    Genetic population structure of the blue sea star (<i>Linckia laevigata</i>) and the boring giant clam (<i>Tridacna crocea</i>) across Malaysia

    Various studies on population genetics in the Indo-Pacific that include the Coral Triangle region have revealed phylogeographic patterns in marine species populations. However, little is known about the population structure and connectivity pattern among the coral reefs in four seas surrounding Malaysia: the Strait of Malacca (Andaman Sea), the South China Sea, the Sulu Sea, and the Sulawesi Sea. In view of this, we examined the genetic population structure of two invertebrate species: the blue sea star (Linckia laevigata) and the boring clam (Tridacna crocea) from seven localities across Malaysia, including Pulau Layang-Layang in the Spratly Islands. Additional samples of L. laevigata were obtained from the Davao Gulf, the Philippines. The analyses were based on partial mitochondrial cytochrome oxidase I sequences of L. laevigata and T. crocea. Populations of L. laevigata and T. crocea showed departure from neutrality, indicating selection or population expansion. However, a mismatch analysis suggested population expansion. Lack of genetic population structure in L. laevigata (ΦST = − 0.001, P = 0.399) implies high connectivity among the sampled reef sites. There was subtle genetic structuring in populations of T. crocea (ΦST = 0.032, P = 0.009), but AMOVA did not detect any structure in the a priori groupings. The genetic population structure and scale in connectivity are most likely attributed to the life history traits of the sea star and giant clam species, and also to the geological history and seasonal current patterns of the region

    The surgical anatomy of the superior gluteal nerve and anatomical radiologic bases of the direct lateral approach to the hip

    In view of the increasing popularity of the direct lateral approach to the hip joint for hemi- or total hip arthroplasty, the location of the superior gluteal nerve (SGN) was studied. This nerve is in danger when using a transgluteal incision. In 20 embalmed specimens the relation of the SGN to the tip of the greater trochanter (TT) was studied as well as the relation to the iliac crest. For this purpose macroscopy, microscopy and CT were used. In 13 hips a so-called most inferior branch was found at an average of 1 cm distal to the inferior branch, the main trunk of the nerve. There was substantial variation in the course of both the inferior and the most inferior branch of the SGN. In order to prevent nerve damage, proximal extension of the transgluteal incision should be limited to 3 cm cranial to TT. Furthermore the incision has to be confined to the distal one third of the distance TT-iliac crest. In tall people extra care should be taken

Real wage and unemployment rate are the arguments of this function. The conditions of arising of direct and opposite dependence between these two economic indicators are investigated. This function is used to analyze processes in a competitive and monopsonic labor market. The expediency and conditions for applying the exogenous increase in labor compensation by fixing minimal wage are substantiated

    Managing Privacy and understanding the handling of personal data has turned into a fundamental right-at least for Europeans-since May 25th with the coming into force of the General Data Protection Regulation. Yet, whereas many different tools by different vendors promise companies to guarantee their compliance to GDPR in terms of consent management and keeping track of the personal data they handle in their processes, interoperability between such tools as well uniform user facing interfaces will be needed to enable true transparency, user-configurable and -manageable privacy policies and data portability (as also implicitly promised by GDPR). We argue that such interoperability can be enabled by agreed upon vocabularies and Linked Data

    Repertoire Analysis of B-Cells Located in Striated Ducts of Salivary Glands of Patients With Sjogren's Syndrome

    A major complication of primary Sjögren's syndrome (pSS) is development of mucosa associated lymphoid tissue (MALT) B-cell lymphoma, particularly in salivary glands. These lymphomas express FcRL4 and are characteristically associated with lymphoepithelial lesions. Neoplastic B-cells may be derived from non-neoplastic glandular intraductal B-cells, also virtually all expressing FcRL4. A characteristic feature of MALT lymphomas is the production of rheumatoid factors (RFs), which are largely encoded by stereotypic immunoglobulin variable heavy chain (IGHV) sequences. The aim of this study was to examine whether there is a relationship between the intraductal and periductal B-cells and whether the intraductal B-cells are selected for RF. RNA was extracted from laser-microdissected infiltrated ductal areas and periductal infiltrates from frozen parotid gland tissue sections of 5 pSS patients. PCR amplified IGHV transcripts were cloned into pCR™4-TOPO vector and subsequently sequenced. Microdissected ducts yielded 96 unique IGHV sequences derived from intraductal B-cells, while 119 unique IGHV sequences were obtained from periductal infiltrates. No major difference in VH-gene usage was observed between intraductal and periductal B-cells. Nearly all (>90%) IGHV sequences derived from both intraductal and periductal B-cells were mutated. Clonal expansions as defined by shared VDJ rearrangements were also present among both intraductal and periductal B-cells: in total 32 clones were found, from which 12 were located within ducts, 15 in periductal areas, and five clones shared members in both areas. We observed 12 IGHV rearrangements encoding for RF sequences from which two were derived from intraductal B-cells and 10 from periductal B-cells. Nine RF sequences were part of a clone. Together these findings indicate that intraductal and periductal B-cells are closely related to each other. Intraductal B-cells are most likely derived from periductal B-cells. We did not obtain evidence that RF-specific B-cells are enriched within the striated ducts. We speculate that in principle any activated B-cell can enter the striated ducts from the periductal infiltrate, irrespective of its antigenic specificity. Within the ducts, these B-cells may receive additional activation and proliferation signals, to further expand at these sites and by acquisition of driver-mutations develop toward lymphoma

    Persistence of Antipsychotic Use After Clozapine Discontinuation:A Real-World Study Across Antipsychotics

    Although clozapine treatment is often discontinued due to limited efficacy or low tolerability, there is a lack of guidelines and evidence on treatment options after discontinuation of clozapine in patients with schizophrenia. Persistence has proven to be an adequate indicator for treatment effectiveness in patients with schizophrenia. The aim of this study was, therefore, to compare persistence of antipsychotic use between antipsychotic treatment options in patients after stopping clozapine treatment. Registry data from a prescription database representative of the Dutch population (1996-2017) was collected to investigate persistence in patients with schizophrenia who had been using clozapine for >= 90 days. Persistence with antipsychotics after clozapine discontinuation was analyzed using Cox-proportional hazard regression models. Our study population consisted of 321 participants, of whom 138 re-initiated clozapine and 183 started some other antipsychotic in the year after clozapine discontinuation (N = 518 antipsychotic use periods, N = 9,178 months). Second-generation antipsychotics (SGAs) as a group were associated with better persistence compared to first-generation antipsychotics (adjusted hazard ratio (aHR), 0.73; 95% confidence interval (CI) 0.57-0.93; P = 0.011). Compared with other antipsychotics, the following oral monotherapy antipsychotics were associated with significantly better persistence: restarting clozapine (aHR 0.48; 95% CI 0.32-0.71; P <0.001) and switching to risperidone (aHR 0.52; 95% CI 0.32-0.84; P = 0.008) or olanzapine (aHR 0.55; 95% CI 0.35-0.87; P = 0.010). Sensitivity analyses confirmed the results. In conclusion, oral SGAs are associated with better persistence than alternative antipsychotic treatment options in patients discontinuing clozapine for undefined reasons. Especially clozapine (except in those with previous serious adverse reactions to clozapine), olanzapine and risperidone should be considered as oral monotherapy for these patients

    Creating a vocabulary for data privacy : the first-year report of data privacy vocabularies and controls community group (DPVCG)

    Managing privacy and understanding handling of personal data has turned into a fundamental right, at least within the European Union, with the General Data Protection Regulation (GDPR) being enforced since May 25th 2018. This has led to tools and services that promise compliance to GDPR in terms of consent management and keeping track of personal data being processed. The information recorded within such tools, as well as that for compliance itself, needs to be interoperable to provide sufficient transparency in its usage. Additionally, interoperability is also necessary towards addressing the right to data portability under GDPR as well as creation of user-configurable and manageable privacy policies. We argue that such interoperability can be enabled through agreement over vocabularies using linked data principles. The W3C Data Privacy Vocabulary and Controls Community Group (DPVCG) was set up to jointly develop such vocabularies towards interoperability in the context of data privacy. This paper presents the resulting Data Privacy Vocabulary (DPV), along with a discussion on its potential uses, and an invitation for feedback and participation
