This long-term experiment suggests that landscape structure influences many aspects of community structure and dynamics during succession, and that such effects become more pronounced with the passage of time
