    Efficiency Options for a Focus Company: Comparison and Analysis of Potential Industrial Ecology and In-House Process Change Initiatives

    Industrial ecology is a popular option in the literature for improving the efficiency of industrial facilities, most notably through the eco-industrial park (EIP) concept. Relatively little attention has been paid to overall efficiencies achievable at the local factory scale through in-house process changes (IHPCs) - efficiency improvements that are undertaken in a factory to improve their processes, without the aid of their neighbours. This thesis compares the EIP model to IHPCs using a case study of an existing factory (the focus company) in an industrialized zone and answers the question: what are the drivers, barriers, and opportunities to encourage a promising company to participate in industrial ecology in an existing industrial cluster and how does the industrial ecology option compare with more conventional in-house environmental efficiency opportunities? An analysis of the focus company’s major operating parameters, as well as data from interviews with the focus company, the local municipality, and the local utility provider are used to compare what the literature says about the implementation of industrial ecology in a case study setting to the results found through this study. The results from this study confirm a number of the barriers to implementing industrial ecology as found in the literature, and further emphasize that improved education and a more grass roots, consultant-led committee for the local industrial site would help with the implementation of industrial ecology principles. In a general sense, EIP projects are on a larger scale than IHPCs and therefore capable of greater efficiency improvements, whereas IHPCs are easier to manage due to being internally managed initiatives. It was found that for this particular case study, EIP options seem to be limited to a collaborative energy production plan, whereas a structured IHPC plan is best for the focus company’s continued efficiency improvement. It was also found that the focus company has begun to tackle the higher hanging fruit in terms of energy efficiency, while leaving the comparably easily accessible lower hanging fruit options untouched. This leaves a myriad of low and/or no cost options to further improve the efficiency of the facility

    Preliminary study of the oral mycobiome of children with and without dental caries

    Children’s oral health is in a dire state, with dental decay (caries) being one of the most common chronic diseases. While the role of bacteria in the oral microbiome and dental caries is established, the contribution of fungi is relatively unknown. We assessed the oral mycobiome in childhood (n = 17), to determine if the composition of fungi varies between children with and without caries. Oral mycobiome composition was assessed by using Illumina MiSeq to sequence the ITS2 region, which was amplified from dental plaque. This revealed that the oral mycobiome in the investigated children contained 46 fungal species. Candida albicans was the most abundant species and was ubiquitous in all samples, indicating this species may not be involved in caries development as previously suggested. While the overall diversity of fungi was similar, independent of caries status (p > 0.05), we found caries influenced the abundance of specific fungi. Children without caries had a significantly higher abundance of 17 species compared to children with caries, which had three enriched species (p < 0.001). While the differentially abundant species between health and caries may be specific to an Australian population, our findings indicate the mycobiome plays a role in oral health.Jacquelyn M.     The geometry of r-adaptive meshes generated using optimal transport methods

    The principles of mesh equidistribution and alignment play a fundamental role in the design of adaptive methods, and a metric tensor M and mesh metric are useful theoretical tools for understanding a methods level of mesh alignment, or anisotropy. We consider a mesh redistribution method based on the Monge-Ampere equation, which combines equidistribution of a given scalar density function with optimal transport. It does not involve explicit use of a metric tensor M, although such a tensor must exist for the method, and an interesting question to ask is whether or not the alignment produced by the metric gives an anisotropic mesh. For model problems with a linear feature and with a radially symmetric feature, we derive the exact form of the metric M, which involves expressions for its eigenvalues and eigenvectors. The eigenvectors are shown to be orthogonal and tangential to the feature, and the ratio of the eigenvalues (corresponding to the level of anisotropy) is shown to depend, both locally and globally, on the value of the density function and the amount of curvature. We thereby demonstrate how the optimal transport method produces an anisotropic mesh along a given feature while equidistributing a suitably chosen scalar density function. Numerical results are given to verify these results and to demonstrate how the analysis is useful for problems involving more complex features, including for a non-trivial time dependant nonlinear PDE which evolves narrow and curved reaction fronts

    Development of the oral resistome during the first decade of life

    Published online: 09 March 2023Antibiotic overuse has promoted the spread of antimicrobial resistance (AMR) with significant health and economic consequences. Genome sequencing reveals the widespread presence of antimicrobial resistance genes (ARGs) in diverse microbial environments. Hence, surveillance of resistance reservoirs, like the rarely explored oral microbiome, is necessary to combat AMR. Here, we characterise the development of the paediatric oral resistome and investigate its role in dental caries in 221 twin children (124 females and 97 males) sampled at three time points over the first decade of life. From 530 oral metagenomes, we identify 309 ARGs, which significantly cluster by age, with host genetic effects detected from infancy onwards. Our results suggest potential mobilisation of ARGs increases with age as the AMR associated mobile genetic element, Tn916 transposase was co-located with more species and ARGs in older children. We find a depletion of ARGs and species in dental caries compared to health. This trend reverses in restored teeth. Here we show the paediatric oral resistome is an inherent and dynamic component of the oral microbiome, with a potential role in transmission of AMR and dysbiosis.Smitha Sukumar, FangWang, Carra A.     A secreted PD-L1 splice variant that covalently dimerizes and mediates immunosuppression

    Targeting immune checkpoint pathways, such as programmed death ligand-1 (PD-L1, also known as CD274 or B7-H1) or its receptor programmed cell death-1 (PD-1) has shown improved survival for patients with numerous types of cancers, not limited to lung cancer, melanoma, renal cell carcinoma, and Hodgkin lymphoma. PD-L1 is a co-inhibitory molecule whose expression on the surface of tumor cells is associated with worse prognosis in many tumors. Here we describe a splice variant (secPD-L1) that does not splice into the transmembrane domain, but instead produces a secreted form of PD-L1 that has a unique 18 amino acid tail containing a cysteine that allows it to homodimerize and more effectively inhibit lymphocyte function than monomeric soluble PD-L1. We show that recombinant secPD-L1 can dimerize and inhibit T-cell proliferation and IFN-gamma production in vitro. The secPD-L1 variant is expressed by malignant cells in vitro that also express high levels of full-length PD-L1. Transcriptomic analysis of gene expression across The Cancer Genome Atlas found the strongest association of secPD-L1 with full-length PD-L1, but also with subsets of immunologic genes, such as in myeloid-derived suppressor cells. Moreover, the splice variant is also expressed in normal tissues and within normal peripheral blood cells it is preferentially expressed in activated myeloid cells. This is the first report of a form of secreted PD-L1 that homodimerizes and is functionally active. SecPD-L1 may function as a paracrine negative immune regulator within the tumor, since secPD-L1 does not require a cell-to-cell interaction to mediate its inhibitory effect

    Horizontal Branch Stars: The Interplay between Observations and Theory, and Insights into the Formation of the Galaxy

    We review HB stars in a broad astrophysical context, including both variable and non-variable stars. A reassessment of the Oosterhoff dichotomy is presented, which provides unprecedented detail regarding its origin and systematics. We show that the Oosterhoff dichotomy and the distribution of globular clusters (GCs) in the HB morphology-metallicity plane both exclude, with high statistical significance, the possibility that the Galactic halo may have formed from the accretion of dwarf galaxies resembling present-day Milky Way satellites such as Fornax, Sagittarius, and the LMC. A rediscussion of the second-parameter problem is presented. A technique is proposed to estimate the HB types of extragalactic GCs on the basis of integrated far-UV photometry. The relationship between the absolute V magnitude of the HB at the RR Lyrae level and metallicity, as obtained on the basis of trigonometric parallax measurements for the star RR Lyrae, is also revisited, giving a distance modulus to the LMC of (m-M)_0 = 18.44+/-0.11.     Exome sequencing identifies genetic variants in anophthalmia and microphthalmia

    Anophthalmia and microphthalmia (A/M) are rare birth defects affecting up to 2 per 10,000 live births. These conditions are manifested by the absence of an eye or reduced eye volumes within the orbit leading to vision loss. Although clinical case series suggest a strong genetic component in A/M, few systematic investigations have been conducted on potential genetic contributions owing to low population prevalence. To overcome this challenge, we utilized DNA samples and data collected as part of the National Birth Defects Prevention Study (NBDPS). The NBDPS employed multi-center ascertainment of infants affected by A/M. We performed exome sequencing on 67 family trios and identified numerous genes affected by rare deleterious nonsense and missense variants in this cohort, including de novo variants. We identified 9 nonsense changes and 86 missense variants that are absent from the reference human population (Genome Aggregation Database), and we suggest that these are high priority candidate genes for A/M. We also performed literature curation, single cell transcriptome comparisons, and molecular pathway analysis on the candidate genes and performed protein structure modeling to determine the potential pathogenic variant consequences on PAX6 in this disease

    The Sudbury Neutrino Observatory

    The Sudbury Neutrino Observatory is a second generation water Cherenkov detector designed to determine whether the currently observed solar neutrino deficit is a result of neutrino oscillations. The detector is unique in its use of D2O as a detection medium, permitting it to make a solar model-independent test of the neutrino oscillation hypothesis by comparison of the charged- and neutral-current interaction rates. In this paper the physical properties, construction, and preliminary operation of the Sudbury Neutrino Observatory are described.
