    Using Unoccupied Aerial Vehicles to estimate availability and group size error for aerial surveys of coastal dolphins

    Aerial surveys are frequently used to estimate the abundance of marine mammals, but their accuracy is dependent upon obtaining a measure of the availability of animals to visual detection. Existing methods for characterizing availability have limitations and do not necessarily reflect true availability. Here, we present a method of using small, vessel-launched, multi-rotor Unoccupied Aerial Vehicles (UAVs, or drones) to collect video of dolphins to characterize availability and investigate error surrounding group size estimates. We collected over 20 h of aerial video of dive-surfacing behaviour across 32 encounters with Australian humpback dolphins Sousa sahulensis off north-western Australia. Mean surfacing and dive periods were 7.85 sec (se = 0.26) and 39.27 sec (se = 1.31) respectively. Dolphin encounters were split into 56 focal follows of consistent group composition to which example approaches to estimating availability were applied. Non-instantaneous availability estimates, assuming a 7 sec observation window, ranged between 0.22 and 0.88, with a mean availability of 0.46 (CV = 0.34). Availability tended to increase with increasing group size. We found a downward bias in group size estimation, with true group size typically one individual more than would have been estimated by a human observer during a standard aerial survey. The variability of availability estimates between focal follows highlights the importance of sampling across a variety of group sizes, compositions and environmental conditions. Through data re-sampling exercises, we explored the influence of sample size on availability estimates and their precision, with results providing an indication of target sample sizes to minimize bias in future research. We show that UAVs can provide an effective and relatively inexpensive method of characterizing dolphin availability with several advantages over existing approaches. The example estimates obtained for humpback dolphins are within the range of values obtained for other shallow-water, small cetaceans, and will directly inform a government-run program of aerial surveys in the region

    Filaggrin-stratified transcriptomic analysis of pediatric skin identifies mechanistic pathways in patients with atopic dermatitis

    BackgroundAtopic dermatitis (AD; eczema) is characterized by a widespread abnormality in cutaneous barrier function and propensity to inflammation. Filaggrin is a multifunctional protein and plays a key role in skin barrier formation. Loss-of-function mutations in the gene encoding filaggrin (FLG) are a highly significant risk factor for atopic disease, but the molecular mechanisms leading to dermatitis remain unclear.ObjectiveWe sought to interrogate tissue-specific variations in the expressed genome in the skin of children with AD and to investigate underlying pathomechanisms in atopic skin.MethodsWe applied single-molecule direct RNA sequencing to analyze the whole transcriptome using minimal tissue samples. Uninvolved skin biopsy specimens from 26 pediatric patients with AD were compared with site-matched samples from 10 nonatopic teenage control subjects. Cases and control subjects were screened for FLG genotype to stratify the data set.ResultsTwo thousand four hundred thirty differentially expressed genes (false discovery rate, P < .05) were identified, of which 211 were significantly upregulated and 490 downregulated by greater than 2-fold. Gene ontology terms for “extracellular space” and “defense response” were enriched, whereas “lipid metabolic processes” were downregulated. The subset of FLG wild-type cases showed dysregulation of genes involved with lipid metabolism, whereas filaggrin haploinsufficiency affected global gene expression and was characterized by a type 1 interferon–mediated stress response.ConclusionThese analyses demonstrate the importance of extracellular space and lipid metabolism in atopic skin pathology independent of FLG genotype, whereas an aberrant defense response is seen in subjects with FLG mutations. Genotype stratification of the large data set has facilitated functional interpretation and might guide future therapy development

    Surface plasmon resonance study of the actin-myosin sarcomeric complex and tubulin dimers

    Biosensors based on the principle of surface plasmon resonance (SPR) detection were used to measure biomolecular interactions in sarcomeres and changes of the dielectric constant of tubulin samples with varying concentration. At SPR, photons of laser light efficiently excite surface plasmons propagating along a metal (gold) film. This resonance manifests itself as a sharp minimum in the reflection of the incident laser light and occurs at a characteristic angle. The dependence of the SPR angle on the dielectric permittivity of the sample medium adjacent to the gold film allows the monitoring of molecular interactions at the surface.     Dapagliflozin, Inflammation and Left Ventricular Remodelling in Patients with Type 2 Diabetes and Left Ventricular Hypertrophy

    Background and AimsSodium-glucose co-transporter 2 (SGLT2) inhibitors have beneficial effects in heart failure (HF), including reverse remodelling, but the mechanisms by which these benefits are conferred are unclear. Inflammation is implicated in the pathophysiology of heart failure (HF) and there are some pre-clinical data suggesting that SGLT2 inhibitors may reduce inflammation. There is however a lack of clinical data. The aim of our study was to investigate whether improvements in cardiac remodelling caused by dapagliflozin in individuals with type 2 diabetes (T2D) and left ventricular hypertrophy (LVH) were associated with its effects on inflammation.MethodsWe measured C-reactive protein (CRP), tumor necrosis factor alpha (TNF-α), interleukin-1ÎČ (IL-1ÎČ), interleukin 6 (IL-6), and interleukin 10 (IL-10) and neutrophil-to-lymphocyte ratio (NLR) in plasma samples of 60 patients with T2D and left ventricular hypertrophy (LVH) but without symptomatic HF from the DAPA-LVH trial in which participants were randomised dapagliflozin 10mg daily or placebo for 12 months and underwent cardiac magnetic resonance imaging (CMR) at baseline and end of treatment. The primary analysis was to investigate the effect of dapagliflozin on inflammation and to assess the relationships between changes in inflammatory markers and LV mass and global longitudinal strain (GLS) and whether the effect of dapagliflozin on LV mass and GLS was modulated by baseline levels of inflammation.ResultsFollowing 12 months of treatment dapagliflozin significantly reduced CRP compared to placebo (mean difference of -1.96; 95% CI -3.68 to -0.24, p=0.026). There were no significant statistical changes in other inflammatory markers. There were modest correlations between improvements in GLS and reduced inflammation (NLR (r=0.311), IL-1ÎČ (r=0.246), TNF-α (r=0.230)) at 12 months.ConclusionsDapagliflozin caused a significant reduction in CRP compared to placebo. There were correlations between reductions in inflammatory markers including IL-1ÎČ and improvements in global longitudinal strain (but not reduced LV mass). Reductions in systemic inflammation might play a contributory role in the cardiovascular benefits of dapagliflozin

    Regularized energy-dependent solar flare hard x-ray spectral index

    The deduction from solar flare X-ray photon spectroscopic data of the energy dependent model-independent spectral index is considered as an inverse problem. Using the well developed regularization approach we analyze the energy dependency of spectral index for a high resolution energy spectrum provided by Ramaty High Energy Solar Spectroscopic Imager (RHESSI). The regularization technique produces much smoother derivatives while avoiding additional errors typical of finite differences. It is shown that observations imply a spectral index varying significantly with energy, in a way that also varies with time as the flare progresses.     CARMA CO(J = 2 - 1) Observations of the Circumstellar Envelope of Betelgeuse

    We report radio interferometric observations of the 12C16O 1.3 mm J = 2-1 emission line in the circumstellar envelope of the M supergiant Alpha Ori and have detected and separated both the S1 and S2 flow components for the first time. Observations were made with the Combined Array for Research in Millimeter-wave Astronomy (CARMA) interferometer in the C, D, and E antenna configurations. We obtain good u-v coverage (5-280 klambda) by combining data from all three configurations allowing us to trace spatial scales as small as 0.9\arcsec over a 32\arcsec field of view. The high spectral and spatial resolution C configuration line profile shows that the inner S1 flow has slightly asymmetric outflow velocities ranging from -9.0 km s-1 to +10.6 km s-1 with respect to the stellar rest frame. We find little evidence for the outer S2 flow in this configuration because the majority of this emission has been spatially-filtered (resolved out) by the array. We also report a SOFIA-GREAT CO(J= 12-11) emission line profile which we associate with this inner higher excitation S1 flow. The outer S2 flow appears in the D and E configuration maps and its outflow velocity is found to be in good agreement with high resolution optical spectroscopy of K I obtained at the McDonald Observatory. We image both S1 and S2 in the multi-configuration maps and see a gradual change in the angular size of the emission in the high absolute velocity maps. We assign an outer radius of 4\arcsec to S1 and propose that S2 extends beyond CARMA's field of view (32\arcsec at 1.3 mm) out to a radius of 17\arcsec which is larger than recent single-dish observations have indicated. When azimuthally averaged, the intensity fall-off for both flows is found to be proportional to R^{-1}, where R is the projected radius, indicating optically thin winds with \rho \propto R^{-2}.Comment: 11 pages, 8 figures To be published in the Astronomical Journal (Received 2012 February 10; accepted 2012 May 25
