    Numerical investigations on scour and flow around two crossing pipelines on a sandy seabed

    When a pipeline is laid on the seabed, local scour often occurs below it due to sea currents. In practical engineering, there are some cases that two pipelines laid on the seabed need to cross with each other. The complex flow structures around two crossing pipelines make the scour characteristics different from that of an isolated single pipeline. In this study, scour below two crossing pipelines was simulated numerically using the CFD software Flow-3D. The study is focused on the effect of the intersecting angle on the equilibrium depth and time scale of scour below the crossing position. Five intersecting angles, i.e., α = 0◦ , 15◦ , 30◦ , 45◦ and 90◦ , are considered, where α = 0◦ and 90◦ represent two pipelines parallel and perpendicular to each other, respectively. The results show that the equilibrium depth and the time scale of scour below the two crossing pipelines are greater than those of an isolated single pipeline. The equilibrium depth and time scale of scour have the largest values at α = 0◦ and decrease with the increase of the intersecting angle. Finally, the flow structures around the crossing pipelines are presented to explain the scour process

    Members of miR-169 family are induced by high salinity and transiently inhibit the NF-YA transcription factor

    <p>Abstract</p> <p>Background</p> <p>MicroRNAs (miRNAs) are endogenously expressed small RNAs with a length of about 21 nt. MiRNAs silence their target genes at the post-transcriptional level. In plants, miRNAs play various developmental and physiological roles by cleavaging mRNAs predominantly. Drought and high salinity are the most severe environmental abiotic stresses and cause crop losses all over the world.</p> <p>Results</p> <p>In this study, we identified miR-169g and miR-169n (o) as high salinity-responsive miRNAs in rice. MiR-169n and miR169o were in a miRNA cluster with a distance of 3707 base pairs (bp). The high degree of conservation and close phylogenic distance of pre-miR-169n and pre-miR-169o indicated that they were derived from a very recent tandem duplication evolutionary event. The existence of a cis-acting abscisic acid responsive element (ABRE) in the upstream region of miR-169n (o) suggested that miR-169n (o) may be regulated by ABA. In our previous study, we found that miR-169g was induced by the osmotic stress caused by drought via a dehydration-responsive element (DRE). Thus, our data showed that there were both overlapping and distinct responses of the miR-169 family to drought and salt stresses. We also showed that these miR-169 members selectively cleaved one of the NF-YA genes, Os03g29760, which is a CCAAT-box binding transcription factor and participates in transcriptional regulation of large number genes. Finally, we found one or more ath-miR-169 member that was also induced by high salinity.</p> <p>Conclusion</p> <p>We identified members of the miR-169 family as salt-induced miRNAs and analyzed their evolution, gene organization, expression, transcriptional regulation motif and target gene. Our data also indicated that the salt-induction of some miR-169 members was a general property in plants.</p

    miR-24 Regulates Apoptosis by Targeting the Open Reading Frame (ORF) Region of FAF1 in Cancer Cells

    BACKGROUND: microRNAs (miRNAs) are small noncoding RNAs that regulate cognate mRNAs at the post-transcriptional stage. Several studies have shown that miRNAs modulate gene expression in mammalian cells by base pairing to complementary sites in the 3'-untranslated region (3'-UTR) of the target mRNAs. METHODOLOGY/PRINCIPAL FINDINGS: In the present study, miR-24 was found to target fas associated factor 1(FAF1) by binding to its amino acid coding sequence (CDS) region, thereby regulating apoptosis in DU-145 cells. This result supports an augmented model whereby animal miRNAs can exercise their effects through binding to the CDS region of the target mRNA. Transfection of miR-24 antisense oligonucleotide (miR-24-ASO) also induced apoptosis in HGC-27, MGC-803 and HeLa cells. CONCLUSIONS/SIGNIFICANCE: We found that miR-24 regulates apoptosis by targeting FAF1 in cancer cells. These findings suggest that miR-24 could be an effective drug target for treatment of hormone-insensitive prostate cancer or other types of cancers. Future work may further develop miR-24 for therapeutic applications in cancer biology

    Transcriptome analysis provides StMYBA1 gene that regulates potato anthocyanin biosynthesis by activating structural genes

    Anthocyanin biosynthesis is affected by light, temperature, and other environmental factors. The regulation mode of light on anthocyanin synthesis in apple, pear, tomato and other species has been reported, while not clear in potato. In this study, potato RM-210 tubers whose peel will turn purple gradually after exposure to light were selected. Transcriptome analysis was performed on RM-210 tubers during anthocyanin accumulation. The expression of StMYBA1 gene continued to increase during the anthocyanin accumulation in RM-210 tubers. Moreover, co-expression cluster analysis of differentially expressed genes showed that the expression patterns of StMYBA1 gene were highly correlated with structural genes CHS and CHI. The promoter activity of StMYBA1 was significantly higher in light conditions, and StMYBA1 could activate the promoter activity of structural genes StCHS, StCHI, and StF3H. Further gene function analysis found that overexpression of StMYBA1 gene could promote anthocyanin accumulation and structural gene expression in potato leaves. These results demonstrated that StMYBA1 gene promoted potato anthocyanin biosynthesis by activating the expression of structural genes under light conditions. These findings provide a theoretical basis and genetic resources for the regulatory mechanism of potato anthocyanin synthesis

    Transcriptional activation of follistatin by Nrf2 protects pulmonary epithelial cells against silica nanoparticle-induced oxidative stress

    Silica nanoparticles (SiO2 NPs) cause oxidative stress in respiratory system. Meanwhile, human cells launch adaptive responses to overcome SiO2 NP toxicity. However, besides a few examples, the regulation of SiO2 NP-responsive proteins and their functions in SiO2 NP response remain largely unknown. In this study, we demonstrated that SiO2 NP induced the expression of follistatin (FST), a stress responsive gene, in mouse lung tissue as well as in human lung epithelial cells (A549). The levels of Ac-H3(K9/18) and H3K4me2, two active gene markers, at FST promoter region were significantly increased during SiO2 NP treatment. The induction of FST transcription was mediated by the nuclear factor erythroid 2-related factor 2 (Nrf2), as evidenced by the decreased FST expression in Nrf2-deficient cells and the direct binding of Nrf2 to FST promoter region. Down-regulation of FST promoted SiO2 NP-induced apoptosis both in cultured cells and in mouse lung tissue. Furthermore, knockdown of FST increased while overexpression of FST decreased the expression level of NADPH oxidase 1 (NOX1) and NOX5 as well as the production of cellular reactive oxygen species (ROS). Taken together, these findings demonstrated a protective role of FST in SiO2 NP-induced oxidative stress and shed light on the interaction between SiO2 NPs and biological systems

    Detecting HI Galaxies with Deep Neural Networks in the Presence of Radio Frequency Interference

    In neutral hydrogen (HI) galaxy survey, a significant challenge is to identify and extract the HI galaxy signal from observational data contaminated by radio frequency interference (RFI). For a drift-scan survey, or more generally a survey of a spatially continuous region, in the time-ordered spectral data, the HI galaxies and RFI all appear as regions which extend an area in the time-frequency waterfall plot, so the extraction of the HI galaxies and RFI from such data can be regarded as an image segmentation problem, and machine learning methods can be applied to solve such problems. In this study, we develop a method to effectively detect and extract signals of HI galaxies based on a Mask R-CNN network combined with the PointRend method. By simulating FAST-observed galaxy signals and potential RFI impacts, we created a realistic data set for the training and testing of our neural network. We compared five different architectures and selected the best-performing one. This architecture successfully performs instance segmentation of HI galaxy signals in the RFI-contaminated time-ordered data (TOD), achieving a precision of 98.64% and a recall of 93.59%.Comment: 17 pages, 9 figures, 1 tables.     Road performance evaluation of prestressed high-strength concrete pile waste powder as alternative filler in asphalt concrete

    As a kind of solid waste, using Prestressed High-Strength Concrete Pile Waste Concrete (PPWC) as the replacement for limestone filler in asphalt concrete can not only reduce the accumulation of PPWC and increase its utilization but also avoid the increased road construction costs and environmental degradation associated with limestone mining. This study aims to investigate the effect of using PPWC filler to replace limestone filler on the road performance of asphalt concrete. Firstly, PPWC was ground into filler particles with a diameter less than 0.075 mm. The particle characteristics such as surface morphology, particle size distribution and chemical composition of PPWC filler and limestone filler were compared. Then, PPWC filler was used to replace limestone filler with different volume fractions to prepare asphalt concrete, and the water damage resistance, high-temperature rutting resistance, low-temperature crack resistance, fatigue resistance and adhesion performance of asphalt concrete were tested. The results showed that PPWC filler has a smaller particle size and rougher surface than limestone filler, and it contains Ca(OH)2 produced by hydration. The addition of PPWC filler can effectively improve the mechanical properties of asphalt concrete without reducing its water damage resistance. PPWC filler can improve the high-temperature rutting resistance and low-temperature crack resistance of asphalt concrete, but reduce its low-temperature fatigue resistance. The low content of PPWC filler will enhance the adhesion between asphalt mortar and aggregate. However, when the content of PPWC filler exceeds 50%, Ca (OH)2 in PPWC will reduce the adhesion between acid asphalt mortar and alkaline basalt aggregate. Therefore, the use of PPWC as filler in asphalt mixtures provides a reliable solution for the sustainable development of road materials

    A Method of Interstory Drift Monitoring Using a Smartphone and a Laser Device

    Interstory drift is an important engineering parameter in building design and building structural health monitoring. However, many problems exist in current interstory drift monitoring methods. The traditional method is imprecise&mdash;double numerical integration of acceleration data&mdash;and other direct monitoring methods need professional equipment. This paper proposes a method to solve these problems by monitoring the interstory drift with a smartphone and a laser device. In this method, a laser device is installed on the ceiling while a smartphone is fixed on a steel projection plate on the floor. Compared with a reference sensor, the method designed in this study shows that a smartphone is competent in monitoring the interstory drift. This method utilizes a smartphone application (APP) named D-Viewer to implement monitoring and data storage just in one place, which is also inexpensive. The results showed that this method has an average percent error of 3.37%, with a standard deviation of 2.67%. With the popularization of the smartphone, this method is promising in acquiring large amounts of data, which will be significant for building assessment after an earthquake
