    Pesticide Product Use and Risk of Non-Hodgkin Lymphoma in Women

    A population-based, incidence caseā€“control study was conducted among women in upstate New York to determine whether pesticide exposure is associated with an increase in risk of non-Hodgkin lymphoma (NHL) among women. The study involved 376 cases of NHL identified through the State Cancer Registry and 463 controls selected from the Medicare beneficiary files and state driverā€™s license records. Information about history of farm work, history of other jobs associated with pesticide exposure, use of common household pesticide products, and potential confounding variables was obtained by telephone interview. Odds ratios (ORs) and 95% confidence intervals (CIs) were estimated using an unconditional logistic regression model. The risk of NHL was doubled (OR = 2.12; 95% CI, 1.21ā€“3.71) among women who worked for at least 10 years at a farm where pesticides were reportedly used. When both farming and other types of jobs associated with pesticide exposure were combined, there was a progressive increase in risk of NHL with increasing duration of such work (p = 0.005). Overall cumulative frequency of use of household pesticide products was positively associated with risk of NHL (p = 0.004), which was most pronounced when they were applied by subjects themselves. When exposure was analyzed by type of products used, a significant association was observed for mothballs. The associations with both occupational and household pesticides were particularly elevated if exposure started in 1950ā€“1969 and for high-grade NHL. Although the results of this caseā€“control study suggest that exposure to pesticide products may be associated with an increased risk of NHL among women, methodologic limitations related to selection and recall bias suggest caution in inferring causation

    Toksikokinetika prometrina u mozgu miŔeva

    Prometryne is a methylthio-s-triazine herbicide. Signifi cant trace amounts are found in the environment, mainly in water, soil, and food plants. The aim of this study was to establish brain and blood prometryne levels after single oral dose (1 g kg-1) in adult male and female mice. Prometryne was measured using the GC/MS assay at 1, 2, 4, 8, and 24 h after prometryne administration. Peak brain and blood prometryne values were observed 1 h after administration and they decreased in a time-dependent manner. Male mice had consistently higher brain and blood prometryne levels than female mice. The observed prometryne kinetics was similar to that reported for the structurally related herbicide atrazine.Prometrin je metiltio-s-triazinski herbicid. Značajne količine prometrina zaostaju u tragovima u okoliÅ”u, poglavito u vodi, tlu i biljkama koje rabimo za prehranu. Cilj je rada izmjeriti količinu prometrina koja se apsorbira u mozgu i krvi nakon primijenjene akutne oralne doze (1 g kg-1 tjelesne mase) u odraslih miÅ”eva obaju spolova. Razine prometrina u mozgu i krvi izmjerene su GC/MS-om tijekom 1., 2., 4., 8. i 24. sata nakon izlaganja. Utvrđeno je da je udio prometrina koji se zadržava u živčanom tkivu relativno nizak ali detektabilan u odnosu na koncentraciju u krvi i koncentraciju primijenjene doze. NajviÅ”e koncentracije u krvi i maseni udjeli u mozgu zabilježeni su tijekom 1. sata nakon izlaganja, a s vremenom izmjerene vrijednosti značajno opadaju. Uočena je značajna razlika između mužjaka i ženki pri čemu mužjaci imaju značajno viÅ”e razine prometrina u mozgu i krvi nego ženke.     Bioactivation of dimethylnitrosamine in intrasanguinous host-mediated assay and its association with in vitro mutagenesis assays

    These studies have revealed the usefulness of in vivo intrasanguine host-mediated assay (HMA) to detect point mutations. Mutations were found to occur at a significant rate in Salmonella typhimurium G-46 employed as indicator organisms recovered from liver, lung, kidney and spleen of DMN-treated animals compared to negative control animals. These differences were true for both male and female animals. The number of Salmonella typhimurium G-46 recovered from the testes was not large enough to make a valid judgement about mutations occurring in testes. The results from in vitro studies do not match with the in vivo host-mediated assay results for mutants occurring in spleen from the male and female mice. The results also do not correlate for in vitro and in vivo studies involving female kidneys. These results suggest there may be no one-to-one correlation between the organ bioactivation in vitro and in vivo, and predictions of in vivo target organ cannot always be made from in vitro studies with isolated microsomal enzymes.
